Hyaluronic acid based nanohydrogels fabricated by microfluidics for the potential targeted release of Imatinib: Characterization and preliminary eval…
2019
Abstract Microfluidics is emerging as an innovative technique for the “on chip” fabrication of nanoparticles for drug delivery applications. Here, by using an amphiphilic derivative of hyaluronic acid as a starting macromolecule, nanohydrogels loaded with Imatinib were produced by the microfluidic procedure in order to develop an innovative therapeutic tool for the treatment of retinal neovascularization. Both cyRGDC functionalized and non-functionalized nanohydrogels were designed and fabricated by using the same technique. The targeting efficiency of the obtained nanosystems was studied in vitro on human retinal pigment epithelial cells (HRPEpiC) and human umbilical vein endothelial cells…
In-situ gelling xyloglucan formulations as 3D artificial niche for adipose stem cell spheroids.
2020
Abstract Three-dimensional spheroidal cell aggregates of adipose stem cells (SASCs) are a distinct upstream population of stem cells present in adipose tissue, with enhanced regeneration properties in vivo. The preservation of the 3D structure of the cells, from extraction to administration, can be a promising strategy to ensure optimal conditions for cell viability and maintenance of stemness potential. With this aim, an artificial niche was created by incorporating the spheroids into an injectable, in-situ gelling solution of partially degalactosylated xyloglucan (dXG) and an ad hoc formulated culture medium for the preservation of stem cell spheroid features. The evolution of the mechani…
Effect of bioglass on growth and biomineralization of SaOS-2 cells in hydrogel after 3D cell bioprinting.
2014
We investigated the effect of bioglass (bioactive glass) on growth and mineralization of bone-related SaOS-2 cells, encapsulated into a printable and biodegradable alginate/gelatine hydrogel. The hydrogel was supplemented either with polyphosphate (polyP), administered as polyP • Ca2+-complex, or silica, or as biosilica that had been enzymatically prepared from ortho-silicate by silicatein. These hydrogels, together with SaOS-2 cells, were bioprinted to computer-designed scaffolds. The results revealed that bioglass (nano)particles, with a size of 55 nm and a molar ratio of SiO2 : CaO : P2O5 of 55 : 40 : 5, did not affect the growth of the encapsulated cells. If silica, biosilica, or polyP …
Temperature and polymer crosslinking degree influence on drug transfer from alpha,beta-polyasparthydrazide hydrogel to model membranes. A calorimetri…
1998
Abstract A non-steroidal anti-inflammatory drug, diflunisal, has been chosen as drug model to be incorporated in α , β -polyasparthydrazide (PAHy) matrices to study the effect of polymer crosslinking degrees on the release processes from hydrogel ( X =0.4 and X =0.8) to a model membrane represented by unilamellar vesicles of dipalmitoylphosphatidylcholine. The technique employed to monitor these processes was differential scanning calorimetry that appears to be particularly suitable to follow the transfer kinetics of a drug from a controlled release system to void biomembrane model. The drug release from the two PAHy hydrogels differently crosslinked by glutaraldehyde to the lipidic model w…
In Vitro and In Silico Studies of Two 1,4-Naphthoquinones and Their Topical Formulation in Bigels.
2021
Background: 1,4-Naphthoquinones (1,4-NQs) are secondary plant metabolites with numerous biological activities. 1,4-NQs display low water solubility and poor bioavailability. Bigels are a new technology with great potential, which are designated as drug delivery systems. Biphasic bigels consisting of solid and liquid components represent suitable formulations improving diffusion and bioavailability of NQs into the skin. Objective: We evaluated the in silico and in vitro activity of 5,8-dihydroxy-1,4-naphthoquinone (M1) and 2,3-dichloro-5,8-dihydroxy-1,4-naphthoquinone (M2) on elastase and assessed their cytotoxicity towards COLO38 melanoma cells. The 1,4-NQs were loaded into bigels for topi…
New method to determine the true transmissibilities and permeabilities of oxygen in hydrogel membranes
1999
In the present paper, an electrochemical method to obtain the true transmissibilities and permeabilities of hydrogels to oxygen is proposed. The method involves the measurement of the electric current arising from the flow of oxygen through a potentiostat cell integrated by a top water layer, the hydrogel membrane, and a thin water layer between the membrane and the cathode, where the oxygen is reduced. Varying the thickness of the top water layer, the true permeation characteristics of the hydrogels are obtained. A parallel study was carried out in which the transmissibilities and permeation coefficients were obtained from the variation of the steady state current with the thickness of the…
Agarose/κ-carrageenan-based hydrogel film enriched with natural plant extracts for the treatment of cutaneous wounds.
2020
Abstract Hydrogels for complex and chronic wound dressings must be conformable, absorb and retain wound exudates and maintain hydration. They can incorporate and release bioactive molecules that can accelerate the healing process. Wound dressings have to be in contact with the wound and epidermis, even for long periods, without causing adverse effects. Hydrogel dressing formulations based on biopolymers derived from terrestrial or marine flora can be relatively inexpensive and well tolerated. In the present article hydrogel films composed by agarose (1.0 wt%), κ-carrageenan at three different concentrations (0.5, 1.0 and 1.5 wt%) and glycerol (3.0 wt%) were prepared without recourse to cros…
Carboxylated-xyloglucan and peptide amphiphile co-assembly in wound healing.
2021
Abstract Hydrogel wound dressings can play critical roles in wound healing protecting the wound from trauma or contamination and providing an ideal environment to support the growth of endogenous cells and promote wound closure. This work presents a self-assembling hydrogel dressing that can assist the wound repair process mimicking the hierarchical structure of skin extracellular matrix. To this aim, the co-assembly behaviour of a carboxylated variant of xyloglucan (CXG) with a peptide amphiphile (PA-H3) has been investigated to generate hierarchical constructs with tuneable molecular composition, structure, and properties. Transmission electron microscopy and circular dichroism at a low c…
Electrically conductive hydrogel composites made of polyaniline nanoparticles and poly(N-vinyl-2-pyrrolidone)
2006
Abstract A novel electrically conductive composite material, consisting of polyaniline (PANI) nanoparticles dispersed in a polyvinyl pyrrolidone (PVP) hydrogel, was prepared ‘in situ’ by water dispersion polymerisation (DP) of aniline using PVP as steric stabiliser, followed by γ-irradiation induced crosslinking of the PVP component. Conversion yield of aniline into PANI particles was determined via HPLC and gas chromatography, while structural confirmation of the synthesised polymer was sought by FTIR. Morphology and dimensions of PANI particles into the coloured, optically transparent hydrogel was determined by electronic microscopy; moreover, swelling behaviour of composite hydrogels in …
Tuning the Structure of Galacturonate Hydrogels: External Gelation by Ca, Zn, or Fe Cationic Cross-Linkers
2019
International audience; We show here how the nature of various divalent cations M2+ (Ca2+, Zn2+, or Fe2+) influences the structure and mechanical properties of ionotropic polygalacturonate (polyGal) hydrogels designed by the diffusion of cations along one direction (external gelation). All hydrogels exhibit strong gradients of polyGal and cation concentrations, which are similar for all studied cations with a constant ratio R = [M2+]/[Gal] equal to 0.25, showing that every M2+ cation interacts with four galacturonate (Gal) units all along the gels. The regions of the hydrogels formed in the early stages of the gelation process are also similar for all cations and are homogeneous, with the s…
