This review summarizes the recent advances concerning metal–organic frameworks (MOFs) modified with several biomolecules (e.g., amino acids, nucleobases, proteins, antibodies, aptamers, etc.) as ligands to prepare affinity-based sorbents for application in the sample preparation field. The preparation and incorporation strategies of these MOF-based affinity materials were described. Specifisku aptamēru atlasīšana izmantojot šūnu - SELEX metodi

Aptamēri pēdējo gadu laikā ir ieguvuši augstāku uzmanību, jo to specifisko saistīšanos pie mērķa var izmantot dažādu slimību diagnostikā un terapijā. Nieru karcinomas ārstēšanas terapijā izmantotie līdzekļi ir toksiski pacientam un nav pietiekami efektīvi. Aizvien vairāk tiek meklētas jaunas pieejas terapijai. Šajā pētījumā izmantojām aptamērus, tos atlasot pret nieru karcinomas šūnām, ar šūnu-SELEX metodi. Pēc 11 atlases cikliem tika veikta aptamēru sekvencēšana, izmantojot otrās paaudzes sekvencēšanu, un iegūto datu analīze. Pēc datu analīzes varēja novērot 15 unikālas aptamēru sekvences, kas specifiski saistās tikai pie RCC-MF šūnām un 4 sekvences, kas saistās ar RC-124 un RCC-MF šūnām, …
New insight into the aptamer conformation and aptamer/protein interaction by surface-enhanced Raman scattering and multivariate statistical analysis

International audience; We study the interaction between one aptamer and its analyte (the MnSOD protein) by the combination of surface-enhanced Raman scattering and multivariate statistical analysis. We observe the aptamer structure and its evolution during the interaction under different experimental conditions (in air or in buffer). Through the spectral treatment by principal component analysis of a large set of SERS data, we were able to probe the aptamer conformations and orientations relative to the surface assuming that the in-plane nucleoside modes are selectively enhanced. Abstract Growing demands for ultrasensitive biosensing have led to the development of numerous signal-amplification strategies. Based on their unique properties (i.e., electro-conductivity, bio-compatibility and ease of synthesis), various iron magnetic nanoparticles (MNPs) have proved to be an excellent nanomaterial for applications in electrochemical biosensing.
