AbstractThe origin of viruses remains an open question. While lack of detectable sequence similarity hampers the analysis of distantly related viruses, structural biology investigations of conserved capsid protein structures facilitate the study of distant evolutionary relationships. Here we characterize the lipid-containing ssDNA temperate bacteriophage ΦCjT23, which infects Flavobacterium sp. (Bacteroidetes). How to reach optimal estimates of confidence intervals in microscopic counting of phytoplankton?

Abstract Present practices in the microscopic counting of phytoplankton to estimate the reliability of results rely on the assumption of a random distribution of taxa in sample preparations. In contrast to that and in agreement with the literature, we show that aggregated distribution is common and can lead to over-optimistic confidence intervals, if estimated according to the shortcut procedure of Lund et al. based on the number of counted cells. We found a good linear correlation between the distribution independent confidence intervals for medians and those for parametric statistics so that 95% confidence intervals can be approximated by using a correction factor of 1.4. AbstractParvoviral genome translocation from the plasma membrane into the nucleus is a coordinated multistep process mediated by capsid proteins. We used fast confocal microscopy line scan imaging combined with image correlation methods including auto-, pair- and cross-correlation, and number and brightness analysis, to study the parvovirus entry pathway at the single-particle level in living cells. Our results show that the endosome-associated movement of virus particles fluctuates from fast to slow. Bacteriophage imaging : past, present and future

The visualization of viral particles only became possible after the advent of the electron microscope. The first bacteriophage images were published in 1940 and were soon followed by many other publications that helped to elucidate the structure of the particles and their interaction with the bacterial hosts. As sample preparation improved and new technologies were developed, phage imaging became important approach to morphologically classify these viruses and helped to understand its importance in the biosphere. In this review we discuss the main milestones in phage imaging, how it affected our knowledge on these viruses and recent developments in the field. peerReviewed

The overall CoCID concept is centred on providing virologists with a next-generation imaging device, which, through increased penetration and depth of focus, as well as through high natural contrast and sensitivity to organelle density (including virus-related organelles), will produce higher-fidelity ultrastructural images of whole intact cells. These insights will, in turn, help increase our understanding of the links between the structural reorganisation of cells and the mechanisms of viral entry, replication, assembly, and egress in cells. Pētījumā tika apskatīta tēma "Apendektomijas epidemioloģija pēc Bērnu klīniskās universitātes slimnīcas datiem 2005. - 2015. gadā”, analizēta bērnu akūta apendicīta saslimstība un to histoloģisku apakštipu izplatība. Pielietota retrospektīva analītiskā pētniecības metode. Teorētiskā daļa balstās uz 44 literatūras avotiem. Pētījumā izmantoti dati no 2279 nosūtījumiem uz patoloģisku izmeklēšanu no slimnīcas arhīva; papildus tiek analizētas materiāla histoloģiskās un imūnhistoķīmiskās pārmaiņas. Izveidota datubāze, veikta datu statistiskā apstrāde. Each class of microscope is limited to imaging specific aspects of cell structure and/or molecular organization. However, imaging the specimen by complementary microscopes and correlating the data can overcome this limitation. Whilst not a new approach, the field of correlative imaging is currently benefitting from the emergence of new microscope techniques. Here we describe the correlation of cryogenic fluorescence tomography (CFT) with soft X‐ray tomography (SXT). This amalgamation of techniques integrates 3D molecular localization data (CFT) with a high‐resolution, 3D cell reconstruction of the cell (SXT). Bismuta selenīda plāno kārtiņu elektroķīmisko īpašību raksturošana nātrija nitrāta ūdens elektrolītā

Bakalaura darba ietvaros tika veikti elektroķīmiskie mērījumi bismuta slelenīda plānajām kārtiņām, izmantojot ciklisko voltamperometriju (CV) un elektroķīmisko impedances spektroskopiju (EIS), kā arī plāno kārtiņu raksturojums, izmantojot skenējošo elektronu mikroskopiju (SEM) un enerģijas dispersīvo rentgenstaru spektroskopiju (EDX). Iegūtie rezultāti statistiski apstrādāti, salīdzināti ar literatūru un izdarīti secinājumi par bismuta selenīdu veikstpēju nātrija nitrāta ūdens elektrolītā. Pētījuma rezultāti norāda uz Bi2Se3 plāno kārtiņu perspektīvu pielietojumu kā anoda elektrodu nātrija jonu baterijās ar ūdens elektrolītu (ASIB).

Parvoviruses are small single-stranded (ss) DNA viruses, which replicate in the nucleoplasm and affect both the structure and function of the nucleus. The nuclear stage of the parvovirus life cycle starts at the nuclear entry of incoming capsids and culminates in the successful passage of progeny capsids out of the nucleus. In this review, we will present past, current, and future microscopy and biochemical techniques and demonstrate their potential in revealing the dynamics and molecular interactions in the intranuclear processes of parvovirus infection. Determining vesicle localization and association in live microscopy may be challenging due to non-simultaneous imaging of rapidly moving objects with two excitation channels. Besides errors due to movement of objects, imaging may also introduce shifting between the image channels, and traditional colocalization methods cannot handle such situations. Our approach to quantifying the association between tagged proteins is to use an object-based method where the exact match of object locations is not assumed.
