Human Topoisomerase I interaction with DNA on flexible nylon substrate for malaria diagnosis

The study of interaction between human Topoisomerase I (hTopo I) and the appropriate sequences of DNA is shown, with the aim to realize a biosensor for malaria diagnosis. Topoisomerases alter the topological state of DNA by catalysing the breaking and rejoining of DNA strands1. The ligase activity is fundamental for our biosensing strategy, revealing the pathology. hTopo I was chosen as a model system for the Plasmodium analogous. The biosensor is a new flexible and easy to degrade biochip printed on nylon by dip-pen lithography (DPL). It is realized exploiting the high specificity of complementary ssDNA2 and hTopo I ligase activity. DPL operation needs ultra-low amounts of DNA (0.5 µL) to …

Polarimetric Plasmonic Sensing with Bowtie Nanoantenna Arrays

2015

We propose a polarimetric plasmonic biosensor based on bowtie nanoantenna array transducers. Through numerical simulations, based on the finite element method (FEM), we study the phase retardation between the components of light polarized parallel and perpendicular to the major axis of the bowties within the arrays. From a design for high volumetric sensitivity at a wavelength of 780 nm, sensitivities ∼5 rad/RIU is obtained, corresponding to a detection limit of ∼10−7 when using a polarimetric readout platform. Similarly, surface sensitivity of the same array is evaluated by simulating the phase retardation changes induced by the coverage of bioreceptors and analytes of the metallic nanostr…

Application of polydopamine functionalized zinc oxide for glucose biosensor design

2021

Zinc oxide (ZnO) nanostructures are widely used in optical sensors and biosensors. Functionalization of these nanostructures with polymers enables optical properties of ZnO to be tailored. Polydopamine (PDA) is a highly biocompatible polymer, which can be used as a versatile coating suitable for application in sensor and biosensor design. In this research, we have grown ZnO-based nanorods on the surface of ITO-modified glass-plated optically transparent electrodes (glass/ITO). Then the deposition of the PDA polymer layer on the surface of ZnO nanorods was performed from an aqueous PDA solution in such a way glass/ITO/ZnO-PDA structure was formed. The ZnO-PDA composite was characterized by S…

Recent advances on application of peptide nucleic acids as a bioreceptor in biosensors development

2019

Abstract The analogs of DNA are unique biomedical tools that are broadly utilized to develop different types of biosensors. Peptide nucleic acids (PNA) are an individual and notable class of nucleic acid analogs due to their unique, novel physicochemical and biochemical characteristics, stability and resistance to nuclease and protease enzymes, significant interactions with complementary strands and remarkable hybridization attributes. Therefore, they are employed in the preparation and fabrication of various types of functional biosensors. In other words, immobilization of PNA as an appropriate diagnostic probe on the surface of electrochemical and optical converters lead to the fabricatio…

Plasmonic Core–Satellite Assemblies as Highly Sensitive Refractive Index Sensors

2015

Highly sensitive and spectrally tunable plasmonic nanostructures are of great demand for applications such as SERS and parallel biosensing. However, there is a lack of such nanostructures for the midvisible spectral regions as most available chemically stable nanostructures offer high sensitivity in the red to far red spectrum. In this work, we report the assembly of highly sensitive nanoparticle structures using a hydroxylamine mediated core–satellite assembly of 20 nm gold nanoparticle satellites onto 60 nm spherical gold cores. The average number of satellites allows tuning the plasmon resonance wavelength from 543 to 575 nm. The core–satellite nanostructures are stable in pH ranges from…

Hybrid Circuits with Nanofluidic Diodes and Load Capacitors

2017

[EN] The chemical and physical input signals characteristic of micro- and nanofluidic devices operating in ionic solutions should eventually be translated into output electric currents and potentials that are monitored with solid-state components. This crucial step requires the design of hybrid circuits showing robust electrical coupling between ionic solutions and electronic elements. We study experimentally and theoretically the connectivity of the nanofluidic diodes in single-pore and multipore membranes with conventional capacitor systems for the cases of constant, periodic, and white-noise input potentials. The experiments demonstrate the reliable operation of these hybrid circuits ove…

Streptavidin-coated TiO2 surfaces are biologically inert: Protein adsorption and osteoblast adhesion studies

2011

Non-fouling TiO2 surfaces are attractive for a wide range of applications such as biosensors and medical devices, where biologically inert surfaces are needed. Typically, this is achieved by controlled surface modifications which prevent protein adsorption. For example, polyethylene glycol (PEG) or PEG-derived polymers have been widely applied to render TiO2 surfaces biologically inert. These surfaces have been further modified in order to achieve specific bio-activation. Therefore, there have been efforts to specifically functionalize TiO2 surfaces with polymers with embedded biotin motives, which can be used to couple streptavidin for further functionalization. As an alternative, here a s…

Reconstitution of a protein monolayer on thiolates functionalized gaas surface

2012

International audience; In the aim to realize an efficient resonant biosensor, gallium arsenide (GaAs) presents many advantages. In addition to its properties of transduction, GaAs is a crystal for which microfabrication processes were developed, conferring the possibility to miniaturize the device and integrate electronic circuit. Moreover, the biofunctionalization could be realized on the crystalline surface without layer deposition, constituting a real advantage to perform reusable sensor. The functionalization of GaAs surface was engaged in order to immobilize a protein monolayer on this substrate. Functionalization was done using a mixed self assembled monolayer of thiolate molecules. …
