Synthesis and studies of new optimised chelating agents for targeting chemokine receptor CXCR4
2012
The objective of this thesis work was to develop CXCR4-targeted tools to localize and treat cancer at an early stage. In this line, we investigated the synthesis of new target-specific radiopharmaceuticals. The work focused on two main axes, i.e. the chelating agent and the carrier, by using the know-how and the expertise of our group in polyazacycloalkanes synthesis and functionalization. In the first part, we were interested in developing new macrocyclic scaffolds of high potential for copper and gallium chelation. We first focused on the development of a new powerful route towards selectively functionalized constrained homocyclens. The second part was based on C-functionalized 1,4,7-tria…
Dysfunction of Oskyddad causes Harlequin-type ichthyosis-like defects in Drosophila melanogaster.
2020
Prevention of desiccation is a constant challenge for terrestrial organisms. Land insects have an extracellular coat, the cuticle, that plays a major role in protection against exaggerated water loss. Here, we report that the ABC transporter Oskyddad (Osy)—a human ABCA12 paralog—contributes to the waterproof barrier function of the cuticle in the fruit fly Drosophila melanogaster. We show that the reduction or elimination of Osy function provokes rapid desiccation. Osy is also involved in defining the inward barrier against xenobiotics penetration. Consistently, the amounts of cuticular hydrocarbons that are involved in cuticle impermeability decrease markedly when Osy activity is reduced. …
Anion-driven encapsulation of cationic guests inside pyridine[4]arene dimers
2019
Pyridine[4]arenes have previously been considered as anion binding hosts due to the electron-poor nature of the pyridine ring. Herein, we demonstrate the encapsulation of Me4N+ cations inside a dimeric hydrogen-bonded pyridine[4]arene capsule, which contradicts with earlier assumptions. The complexation of a cationic guest inside the pyridine[4]arene dimer has been detected and studied by multiple gas-phase techniques, ESI-QTOF-MS, IRMPD, and DT-IMMS experiments, as well as DFT calculations. The comparison of classical resorcinarenes with pyridinearenes by MS and NMR experiments reveals clear differences in their host–guest chemistry and implies that cation encapsulation in pyridine[4]arene…
Exceptional affinity of nanostructured organic-inorganic hybrid materials towards dioxygen: confinement effect of copper complexes
2007
We report the exceptional reactivity towards dioxygen of a nanostructured organic-inorganic hybrid material due to the confinement of copper cyclam within a silica matrix. The key step is the metalation reaction of the ligand, which can occur before or after xerogel formation through the sol-gel process. The incorporation of a Cu(II) center into the material after xerogel formation leads to a bridged Cu(I)/Cu(II) mixed-valence dinuclear species. This complex exhibits a very high affinity towards dioxygen, attributable to auto-organization of the active species in the solid. The remarkable properties of these copper complexes in the silica matrix demonstrate a high cooperative effect for O(2…
Construction of green nanostructured heterogeneous catalysts via non-covalent surface decoration of multi-walled carbon nanotubes with Pd(II) complex…
2017
Abstract Green nanostructured heterogeneous catalysts were prepared via a bottom-up strategy. Designed ligands were synthesized joining covalently an electrondeficient pyrimidine residue and a scorpiand azamacrocycle. The desired molecular properties were easily transferred to nanostructured materials in two steps: first, exploiting their spontaneous chemisorption onto multi-walled carbon nanotubes (MWCNTs) via the pyrimidinic moiety in water at room temperature, then, taking advantage of the easy coordination of Pd(II) to the azamacrocycle in the same conditions. An evenly distribution of catalytic centres was obtained on the MWCNTs surface. Catalytic properties of these materials were ass…
New approach to condensed pyrid-2-ones
2002
We wish to report a simple procedure for the preparation of 5-substituted-thienopyridin-7-ones and 7-substituted-1,6-naphthyridin-5(6H)-ones, in good yields, from the dianions of 3-methylthiophene-2-carboxylic and 2-methylnicotinic acids on treatment with nitriles. Eight-membered heterocycles with two heteroatoms in a 1,3-relationship of interest in medicinal chemistry
2022
This chapter deals with 1,3-diheterocines, eight-membered heterocyclic systems with two heteroatoms, N, O and S, in a 1,3 relationship, exhibiting biological properties and exhaustively covers the literature from 2007 to the end of November 2019. The biological results of 1,3-diazocines, 1,3-oxazocines, 1,3-dioxocins and 1,3-thiazocines, that are the largest class of 1,3-diheterocines, were collected together with the few results available for 1,3-oxathiocin and 1,3-dithiocin derivatives at that time.
Iodolactonization of 3-Alkynylthiophene-2-Carboxylic and 3-Alkynylpicolinic Acids for the Synthesis of Fused Heterocycles
2020
The iodolactonization of 3-alkynylthiophene-2-carboxylic acids and 3-alkynylpicolinic acids has been investigated. Using I2 as the iodine source and NaHCO3 as the base in MeCN, the process took place smoothly to afford thienopyranones and pyranopyridinones, respectively, from 6-endo-dig cyclization. The method also worked nicely for the transformation of 2-(phenylethynyl)thiophene-3-carboxylic acid and 3-(phenylethynyl) isonicotinic acid into 7-iodo-6-phenyl-4H-thieno[3,2-c]- pyran-4-one and 4-iodo-3-phenyl-1H-pyrano[4,3-c]pyridin-1- one, respectively. Although with some 3-alkynylpicolinic acids the process led to a mixture of the 6-endo-dig and 5-exo-dig products, it could be still made se…
Integrable systems, Frobenius manifolds and cohomological field theories
2022
In this dissertation, we study the underlying geometry of integrable systems, in particular tausymmetric bi-Hamiltonian hierarchies of evolutionary PDEs and differential-difference equations.First, we explore the close connection between the realms of integrable systems and algebraic geometry by giving a new proof of the Witten conjecture, which constructs the string taufunction of the Korteweg-de Vries hierarchy via intersection theory of the moduli spaces of stable curves with marked points. This novel proof is based on the geometry of double ramification cycles, tautological classes whose behavior under pullbacks of the forgetful and gluing maps facilitate the computation of intersection…
On the determination of coherent solar climates over a tropical island with a complex topography
2020
Abstract Many tropical islands aim at developing a greener self-sufficient energy production systems based on renewable energy, notably solar-generated electricity. This work explores the mean diurnal and annual solar cycles over La Reunion island (southwest Indian Ocean: 21°S, 55.5°E), and their spatial behavior, using the Solar surfAce RAdiation Heliosat – East (SARAH-E) satellite-derived data at high spatial ( 0.05 ° × 0.05 ° ) and time (hourly) resolutions over period 1999–2016. Comparisons of the SARAH-E data with ground-based measurements over the period 2011–2015 show differences of ~ 15 % for diurnal-seasonal variations. The solar resource over the island displays strong spatial var…
