Transcriptome profiling of citrus fruit response to huanglongbing disease.
2010
Huanglongbing (HLB) or "citrus greening" is the most destructive citrus disease worldwide. In this work, we studied host responses of citrus to infection with Candidatus Liberibacter asiaticus (CaLas) using next-generation sequencing technologies. A deep mRNA profile was obtained from peel of healthy and HLB- affected fruit. It was followed by pathway and protein-protein network analysis and quantitative real time PCR analysis of highly regulated genes. We identified differentially regulated pathways and constructed networks that provide a deep insight into the metabolism of affected fruit. Data mining revealed that HLB enhanced transcription of genes involved in the light reactions of phot…
How to untie G-quadruplex knots and why?
2021
International audience; For over two decades, the prime objective of the chemical biology community studying G-quadruplexes (G4s) has been to use chemicals to interact with and stabilize G4s in cells to obtain mechanistic interpretations. This strategy has been undoubtedly successful, as demonstrated by recent advances. However, these insights have also led to a fundamental rethinking of G4-targeting strategies: due to the prevalence of G4s in the human genome, transcriptome, and ncRNAome (collectively referred to as the G4ome), and their involvement in human diseases, should we continue developing G4-stabilizing ligands or should we invest in designing molecular tools to unfold G4s? Here, …
The on-line curvilinear component analysis (onCCA) for real-time data reduction
2015
Real time pattern recognition applications often deal with high dimensional data, which require a data reduction step which is only performed offline. However, this loses the possibility of adaption to a changing environment. This is also true for other applications different from pattern recognition, like data visualization for input inspection. Only linear projections, like the principal component analysis, can work in real time by using iterative algorithms while all known nonlinear techniques cannot be implemented in such a way and actually always work on the whole database at each epoch. Among these nonlinear tools, the Curvilinear Component Analysis (CCA), which is a non-convex techni…
What can the concept of affective scaffolding do for us?
2020
The concept of affective scaffolding designates the various ways in which we manipulate the environment to influence our affective lives. In this article, I present a constructive critique of recent discussion on affective scaffolding. In Part 1, I summarize how the theories of situated mind and niche construction contribute to a multidimensional notion of scaffolding. In Part 2, I focus specifically on affective scaffolding and argue that current ambiguity over its distinctive criteria causes uncertainty as to how the concept can and should be used. In Part 3, I identify and examine two possible responses to the suggested state of conceptual ambiguity. The first, restrictive option is to k…
Unveiling anion-induced folding in tripodal imidazolium receptors by ion-mobility mass spectrometry.
2021
The anion-induced folding of tripodal imidazolium receptors has been investigated by NMR spectroscopy, electrospray ionization ion mobility mass spectrometry and DFT calculations. Such folding can be switched by anion release upon collision induced dissociation.
Writting practice for developp mind and language
2016
This thesis wanted to put on the opposite between established culture, transmitted by School, and private culture, the VAE candidates’ concerned by this research, as well as the author’s. This putting on the opposite is fed by a purpose: to show to what extent the individual and collective work of writing on practical, allows to articulate first the peculiarity of the subjects, the trajectories, the experiences, the situations and the activities, with the generalization of the institutional speech linked with the standardized procedures of the device VAE ? In a second time does this work of writing allows to build knowledges from the experience, object and source of development, and to make…
Suomen kielen oppimisen mahdollisuudet ja työyhteisön tuki puhdistuspalvelualalla: afrikkalaisten maahanmuuttajien käsityksiä ja kokemuksia
2013
This article presents a triple case study of African immigrants’ beliefs about Finnish language learning and support of the work community in the cleaning industry. Cleaning does not require a high language proficiency and it is therefore a typical first job for immigrants, a so-called entrance job. The theoretical framework of the study combines dialogical and sociocultural perspectives on language learning. Learners’ beliefs about work-related affordances and scaffolding are analysed using a dialogical analysis of beliefs. The interviewees’ shared belief is that in spite of their simple duties, cleaning work does provide affordances for learning, e.g. in the form of work-related announcem…
The Proteome and Lipidome of Synechocystis sp. PCC 6803 Cells Grown under Light-Activated Heterotrophic Conditions*
2015
Cyanobacteria are photoautotrophic prokaryotes with a plant-like photosynthetic machinery. Because of their short generation times, the ease of their genetic manipulation, and the limited size of their genome and proteome, cyanobacteria are popular model organisms for photosynthetic research. Although the principal mechanisms of photosynthesis are well-known, much less is known about the biogenesis of the thylakoid membrane, hosting the components of the photosynthetic, and respiratory electron transport chain in cyanobacteria. Here we present a detailed proteome analysis of the important model and host organism Synechocystis sp. PCC 6803 under light-activated heterotrophic growth condition…
Multifunctionality of F-rich nucleoporins
2020
Nucleoporins (Nups) represent a range of proteins most known for composing the macromolecular assembly of the nuclear pore complex (NPC). Among them, the family of intrinsically disordered proteins (IDPs) phenylalanine-glycine (FG) rich Nups, form the permeability barrier and coordinate the high-speed nucleocytoplasmic transport in a selective way. Those FG-Nups have been demonstrated to participate in various biological processes besides nucleocytoplasmic transport. The high number of accessible hydrophobic motifs of FG-Nups potentially gives rise to this multifunctionality, enabling them to form unique microenvironments. In this review, we discuss the multifunctionality of disordered and …
(DIS)Assembly and Structural Stability of mtHsp60 and its Precursor NaÏve Form
2015
Heat shock protein 60kDa is a molecular chaperone (GroEL human homolog) that assists protein folding in mitochondria (mtHsp60). It is synthesized in the cell cytoplasm as a higher molecular weight precursor form (p-mtHsp60) containing a N-terminal targeting sequence, that is cleaved after import into the mitochondrial matrix [1, 2].It has been established, and demonstrated by various techniques, Hsp60 can accumulate in the cytosol, in various pathological conditions (i.e., cancer and chronic inflammatory diseases). The cytosolical Hsp60 accumulation mechanism may occur with or without mitochondrial release concomitantly, so that in the cytosol the two types of 60 kDa chaperonin proteins, (m…
