Expanding Two-Photon Intravital Microscopy to the Infrared by Means of Optical Parametric Oscillator
2010
Chronic inflammation in various organs, such as the brain, implies that different subpopulations of immune cells interact with the cells of the target organ. To monitor this cellular communication both morphologically and functionally, the ability to visualize more than two colors in deep tissue is indispensable. Here, we demonstrate the pronounced power of optical parametric oscillator (OPO)-based two-photon laser scanning microscopy for dynamic intravital imaging in hardly accessible organs of the central nervous and of the immune system, with particular relevance for long-term investigations of pathological mechanisms (e.g., chronic neuroinflammation) necessitating the use of fluorescent…
Two-photon absorption dye based on 2,5-bis(phenylacrylonitrile)thiophene with aggregration enhanced fluorescence
2016
This paper reports the synthesis and characterization of a 2,5-bis(phenylacrylonitrile) thiophene based two-photon dye, designed to show enhancement in fluorescence quantum yield in nanoaggregated form. Strong solvatochromism has been observed and explained by the favoritism of locally excited (LE) or internal charge transfer (ICT) state depending on the solvent polarity. Aqueous dispersions of nanoparticles have been prepared and investigated regarding their optical properties which were correlated to the LE and ICT state and the molecular structure of the aggregates. Oscillator strengths of sulphur 3s23p34s3So-3s23p34p3P transitions measured by time resolved two-photon laser spectroscopy
1997
Oscillator strengths for the three 3s23p34s 3S1o-3s23p34p 3P0,1,2 transitions in neutral sulphur have been determined directly from lifetime measurements. Two-photon UV laser excitation and subsequent fluorescence detection have been used on atoms produced in a thermal sulphur beam or in a laser-driven plasma. A good agreement was found between data obtained in both schemes. The lifetime data obtained, τ(3P0) = 46.1(1.0) ns, τ(3P1) = 46.1(1.0) ns and τ(3P2) = 46.1(1.0) ns are compared with results of theoretical and experimental data from literature. The absorption oscillator strengths derived are f(3S1o-3P0) = 0.120(0.003), f(3S1o-3P1) = 0.360(0.008) and f(3S1o-3P2) = 0.600(0.013), respect…
Continuous-wave spontaneous lasing in mercury pumped by resonant two-photon absorption
2010
The first continuous-wave two-photon absorption laser-induced stimulated emission (CTALISE) is demonstrated. The 7^1S-6^1P transition in mercury at 1014nm wavelength is used and selective lasing of different isotopes is observed.
Fast whole-brain imaging of seizures in zebrafish larvae by two-photon light-sheet microscopy
2022
Light-sheet fluorescence microscopy (LSFM) enables real-time whole-brain functional imaging in zebrafish larvae. Conventional one photon LSFM can however induce undesirable visual stimulation due to the use of visible excitation light. The use of two-photon (2P) excitation, employing near-infrared invisible light, provides unbiased investigation of neuronal circuit dynamics. However, due to the low efficiency of the 2P absorption process, the imaging speed of this technique is typically limited by the signal-to-noise-ratio. Here, we describe a 2P LSFM setup designed for non-invasive imaging that enables quintuplicating state-of-the-art volumetric acquisition rate of the larval zebrafish bra…
Nanofabrication on 2D and 3D Topography via Positive‐Tone Direct‐Write Laser Lithography
2019
Direct laser writing (DLW) lithography using two‐photon absorption is a powerful technique mostly used for fabrication of complex structures in micro‐ and nanoscale, by photopolymerizing a negative‐tone resist. In contrast, in this study it is demonstrated that DLW is also well suited for fabricating nano‐ to microscale metallic structures using lift‐off and a positive‐tone photoresist. It is shown first that versatile, fast and large area fabrication is possible on flat two‐dimensional insulating substrates, and an expression for how the line width varies with the scanning speed is derived, with excellent agreement with the experiments. Even more interestingly, a unique application for the…
From Seeds to Islands: Growth of Oxidized Graphene by Two-Photon Oxidation
2016
The mechanism of two-photon induced oxidation of single-layer graphene on Si/SiO2 substrates is studied by atomic force microscopy (AFM) and Raman microspectroscopy and imaging. AFM imaging of areas oxidized by using a tightly focused femtosecond laser beam shows that oxidation is not homogeneous but oxidized and nonoxidized graphene segregate into separate domains over the whole irradiated area. The oxidation process starts from point-like “seeds” which grow into islands finally coalescing together. The size of islands before coalescence is 30–40 nm, and the density of the islands is on the order of 1011 cm–2. Raman spectroscopy reveals growth of the D/G band ratio along the oxidation. Sha…
Two-photon light-sheet microscopy for high-speed whole-brain functional imaging of zebrafish neuronal physiology and pathology
2020
We present the development of a custom-made two-photon light-sheet microscope optimized for high-speed (5 Hz) volumetric imaging of zebrafish larval brain for the analysis of neuronal physiological and pathological activity. High-speed volumetric two-photon light-sheet microscopy is challenging to achieve, due to constrains on the signal-to-noise ratio. To maximize this parameter, we optimized our setup for high peak power of excitation light, while finely controlling its polarization, and we implemented remote scanning of the focal plane to record without disturbing the sample. Two-photon illumination is advantageous for zebrafish larva studies since infra-red excitation does not induce a …
Complete Measurement of the Λ Electromagnetic Form Factors.
2019
The exclusive process e+e−→ΛΛ¯, with Λ→pπ− and Λ¯→p¯π+, has been studied at s=2.396 GeV for measurement of the timelike Λ electric and magnetic form factors, GE and GM. A data sample, corresponding to an integrated luminosity of 66.9 pb−1, was collected with the BESIII detector for this purpose. A multidimensional analysis with a complete decomposition of the spin structure of the reaction enables a determination of the modulus of the ratio R=|GE/GM| and, for the first time for any baryon, the relative phase ΔΦ=ΦE−ΦM. The resulting values are R=0.96±0.14(stat)±0.02(syst) and ΔΦ=37°±12°(stat)±6°(syst), respectively. These are obtained using the recently established and most precise value of …
Power-effective scanning with AODs for 3D optogenic applications
2022
Two-photon (2P) excitation is a cornerstone approach widely employed in neuroscience microscopy for deep optical access and sub-micrometric-resolution light targeting into the brain. However, besides structural and functional imaging, 2P optogenetic stimulations are less routinary, especially in 3D. This is because of the adopted scanning systems, often feebly effective, slow and mechanically constricted. Faster illumination can be achieved through acousto-optic deflectors (AODs) although their applicability to large volumes excitation has been limited by large efficiency drop along the optical axis. Here, we present a new AOD-based scheme for 2P 3D scanning that improves the power delivery…
