Composition influence on pulmonary delivery of rifampicin liposomes.

The effects of lipid concentration and composition on the physicochemical properties, aerosol performance and in vitro toxicity activity of several rifampicin-loaded liposomes were investigated. To this purpose, six liposome formulations containing different amounts of soy phosphatidylcholine and hydrogenated soy phosphatidylcholine, with and without cholesterol and oleic acid, were prepared and fully characterized. Audzēju makrofāgu (tumor-associated macrophages, TAM) pārprogrammēšana ir daudzsološa vēža imūnterapijas stratēģija. Maģistra darba ietvaros tika izveidots un raksturots jauns in vitro 3D modelis makrofāgu programmēšanas pētījumiem vēža sferoīdu klātbūtnē. Pirmkārt, tika parādīts, ka 3D kultivētajiem makrofāgiem citokīnu, hemokīnu un virsmas marķieru profils būtiski neatšķiras no monoslānī kultivētajiem makrofāgiem. Otrkārt, tika izpētītas makrofāgu pārprogrammēšanas iespējas 3D apstākļos. Even though it is known for more than one decade that antigen-encoding RNA can deliver antigenic information to induce antigen-specific immunity against cancer, the nature and mechanism of RNA uptake have remained enigmatic. In this study, we investigated the pharmacokinetics of naked RNA administered into the lymph node. We observed that RNA is rapidly and selectively uptaken by lymph node dendritic cells (DCs). Furthermore, in vitro and in vivo studies revealed that the efficient internalization of RNA by human and murine DCs is primarily driven by macropinocytosis. In vitro antitumor effects of the cold-water extracts of Mediterranean species of genus Pleurotus (higher Basidiomycetes) on human colon cancer cells

The aim of this study was to evaluate whether the cold-water extracts of Pleurotus eryngii var. ferulae (CWE-Pef) and Pleurotus nebrodensis (CWE-Pn), 2 of the most prized wild and cultivated edible mushrooms, can affect the tumor phenotype of human colon cancer HCT116 cells. Our results showed that treatment with CWE- Pef and CWE-Pn resulted in a significant inhibition of the viability of HCT116 cells and promoted apoptosis, as also demonstrated by the increase of Bax-to-Bcl-2 messenger RNA ratio.
