Chiral determination of amphetamine and related compounds using chloroformates for derivatization and high-performance liquid chromatography
1999
The enantiomeric determination of amphetamine and various amphetamine-type compounds by liquid chromatography after chiral derivatization with 9-fluorenylmethyl chloroformate-L-proline (FMOC-L-Pro) is reported. The results obtained were compared with those achieved after achiral derivatization with 9-fluorenylmethyl chloroformate and subsequent separation of the derivatives on a beta-cyclodextrin chiral stationary phase. Conditions for the derivatization of amphetamines with FMOC-L-Pro were investigated, including the effect of the derivatization reagent concentration, pH and reaction time, using amphetamine, ephedrine and pseudoephedrine as model compounds. On the basis of these studies, p…
Simple high-performance liquid chromatographic assay for polyamines and their monoacetyl derivatives.
1993
A rapid reversed-phase high-performance liquid chromatographic method, using pre-column derivatization with benzoyl chloride and ultraviolet detection at 254 nm, was developed for the simultaneous measurement of polyamines and their monoacetyl derivatives. Calibration curves were linear for concentrations from 1.25 to 25 nmol/ml. The method was employed to assay these compounds in chick embryo retina explants using organic solvent extraction and 1,7-diaminoheptane as an internal standard. This simple and sensitive method can be applied to routine determinations of these compounds in various biological samples.
Seafood freshness determination through vapour phase Fourier transform infrared spectroscopy.
2006
Abstract A new vapour-phase manifold has been developed to determine trimethylamine (TMA) in fish and cephalopod samples by Fourier transform infrared (FT-IR) spectroscopy. Samples were treated off-line for 1 h with trichloroacetic acid (TCA), filtered and washed. The obtained extracts were aspirated and alkalinized with NaOH 2.0 M, in an on-line system. TMA was separated from the solution in a gas phase separator and then transported by means of a nitrogen carrier into a home made 10 cm pathlength IR gas cell, where the corresponding FT-IR spectra were acquired by accumulating 30 scans per spectrum with 2 cm−1 nominal resolution. The method was applied to the determination of TMA in natura…
Liquid chromatographic determination of aliphatic amines in water using solid support assisted derivatization with 9-fluorenylmethyl chloroformate
2002
A simple and sensitive method has been developed for the liquid chromatographic determination of short-chain aliphatic amines in water. Analytes are retained in solid-phase extraction (SPE) cartridges, and then derivatized by drawing an aliquot of the fluorogeneic reagent 9-fluorenylmethyl chloroformate (FMOC) through the cartridges. After a certain reaction time the derivatives formed are desorbed with acetonitrile. The collected extracts are then chromatographed on a LiChrospher 100 RP18 125 mm×4 mm i.d., 5 μm, column using an acetonitrile-water gradient. The influence of experimental conditions (SPE material, volume of sample, concentration of FMOC, time of reaction and pH) has been inve…
Identification and determination of amphetamine and methamphetamine in street drugs
2000
Abstract A procedure to identify and quantify amphetamine and/or methamphetamine in street drugs is proposed. The procedure is based on the application of the ACC method (Apparent content curves method) to emission and absorption data of samples. Discrimination and quantification of amphetamine and methamphetamine in samples is carried out based on their different behavior in acid and basic medium. Potential interfering drugs are tested and the accuracy of the method is verified in prepared and real samples. Results obtained are in agreement with a HPLC procedure used as reference.
Chromatographic analysis of phenethylamine-antihistamine combinations using C8, C18 or cyano columns and micellar sodium dodecyl sulfate-pentanol mix…
2001
The chromatographic behaviour of binary and ternary mixtures of several phenethylamines (phenylephrine, phenylpropanolamine, ephedrine, pseudoephedrine and methoxyphenamine) and antihistamines (pheniramine, carbinoxamine, doxylamine, chlorpheniramine, dexchlorpheniramine, dexbrompheniramine, diphenhydramine, tripolidine, azatadine and phenyltoloxamine), found in cough-cold pharmaceutical preparations, was studied using C8, C18 and cyano columns, micellar mobile phases of sodium dodecyl sulfate (SDS) and pentanol and UV detection. Using a C8 column and mobile phases of 0.05 mol l-1 SDS-6% v/v pentanol or 0.15 mol l-1 SDS-2% v/v pentanol at pH 7, more than 30 different phenethylamine-antihist…
Determination of Lobenzarit Disodium in Human Plasma by High-Performance Liquid Chromatography
1998
Abstract A reversed-phase high performance liquid chromatographic procedure was developed for determination of lobenzarit disodium in human plasma over the range 0.5−17.5 μg/mL with diphenylamine as internal standard. Plasma samples were extracted with acetonitrile and an aliquot was injected onto a Lichrospher 100 RP-18 column with ultraviolet absorbance detection at 308 nm. Composition of the mobile phase was acetonitrile-water-glacial acetic acid (50:50:0.2 v/v/v).
Off-line dansylation of amines using C18 solid-phase packings: study of the fluorescence and chemiluminescence detection by post-column derivatizatio…
1999
Abstract Dansylation of amines (primaries and secondaries) using C18 solid-phase supports is described. The time of analysis and the handing sample have been markedly improved with respect to those required for solution dansylation. The dansylated amine derivates were injected onto the liquid chromatography system and fluorescence detected. The sensitivity and selectivity is better than the other dansylation procedures described in the literature and also better than other derivatization reagents such 1,2-napthoquinone 4-sulfonate (NQS) or 3,5-dinitrobenzoyl chloride (3,5-DNB). Dansyl compounds can also form chemiluminescent derivatives, therefore a post-column derivatization procedure with…
Determination of diphenhydramine hydrochloride by flow injection with Bromophenol Blue and turbidimetric measurement
1990
The study of a number of diphenhydramine-dye systems was carried out in order to determine the most suitable precipitate for the turbidimetric determination of diphenhydramine using flow injection (FI). The reagent selected was Bromophenol Blue. The chemical and FI variables were optimised. The calibration graph was linear over the concentration range 50-230 p.p.m. of diphenhydramine hydrochloride. A number of interfering substances were also investigated.
Improvement of peak shape and separation performance of beta-blockers in conventional reversed-phase columns using solvent modifiers.
2003
A comparative study of peak shape, elution behavior, and resolution of 16 beta-blockers (acebutolol, alprenolol, atenolol, bisoprolol, carteolol, celiprolol, esmolol, labetalol, metoprolol, nadolol, oxprenolol, pindolol, practolol, propranolol, sotalol, and timolol) chromatographed with hybrid mobile phases of triethylamine (TEA)-acetonitrile and sodium dodecyl sulfate (SDS)-propanol is performed using conventional reversed-phase columns and isocratic elution. Both solvent modifiers (TEA and SDS) prevent the interaction of the basic drugs with the alkyl-bonded phase. However, the protection mechanisms of silanols on the packing are different. Whereas TEA associates with the silanol sites (b…
