Photoactive yellow proteins (PYPs) make up a diverse class of blue-light-absorbing bacterial photoreceptors. Electronic excitation of the p-coumaric acid chromophore covalently bound within PYP results in triphasic quenching kinetics; however, the molecular basis of this behavior remains unresolved. Here we explore this question by examining the excitation-wavelength dependence of the photodynamics of the PYP from Halorhodospira halophila via a combined experimental and computational approach. Ethoxyresorufin-O-deethylase (EROD) activity is a biomarker of exposure to planar aromatic hydrocarbons, and it is often measured from the S9 fraction. The effect of the liver S9 fraction of seven boreal freshwater fish species on the fluorescence of resorufin was studied. The S9 fractions diminished resorufin fluorescence by 40–80%, and there were large differences between species. Thus, using a resorufin standard curve without the S9 fraction leads to a large underestimation of the EROD activity. The synthesis, NMR and UV-Vis spectroscopy measurements and X-ray diffraction analysis of four new metal complexes of the amino terpyridine ligand 4́-[4-(4-aminophenyl)phenyl]-2,2́:6́,2́́-terpyridine L, namely [FeL2](ClO4)2 (1), [ZnL2](ClO4)2 (2), [CdL2](ClO4)2 (3) and [PtMe3IL] (4), are reported. The X-ray crystal structures of complexes 1-3 are 1:2 metal:ligand structures with tridentate ligands decorated around the octahedral metal centers. In complex 4, with L in a bidentate coordination mode, the Pt(IV) coordinated methyl and iodine groups form a fac-arrangement. Fluorescence enhancement of quinolines by protonation.

A study of the fluorescence enhancement of isoquinoline, acridine (benzo[b]quinoline) and benzo[h]quinoline is reported with six organic acids of different pKa values. Protonation was found to be an effective tool in the fluorescence enhancement of quinolines. A significant increase in the fluorescence intensity is observed only when strong acids are used, resulting in an over 50-fold increase in fluorescence with trifluoroacetic or benzenesulfonic acid and isoquinoline in a 1.5 : 1 ratio. The benzenesulfonic acid was found to be the most effective in the protonation of the bases despite its higher pKa value compared to trifluoro- and trichloroacetic acid. Comparison of epifluorescence microscopy and flow cytometry in counting freshwater picophytoplankton

The smaller the phytoplankton, the greater effort is required to distinguish individual cells by optics-based methods. Flow cytometry is widely applied in marine picophytoplankton research, but in freshwater research its role has remained minor. We compared epifluorescence microscopy and flow cytometry in assessing the composition, abundance and cell sizes of autofluorescent picophytoplankton in epilimnia of 46 Finnish lakes. Phycocyaninrich picocyanobacteria were the most dominant. An unexpected and previously unknown resorcinarene mono-crown with a fused benzofuran moiety in its macrocyclic core was obtained as a byproduct from a bridging reaction of tetramethoxy resorcinarene with tetraethylene glycol ditosylate. The formation of the fused benzofuran moiety in the resorcinarene macrocycle resulted in a unique rigid and puckered boat conformation, as shown by XRD studies in the solid state. Modification of the macrocycle was also observed to affect the photophysical properties in solution by enhancing the fluorescence brightness compared with a conventional resorcinarene macrocycle. Computational modeling of protein diffusion in the cell nucleus using the lattice-Boltzmann method

The number of fluorescence applications and methods of analysis is growing in the biological and related sciences. The aim of this work was to accurately determine the diffusion coefficient in nucleus, diffusion in the cytoplasm and nucleo-cytoplasmic translocation should be considered as well. Fluorescence recovery after photobleaching (FRAP) experiments were used in order to study protein diffusion. A numerical model for FRAP was used to determine the diffusion properties of proteins. The present method removed the difficulties of the conventional analysis, could produce, interesting results for protein diffusion in the cell.
