Association of immunoglobulin GM allotypes with longevity in long-living individuals from Southern Italy

Abstract Background The aim of this study was to analyse the role of GM allotypes, i.e. the hereditary antigenic determinants expressed on immunoglobulin polypeptide chains, in the attainment of longevity. The role played by immunoglobulin allotypes in the control of immune responses is well known as well as the role of an efficient immune response in longevity achievement. So, it is conceivable that particular GM allotypes may contribute to the generation of an efficient immune response that supports successful ageing, hence longevity. GADD45a physically and functionally interacts with TET1

AbstractDNA demethylation plays a central role during development and in adult physiology. Different mechanisms of active DNA demethylation have been established. For example, Growth Arrest and DNA Damage 45-(GADD45) and Ten-Eleven-Translocation (TET) proteins act in active DNA demethylation but their functional relationship is unresolved. Here we show that GADD45a physically interacts – and functionally cooperates with TET1 in methylcytosine (mC) processing. In reporter demethylation GADD45a requires endogenous TET1 and conversely TET1 requires GADD45a. Redox regulation of genome stability by effects on gene expression, epigenetic pathways and DNA damage/repair

Reactive oxygen and nitrogen species (e.g. H2O2, nitric oxide) confer redox regulation of essential cellular signaling pathways such as cell differentiation, proliferation, migration and apoptosis. In addition, classical regulation of gene expression or activity, including gene transcription to RNA followed by translation to the protein level, by transcription factors (e.g. NF-κB, HIF-1α) and mRNA binding proteins (e.g. GAPDH, HuR) is subject to redox regulation. Dynamical twisted mass fermions with light quarks

We present results of dynamical simulations with 2 flavours of degenerate Wilson twisted mass quarks at maximal twist in the range of pseudo scalar masses from 300 to 550 MeV. The simulations are performed at one value of the lattice spacing a \lesssim 0.1 fm. In order to have O(a) improvement and aiming at small residual cutoff effects, the theory is tuned to maximal twist by requiring the vanishing of the untwisted quark mass. Precise results for the pseudo scalar decay constant and the pseudo scalar mass are confronted with chiral perturbation theory predictions and the low energy constants F, \bar{l}_3 and \bar{l}_4 are evaluated with small statistical errors.

Role of TET3 and 5hmC in the small intestinal epithelium homeostasis

The Ten-eleven translocation (TET) enzymes oxidize the DNA base 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC). This DNA hydroxymethylation process generates a stable epigenetic mark, the 5hmC, that is able to regulate gene transcription. In fact, TET proteins and the 5hmC modification are relevant during embryonic development and cellular differentiation. Specifically, in the case of the small intestinal epithelium, the highest DNA hydroxymethylation levels are found at the villus, where 5hmC is preferentially deposited at genes that define mature enterocytes.
