Demonstration that the Group II Intron from the Clostridial Conjugative Transposon Tn5397 Undergoes Splicing In Vivo

Previous work has identified the conjugative transposon Tn5397 from Clostridium difficile. This element was shown to contain a group II intron. Tn5397 can be conjugatively transferred from C. difficile to Bacillus subtilis. In this work we show that the intron is spliced in both these hosts and that nonspliced RNA is also present. We constructed a mutation in the open reading frame within the intron, and this prevented splicing but did not prevent the formation of the circular form of the conjugative transposon (the likely transposition intermediate) or decrease the frequency of intergeneric transfer of Tn5397. Synthesis and noncovalent protein conjugation of linear-hyperbranched PEG-poly(glycerol) alpha,omega(n)-telechelics.

Linear-hyperbranched, heterobifunctional alpha,omega(n) telechelic block copolymers consisting of a linear poly(ethylene glycol) (PEG) chain and a hyperbranched polyglycerol (PG) block have been prepared in five steps, using a protected amino-functional initiator. The polyfunctionality omega(n) (OH groups) can be adjusted by the degree of polymerization (DP(n)) of the polyglycerol block. Subsequent introduction of a single biotin unit by amidation in alpha-position permitted noncovalent bioconjugation with avidin.

Measurement of CP asymmetry in D 0 → K - K + and D 0 → π - πdecays

Time-integrated $CP$ asymmetries in $D^0$ decays to the final states $K^- K^+$ and $\pi^- \pi^+$ are measured using proton-proton collisions corresponding to $3\mathrm{\,fb}^{-1}$ of integrated luminosity collected at centre-of-mass energies of $7\mathrm{\,Te\kern -0.1em V}$ and $8\mathrm{\,Te\kern -0.1em V}$. The $D^0$ mesons are produced in semileptonic $b$-hadron decays, where the charge of the accompanying muon is used to determine the initial flavour of the charm meson. 2-Diphenylphosphanyl-1,3,2-diazaphospholidines were prepared via metathesis from 2-chloro-1,3,2-diazaphospholidines and LiPPh2. For some of the products, symmetrisation to tetraphenyldiphosphane and 2,2′-bis-1,3,2-diazaphospholidinyls was observed. Most of the derivatives were characterised by single-crystal X-ray diffraction, which showed that all compounds studied feature elongated exocyclic P–Cl or P–P-bonds, respectively. The extent of this bond lengthening is in the P-phosphanyl-substituted species similar and in the P-chloro-derivatives less pronounced than in corresponding CC-unsaturated 1,3,2-diazaphospholenes. Characterization of PP-cLT Waveguides for Second-Harmonic-Generation and Wavelength-Conversion in the C + L Band of Optical Communications

We report the characterization of single-mode optical waveguides at telecom wavelength, realized in congruent lithium-tantalate. We demonstrate that waveguides realized by proton-exchange show a nonlinear coefficient matching that expected in a bulk crystal.

SYNTHESIS, PHYSICO-CHEMICAL AND BIOLOGICAL CHARACTERIZATION OF A PACLITAXEL MACROMOLECULAR PRODRUG

Paclitaxel was attached to poly(hydroxyethylaspartamide) via a succinic spacer arm by a two-step protocol: (1) synthesis of 2'-O-succinyl-paclitaxel; (2) synthesis of PHEA-2'-O-succinyl-paclitaxel. The 2'-O-succinyl-paclitaxel derivative and the macromolecular conjugate were characterized by UV, IR, NMR and mass spectrometry analysis. The reaction yields were over 95% and the purity of products over 98%. Paclitaxel release and degradation from 2'-O-succinyl-paclitaxel occurred at a faster rate at pH 5.5 than 7.4. SUMMARYThe process leading to gene recombination can be interrupted in the filamentous bacteriaStreptomyces coelicolorby growing mixed cultures on cellophane disks lying on complete medium. The mycelium is harvested, broken, diluted and the broken hyphae plated at different time intervals. By this means some markers can be excluded from heteroclones or from recombinant progeny in early samples. The recombinant pattern clearly changes with time, with an increase of markers contributed to the recombinant progeny. A Very Low Band Gap Diketopyrrolopyrrole-Porphyrin Conjugated Polymer

International audience; A porphyrin-diketopyrrolopyrrole-containing polymer (poly(porphyrin-diketopyrrolopyrrole) (PPDPP)) shows impressive molar absorption coefficients from lambda=300 to 1000 nm. The photophysical and structural properties of PPDPP have been studied. With PPDPP as the electron donor and [ 6,6]phenyl C-71 butyric acid methyl ester (PC71BM) as the electron acceptor, the bulk heterojunction polymer solar cell showed overall power conversion efficiencies of 4.18 and 6.44% for as-cast and two-step annealing processed PPDPP: PC71BM (1: 2) active layers, respectively. Electronic Structure of Oxygen Dangling Bond in GlassySiO2: The Role of Hyperconjugation

The electronic structure and the nature of optical transitions in oxygen dangling bond in silica glass, the nonbridging oxygen hole center (NBOHC), were calculated. The calculation reproduced well the peak positions and oscillator strengths of the well-known optical absorption bands at 2.0 and 4.8 eV, and of the recently discovered absorption band at 6.8 eV. The 2.0 eV band was attributed to transition from the $\ensuremath{\sigma}$ bond between Si and dangling oxygen to nonbonding $\ensuremath{\pi}$ orbital on the dangling oxygen. Stacking as a key property for creating nanoparticles with tunable shape: The case of squalenoyl-doxorubicin

The development of elongated nanoparticles for drug delivery is of growing interest in recent years, due to longer blood circulation and improved efficacy compared to spherical counterparts. Squalenoyl-doxorubicin (SQ-Dox) conjugate was previously shown to form elongated nanoparticles with improved therapeutic efficacy and decreased toxicity compared to free doxorubicin. By using experimental and computational techniques, we demonstrate here that the specific physical properties of SQ-Dox, which include stacking and electrostatic interactions of doxorubicin as well as hydrophobic interactions of squalene, are involved in the formation of nanoassemblies with diverse elongated structures.
