Quantitative determination of casein genetic variants in goat milk: Application in Girgentana dairy goat breed

The study was conducted to develop a high-performance liquid chromatographic (HPLC) method to quantify casein genetic variants (αs2-, β-, and κ-casein) in milk of homozygous individuals of Girgentana goat breed. For calibration experiments, pure genetic variants were extracted from individual milk samples of animals with known genotypes. The described HPLC approach was precise, accurate and highly suitable for quantification of goat casein genetic variants of homozygous individuals. In Schizosaccharomyces pombe, ribosomal protein gene (RPG) promoters contain a TATA analogue element called the HomolD box. The HomolD-binding protein Rrn7 forms a complex with the RNA polymerase II machinery. Despite the importance of ribosome biogenesis to cell survival, the mechanisms involved in the regulation of transcription of eukaryotic RPGs are unknown. Abstract A study has been made of the influence of two pools of caseinophosphopeptides (CPPs) obtained from α s - and β-casein (CN) fractions, and of three specific CPPs (β-CN(1–25)4P, α s1 -CN(64–74)4P and α s2 -CN(1–19)4P), on iron bioavailability (ferritin synthesis) and zinc bioavailability (retention, transport and uptake of zinc) in Caco-2 cells. α-CPP and β-CPP pools did not improve ferritin synthesis, but the three specific CPPs showed an increase in ferritin synthesis in Caco-2 cells versus iron sulphate, β-CN(1–25)4P being the most effective. In this work we analyzed the pathogenic in vivo and in vitro activities for both fish and mammals of extracellular products (ECP) of several isolates of Vibrio damsela implicated in disease problems in marine culture. The ECP from all the strains were strongly lethal for fish (LD50 ranging from 0.06 to 3.7 μg protein/g fish) and mice (LD50 ranging from 0.02 to 0.43 μg protein/g mouse), causing death between 4 and 72 h after inoculation. These ECP samples possessed low proteolytic activity without production of caseinase, gelatinase, or elastase. Efficacy of Brevibacillus brevis strain and its influence on larval rearing of sea bass (Dicentrarchus labrax) were investigated in the present work. Biochemical analyses permit to identify this strain as B. brevis. This bacterium has an inhibitory effect against fish pathogenic bacteria, especially the genus of Vibrio. Enzymatic characterization revealed that B. brevis was lipase positive, amylase, lecithinase and caseinase negative. Adherence assays to abiotic surfaces and challenge test with Artemia larvae demonstrate that B. brevis was fairly adherent and play an important role in the enhancement of the protection of Artemia culture against pathogens. In this work, we present the results obtained in an analytical study of the different types of proteinaceous binding media most commonly used in paintings, using GC-FID as the technique of analysis and GC-MS as a confirmatory technique. The application of this methodology requires prior hydrolysis of the proteins in the binding media to obtain free amino acids and then volatile derivatives, in this case by reaction with chloroformates due to advantages of speed, safety and the aqueous medium in which the reaction occurs. Extracts of resting pine seeds inhibited the proteinase activities present in extracts of endosperms of germinating seeds (hydrolysis of haemoglobin at pH 3.7 and hydrolysis of casein at pH 5.4 and 7.0). Heating the extracts of resting seeds at 60°C destroyed their own proteinase activity but their proteinase inhibitor activity decreased by only 25 to 30%. Some properties of the inhibitor(s) were studied using extracts treated at 60°C. Abstract Viscosities of casein solutions were measured within the dilute range in ammoniacal water and in saline solvents containing different amounts and different kinds of salt. All these data are modeled quantitatively by means of an approach accounting for the polyelectrolyte character of casein micelles. Two parameters are required: The intrinsic viscosity [ η ] and a viscometric interaction parameter β . The behavior of casein is compared with that of chain-like polyelectrolytes. For both polymers one observes a pronounced reduction of [ η ] with increasing salt concentration. The ability of dairy Debaryomyces hansenii, Kluyveromyces lactis and Kluyveromyces marxianus strains to release the casein-derived antihypertensive sequences RYLGY, AYFYPEL, LHLPLP, HLPLP, VPP and/or IPP was examined. Yeast strains were grown in medium with casein as sole nitrogen source and the yeast casein hydrolysates (CSHs) were analysed by HPLC-MS/MS to search for the six antihypertensive sequences. Only LHLPLP and HLPLP were identified in CSHs and exclusively in D. hansenii Dh1 and Dh14 hydrolysates in which both antihypertensive sequences represented approximately 6 (CSH Dh1) and 10% (CSH Dh14) of total peptide content.
