Effect of eye fluke infection on the growth of whitefish (Coregonus lavaretus) —An experimental approach

Abstract Effect of the eye fluke Diplostomum spathaceum on fish growth has remained somewhat unclear because 1) the question has not been subjected to experimental examination with treatment-control setup and 2) growth has not been related to the coverage of parasite-induced cataracts in a quantitative manner. Comparative analysis of the coordinated motion of Hsp70s from different organelles observed by single-molecule three-color FRET.

Cellular function depends on the correct folding of proteins inside the cell. Heat-shock proteins 70 (Hsp70s), being among the first molecular chaperones binding to nascently translated proteins, aid in protein folding and transport. They undergo large, coordinated intra- and interdomain structural rearrangements mediated by allosteric interactions. Here, we applied a three-color single-molecule Forster resonance energy transfer (FRET) combined with three-color photon distribution analysis to compare the conformational cycle of the Hsp70 chaperones DnaK, Ssc1, and BiP. O-Glycoprotein 2-acetamino-2-deoxy-β-d-glucopyranosidase (O-GlcNAcase) hydrolyzes O-linked 2-acetamido-2-deoxy-β-d-glucopyranoside (O-GlcNAc) residues from post-translationally modified serine/threonine residues of nucleocytoplasmic protein. The chemical process involves substrate-assisted catalysis, where two aspartate residues have been identified as the two key catalytic residues of O-GlcNAcase. Influence of negative allosteric cooperativity in cation transport.

Abstract The bis-macrocyclic ether5 seems to have a negative allosteric cooperativity and is able to transport double the amount of Na+ and K+ cations as monocyclic systems. This compound could be used as a simple model of the plasma membrane Na+−K+ ATPase which actively pumps Na+ out and K+ into the cell, respectively.

Kinetics of alkaline phosphatase from pig kidney. Influence of complexing agents on stability and activity

Metal ion-complexing agents, like KCN, EDTA etc., inactivate alkaline phosphatase of pig kidney. This inactivation is reversible at low concentrations of the complexing agents and irreversible at high concentrations. The reversible inhibition is probably due to removal of Zn2+ ions from the active site, where they are necessary for catalytic action, whereas the irreversible inhibition results from the removal of Zn2+ ions necessary for preservation of the structure. The inactivation is pseudo-first order. Characterization of vitellogenins in Spilostethus pandurus (Hemiptera): Immunoelectrophoretic studies and short-term labelling experiments

Abstract The haemolymph of Spilostethus pandurus contains three female-specific proteins (I, II and III) as revealed by SDS-PAGE. Antibodies prepared against them recognize three fractions (4, 5 and 6) in ovarian extracts which have similar molecular weights suggesting that I, II and III are vitellogenins. Upon injection of a 14C amino acid mixture into vitellogenic females, labelled proteins first appear in the fat body, then in the haemolymph and next in the ovarian follicles.
