Legionella pneumophila identifikācija, izmantojot MALDI-TOF MS un SBT metodes

Legionella pneumophila identifikācijai un subtipēšanai tika pielietotas divas metodes – MALDI-TOF MS un SBT. Tika izmantoti 22 no ūdens izolēti ZI ”BIOR” Legionella pneumophila celmi. Ar lateksa aglutinācijas metodi tika noteiktas izmantojamās Legionella pneumophila serogrupas – 1, 2, 3 un 6. MALDI-TOF MS ekstrakcijas metode nodrošināja 100 % identifikāciju visiem Legionella pneumophila izolātiem. Pēc 17 iegūtajiem masas spektra signāliem celmi tika efektīvi subtipēti. Ar SBT metodi celmiem tika iegūti alēļu profili un sekvences tipa numuri, kopumā 15 dažādi Legionella pneumophila ST, no kuriem divi – ST2191 un ST2192 iepriekš nebija iekļauti starptautiskajā ESGLI datubāzē. A portable fibre-optic spectrometry set-up has been assembled and tested for applications in skin diffuse reflectance spectrometry, laser fluorescence spectrometry and multi-wavelength reflection photoplethysmography (multi-PPG) studies. The spectrometry set was tested by diffuse reflectance and fluorescence measurements for diagnostics of skin vascular malformations and pigmented diseases such as nevi and melanoma . Adenoviral transduction of the VEGF gene in an oversized skin flap increases flap survival and perfusion. In this study we investigated the potential of magnetofection of magnetic lipospheres containing VEGF165-cDNA on survival and perfusion of ischemic skin flaps and evaluated the method with respect to the significance of applied magnetic field and ultrasound.

Sphingomyelin induces structural alteration in canine parvovirus capsid

One of the essential steps in canine parvovirus (CPV) infection, the release from endosomal vesicles, is dominated by interactions between the virus capsid and the endosomal membranes. In this study, the effect of sphingomyelin and phosphatidyl serine on canine parvovirus capsid and on the phospholipase A2 (PLA2) activity of CPV VP1 unique N-terminus was analyzed. Accordingly, a significant (P ≤ 0.05) shift of tryptophan fluorescence emission peak was detected at pH 5.5 in the presence of sphingomyelin, whereas at pH 7.4 a similar but minor shift was observed.
