Cardenolide variation within and among natural populations of Digitalis obscura

Summary Cardenolide content in 49 wild-growing Digitalis obscura plants from six natural populations on the Iberian Peninsula was determined by HPLC. Series A and B glycosides were the predominant cardenolides in all samples, but absolute values varied among and within populations. Hierarchic analysis of variance showed that the proportion of variation attributable to individuals was significantly higher than that attributable to population differences. Furthermore, analyses from greenhouse-grown plants, generated from seeds collected from selected parental genotypes, attributed nearly all the variability in cardenolide content to single plants. From the roots of three species of Acanthophyllum (Caryophyllaceae), two new gypsogenic acid glycosides, 1 and 2, were isolated, 1 from A. sordidum and A. lilacinum, 2 from A. elatius and A. lilacinum, together with three known saponins, glandulosides B and C, and SAPO50. The structures of 1 and 2 were established mainly by 2D NMR techniques as 23-O-β-D-galactopyranosylgypsogenic acid-28-O-β-D-glucopyranosyl-(13)-[β-D-glucopyranosyl-(16)]-β-D-galactopyranoside (1) and gypsogenic acid-28-O-β-D-glucopyranosyl-(13)-[β-D-glucopyranosyl-(16)]-β-D-galactopyranoside (2). In order to evaluate the potential aroma of some Portuguese grapes, several glycosidically bound aroma compounds of ten grape varieties grown in Evora, Alentejo, Portugal, were released, identified and quantified using gas chromatography–mass spectrophotometry. White varieties were Arinto, Perrum, Rabo-de-Ovelha, Roupeiro and Antao Vaz. Red varieties were Trincadeira, Aragonez, Tinta Caiada, Moreto and Castelao. Materials and Methods We carried out a retrospective review of the charts of 55 patients with definite MD in accordance with the AAO-HNS (American Academy of Otolaryngology–Head and Neck Surgery) Diagnostic Scale who were treated at Gruppo Otologico (Piacenza, Rome, Italy). Ruta chalepensis L. (Rutaceae) leaf extract was investigated for its chemical profile and antioxidant and hypoglycaemic properties. The antioxidant effects were investigated by 2,2-diphenyl-1-picrylhydrazyl (DPPH), Chi-carotene bleaching, and metal chelating activity assays. The carbohydrate-hydrolysing enzymes inhibition assay was used to test the hypoglycaemic potential. R. chalepensis showed a high content of hesperidin and rutin with values of 591.9 and 266.7 mg/g dry extract, respectively. The extract exhibited a promising protection of lipid peroxidation (IC50 value of 16.9 mu g/mL) and inhibited both alpha-amylase and alpha-glucosidase enzymes in a concentration-dependent manner. Abstract The synthesis of metallocenic derivatives of aminoglycoside antibiotics, i. e. kanamycin A and tobramycin, is described. The organometallic-labelled compounds have been obtained from the reaction between the polyaminodrugs and organochromium esters of N-hydroxysuccinimide. The reaction proceeded selectively at the 6′-N position, as might be deduced both from the mass and the pH-dependent 13 C-NMR spectra. The procedure could be regarded as generally useful for the metallolabelling of aminoglycoside antibiotics. As an example of application a competitive immunoassay based on the use of these labels is proposed.

Two anti-inflammatory principles were isolated from the methanol extract of the leaves of Erythrospermum monticolum (Flacourtiaceae). The isolation was based on a guided bioassay of the inhibitory activity on TPA-induced ear edema in mice. These compounds were identified as quercetin 3-O-xylosyl(1-->2) rhamnoside and quercetin 3-O-rhamnoside. In addition, their effects on a chronic topic inflammation model were evaluated.
