Analysis of uncontrolled phosphorus precipitation in anaerobic digesters under thermophilic and mesophilic conditions.

This study compares the operation of mesophilic and thermophilic anaerobic digestion of sewage sludge and their effects in uncontrolled phosphorus precipitation. The research has been carried out using a pilot plant consisting of two digesters of 1.6 m3 working volume, treating the mixed sludge of Alzira WWTP (Valencia, Spain). The digesters were operated in parallel, at different conditions: mesophilic (38 ± 2.0°C) and thermophilic (55 ± 2.5°C) temperatures and organic loading rates (OLR) ranging from 1.1 to 1.7 kg volatile solids (VS) m−3 d−1 and different hydraulic retention times (HRT) 20, 15 and 12 days. The genome sequence of Defluviitoga tunisiensis L3 originating from a thermophilic biogas-production plant was established and recently published as Genome Announcement by our group. The circular chromosome of D. tunisiensis L3 has a size of 2,053,097bp and a mean GC content of 31.38%. To analyze the D. tunisiensis L3 genome sequence in more detail, a phylogenetic analysis of completely sequenced Thermotogae strains based on shared core genes was performed. CitA (citrate) and DcuS (C4-dicarboxylate) sensor kinases in thermophilic Geobacillus kaustophilus and Geobacillus thermodenitrificans

The thermophilic Geobacillus thermodenitrificans and Geobacillus kaustophilus are able to use citrate or C4-dicarboxylates like fumarate or succinate as the substrates for growth. The genomes of the sequenced Geobacillus strains (nine strains) each encoded a two-component system of the CitA family. The sensor kinase of G. thermodenitrificans (termed CitAGt) was able to replace CitA of Escherichia coli (CitAEc) in a heterologous complementation assay restoring expression of the CitAEc-dependent citC-lacZ reporter gene and anaerobic growth on citrate. Microbial community characterization during anaerobic digestion of Scenedesmus spp. under mesophilic and thermophilic conditions

[EN] Microbial communities were thoroughly characterized in a mesophilic anaerobic membrane bioreactor (AnMBR) and a thermophilic continuous stirred tank reactor (CSTR), which were both treating recalcitrant microalgal biomass dominated by Scenedesmus. 16S rRNA amplicon sequencing analysis was performed when the AnMBR achieved 70% algal biodegradation and revealed high microbial diversity, probably due to the high solid retention time (SRT) of the AnMBR configuration. Abstract Harvested microalgae Chlorella spp. and primary sludge were co-digested in a laboratory-scale anaerobic membrane bioreactor (AnMBR) under thermophilic conditions (55 °C). The system was run for 700 days divided into four experimental phases to determine the influence of the organic loading rate on the process performance and the microbial community. The rise in organic loading rate from 0.17 to 0.5 gCOD·L−1·d−1 led to a 35% improvement in methane production. Complete genome sequence of the strain Defluviitoga tunisiensis L3, isolated from a thermophilic, production-scale biogas plant.

An anaerobic, thermophilic bacterium belonging to the phylum Thermotogae was isolated from a rural, thermophilic biogas plant (54 degrees C) producing methane-rich biogas from maize silage, barley, cattle and pig manure. Here we report the first complete genome sequence of the Defluviitoga tunisiensis strain L3, an isolate from the family Thermotogaceae. The strain L3 encodes several genes predicted to be involved in utilization of a large diversity of complex carbohydrates including cellobiose and xylan for the production of acetate, hydrogen (H-2) and carbon dioxide (CO2). Start-up and Operation of Laboratory-Scale Thermophilic Upflow Anaerobic Sludge Blanket Reactors Treating Vegetable Processing Wastewaters

Thermophilic anaerobic treatment of hot vegetable processing wastewaters was studied in laboratory-scale UASB reactors at 55°C. The high-strength wastewater streams, deriving from steam peeling and blanching of carrot, potato and swede were used. The reactors were inoculated with mesophilic granular sludge. Stable thermophilic methanogenesis with about 60% COD removal was reached within 28 days. During the 134 day study period the loading rate was increased up to 24 kg COD m−3 day−1. High treatment efficiency of more than 90% COD removal and concomitant methane production of 7·3 m3 CH4 m−3 day−1 were achieved. A thermo-alkaline lipase from a new thermophileGeobacillus thermodenitrificansAV-5 with potential application in biodiesel production

BACKGROUND A thermophilic lipase-producing Geobacillus thermodenitrificans strain AV-5 was isolated from the Mushroom Spring of Yellowstone National Park in WY, USA and studied as a source of lipase for transesterification of vegetable oils to biodiesel. RESULTS A maximum activity of 330 U mL−1 was produced on 2% (v/v) waste cooking oil at 50 °C, pH 8, aeration rate of 1 vvm and agitation speed of 400 rpm. However, the higher lipase productivity (14.04 U mL−1 h−1) was found at a volumetric oxygen transfer coefficient (kLa) value of 18.48 h−1. A Thermophilic Tetramolecular G-Quadruplex/Hemin DNAzyme.

International audience; The quadruplex-based DNAzyme system is one of the most useful artificial enzymes or catalysts; their unique properties make them reliable alternatives to proteins for performing catalytic transformation. The first prototype of a thermally stable DNAzyme system is presented. This thermophilic DNAzyme is capable of oxidizing substrates at high temperatures (up to 95 degrees C) and long reaction times (up to 18 h at 75 degrees C).
