Determination of acetylcholine, nitrostigmine and acetylcholinesterase activity in four patients with severe nitrostigmine (E 605 forte) intoxication.

Concentrations of acetylcholine and nitrostigmine as well as acetylcholinesterase activity were determined in 4 patients with severe nitrostigmine intoxication. In the acute phase of the intoxication, acetylcholine levels up to 36 and 55 ng/ml were found in blood plasma and cerebrospinal fluid, respectively. At the same time no activity of acetylcholinesterase in plasma and erythrocytes was detected. Acetylcholinesterase activity gradually recovered at the end of the second week. In the first 3 days after the ingestion plasma nitrostigmine concentrations ranged from 409 to 86 ng/ml. The aim was to evaluate the tone and electric activity of the quadriceps muscle at rest and different torque levels. The second aim was to study whether thickness of soft tissues and change in the joint position would affect muscle tone. Eighteen healthy subjects participated. Stretch-shortening cycle refers to the mechanical condition in which store and recoil of elastic energy occur in the skeletal muscle. This leads to a greater work output when compared to a simple shortening contraction. The subjects performed vertical jumps with and without preliminary counter-movement and with small and large knee angular displacement. The ultrastructure of collagen fibrils was investigated in normal (n 39) and ruptured (n 23) human anterior cruciate ligaments. The normal ligament had a complex three-dimensional structure. Collagen fibrils predominantly had a unidirectional course with parallel arrangement and a mean diameter of 75 (20-185) nm. Four days after anterior cruciate ligament rupture, the mean fibril diameter was increased; it later decreased, probably due to synthesis of young, thin 30-40 nm fibrils. Strains within the bone tissue play a major role in bone (re)modeling. These small strains can be assessed using experimental strain gage measurements, which are challenging and invasive. Further, the strain measurements are, in practise, limited to certain regions of superficial bones only, such as the anterior surface of the tibia.
