Bioorthogonal red and far-red fluorogenic probes for wash-free live-cell and super-resolution microscopy

AbstractSmall-molecule fluorophores enable the observation of biomolecules in their native context with fluorescence microscopy. Specific labelling via bioorthogonal tetrazine chemistry confers minimal label size and rapid labelling kinetics. At the same time, fluorogenic tetrazine-dye conjugates exhibit efficient quenching of dyes prior to target binding. However, live-cell compatible long-wavelength fluorophores with strong fluorogenicity have been difficult to realize. Here, we report close proximity tetrazine-dye conjugates with minimal distance between tetrazine and fluorophore. This paper describes a general method for the synthesis of novel [1,2,3]triazolo[1,5-a]pyridylboronic acids and esters, and the first results on Suzuki cross-coupling reactions with these new compounds and [1,2,3]triazolo[5,1-a]isoquinolylboronic acid, reacting with a variety of aryl halides as a route to 7-aryltriazolopyridines and 5-aryltriazoloisoquinolines.

The synthesis of fluorine containing pyrazoles has been boosted in the past decades due to the interesting properties that confer these building blocks in pharmaceutical and agricultural active ingredients. There are two main methods for the synthesis of fluorine-containing organic compounds, namely, the direct replacement of an atom or functional group such as diazo, hydroxyl, halogen or hydrogen by fluorine, and the modification of fluorine-containing building blocks. Diazotization of the 3-aminopyrrole-4-carboxamides 6a,b under different conditions directly led to the new ring system pyrrolo[3,4-d]-1,2,3-triazine in excellent yields through an intramolecular coupling reaction. In all the cases it was impossible to isolate the intermediate 3-diazopyrroles.
