Characterization of 1-aminocyclopropane-1-carboxylic acid oxidase from barley (Hordeum vulgare L.) seedlings and pine (Pinus sylvestris L.) needles

Abstract Biochemical characteristics of 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase extracted from barley ( Hordeum vulgare L.) leaves and pine ( Pinus sylvestris L.) needles were investigated. ACC oxidase from barley leaves was soluble while for complete recovery of ACC oxidase from pine needles addition of Triton X-100 to extraction medium was necessary. The enzyme required Fe 2+ , ascorbate and NaHCO 3 for maximum activity. A non-linear time course of ACC oxidase reaction indicated possible catalytic inactivation of the enzyme. The maximum activity was measured at pH 7.0–7.2 for ACC oxidase from both barley leaves and pine needles. The apparent K m for ACC was found to be 77 and …

The influence of α-aminophosphonic acids on the activity of aminopeptidase from barley seeds—an approach to determine the enzyme specificity

Inhibitory potencies of 24 α-aminophosphonic acids against barley seeds (Hordeum vulgare L.) metallo-aminopeptidase have been determined to evaluate structural requirements of this enzyme. The enzyme was sensitive mostly to the influence of phosphonic acid analogues of phenylalanine and its homologues, thus showing narrow specificity if compared with porcine aminopeptidases M1 and M17 and with Plasmodium aminopeptidase M17.

A barley (Hordeum vulgare L.) LEA3 protein, HVA1, is abundant in protein storage vacuoles

The HVA1 protein belongs to the LEA3 group, which is expressed during the late stage of seed maturation. It is also induced by exogenous abscisic acid (ABA) and a variety of environmental stresses in germinating barley (Hordeum vulgare L.). In the present work, the potential role of HVA1 was investigated by studying its tissue distribution and subcellular localization in mature and stressed seeds by immuno-microscopic methods. The biomass of three agricultural crops, winter wheat (Triticum aestivum L.), barley (Hordeum vulgare L.), and canola (Brassica napus L.), was studied using multi-temporal dual-polarimetric TerraSAR-X data. The radar backscattering coefficient sigma nought of the two polarization channels HH and VV was extracted from the satellite images. Purification and partial characterization of aminopeptidase from barley (Hordeum vulgare L.) seeds.

Aminopeptidases (EC 3.4.11) are proteolytic enzymes, which hydrolyze one amino acid from N-terminus of peptidic substrates. Inhibitors of plant aminopeptidases can find an application in agriculture as herbicides. Isolation and partial characterization of aminopeptidase from barley (Hordeum vulgare L.) seeds has been described. The enzyme was purified to molecular homogeneity using a six-step purification procedure (precipitation with (NH4)2SO4, followed by chromatography on Sephadex G-25, DEAE-Sepharose, Sephacryl HR 300, Macro-Prep Q and Phenyl-Sepharose HP columns).
