Gradient high-performance liquid chromatography of statistical and block copolymers of styrene and t-butyl methacrylate

Copolymers of styrene (S) and tert-butyl methacrylate (TBMA) containing 24–92 mass % of the latter monomer were investigated using a silica column with isooctane/tetrahydrofuran (THF) gradients and on a phenyl bonded-phase column by methanol (MeOH)/THF gradients using UV detection. In both cases, retention decreased with increasing TBMA content of the sample. This is in contrast to the behavior of copolymers of S and methyl methacrylate (MMA) whose retention in gradient elution on silica columns increases with MMA content due to the polarity of this unit. A novel highly selective sample cleanup procedure based on the use of molecularly imprinted polymers (MIPs) as solid-phase extraction materials has been evaluated with respect to its applicability and routine use in environmental analysis. The method comprises the combination of a restricted access material (RAM) and a MIP allowing a selective sample preparation to be achieved in the online mode. This combination is called the size-selective sample separation and solvent switch (six-SPE). The RAM column combines size exclusion and adsorption chromatography, reducing the concentration of matrix molecules by a cutoff of 15 kDa. A three-factor optimisation strategy for micellar liquid chromatography

An interpretive optimisation methodology for micellar liquid chromatography (MLC) is shown, taking into account pH, surfactant (sodium dodecyl sulphate) and organic modifier (propanol) concentration. Two objectives are considered: to develop a highly practical straightforward three-factor optimisation for practical MLC, and, in order to avoid unecessary experiments, to link two and three-factor optimisations through a step-wise construction of the experimental design at different pH levels. Development of HPLC methods for the purification and analysis of plasma membrane glycoproteins.

High resolution HPLC techniques such as affinity chromatography (AC), ion exchange chromatography (IEC), and size exclusion chromatography (SEC) were used successfully for separations of hydrophobic plasma membrane glycoproteins. We have tested a lot of commercially available columns for IEC and SEC and performed the purification of the crude plasma membrane extract with the most suitable columns. By using immobilized ligands with different specificities and sequential affinity chromatography, it is possible to obtain a preliminary structural characterization of the interesting carbohydrate residues of membrane glycoproteins.

Resveratrol exerts a variety of biological and pharmacological activities, which are observed despite its extremely low bioavailability and rapid clearance from the circulation due to extensive sulfation and glucuronidation in the intestine and liver.
