Plasmonic Resonant Nanoantennas Induce Changes in the Shape and the Intensity of Infrared Spectra of Phospholipids.

2021

Surface enhanced infrared absorption spectroscopic studies (SEIRAS) as a technique to study biological molecules in extremely low concentrations is greatly evolving. In order to use the technique for identification of the structure and interactions of such biological molecules, it is necessary to identify the effects of the plasmonic electric-field enhancement on the spectral signature. In this study the spectral properties of 1,2-Dipalmitoyl-sn-glycero-3 phosphothioethanol (DPPTE) phospholipid immobilized on gold nanoantennas, specifically designed to enhance the vibrational fingerprints of lipid molecules were studied. An AFM study demonstrates an organization of the DPPTE phospholipid in…

Brain choline has a typical precursor profile.

1998

Choline is product and precursor to both acetylcholine and membrane phospholipids, and, in the brain, is ultimately provided by the circulation. The brain is protected from excess choline and choline deprivation by a refined system of homeostatic mechanisms that maintain a level of extracellular choline that, for its role as precursor, meets saturation criteria under normal conditions. The kinetic and activity profiles of choline are typical for a biosynthetic precursor.

Scanning Force Microscopy of Artificial Membranes

2002

Visualization of biological membranes by scanning force microscopy (SFM) has tremendously improved the current understanding of protein ‐ lipid interactions under physiological conditions. SFM is the only tool to directly image processes on surfaces in aqueous solution at molecular resolution. Besides being a supportive means to confirm results on lipid phases and domains obtained from fluorescence spectroscopy, calorimetry, and X-ray crystallography, SFM has contributed distinct aspects on the formation of 2D crystals of various membrane-confined proteins and morphological changes of membranes due to the interaction of peptides and proteins. This review will focus on recent results in SFM …

A Study of Lipid-Lipid and Lipid-Polypeptide Interactions by High Performance Liquid Chromatography

1984

Abstract Ternary systems containing phosphatidylcholine-cholesterol, phosphatidylcholine-gramicidin A or cholesterol-gramicidin A in tetrahydrofuran have been examined by high performance liquid chromatography. Preferential solvation of cholesterol and especially gramicidin A by phosphatidylcholine is observed. These results are interpreted in terms of hydrophobic interactions between membrane components.

FT-IR investigation of the urea state in lecithin and sodium bis(2-ethylhexyl)phosphate reversed micelles

2003

Abstract FT-IR spectra of urea/lecithin/CCl4 and urea/sodium bis(2-ethylhexyl) phosphate (NaDEHP)/CCl4 systems as a function of the urea-to-surfactant molar ratio (Rurea) at a fixed surfactant concentration (0.1 mol kg−1) have been recorded at 25 °C. Analysis of the absorption spectra leads the to hypothesis that urea is confined within the hydrophilic micellar core of lecithin and NaDEHP reversed micelles. The encapsulation of urea involves some changes of the urea NH stretching band with respect to that of the pure solid urea, attributable to confinement effects. The stretching modes of the surfactant head group are affected by the presence of urea, indicating specific urea-surfactant hea…

Determination of heavy metal content and lipid profiles in mussel extracts from two sites on the moroccan atlantic coast and evaluation of their biol…

2012

Mussels may concentrate pollutants, with possibly significant side effects on human health. Therefore, mussels (Mytilus galloprovincialis) from two sites of the Moroccan Atlantic coast (Jorf Lasfar [JL], an industrial site, and Oualidia [OL], a vegetable-growing area), were subjected to biochemical analyses to quantify the presence of heavy metals (Cd, Cr, and Pb) and to establish the lipid profile: fatty acid, cholesterol, oxysterol, phytosterol and phospholipid content. In addition, mussel lipid extracts known to accumulate numerous toxic components were tested on murine pancreatic β-cells (MIN6), and their biological activities were measured with various flow cytometric and biochemical m…

Polyunsaturated fatty acids in cultured cardiomyocytes : effect on physiology and beta-adrenoceptor function

1992

This study was carried out to investigate the influence of the membrane fatty acid composition on the basal electrical and contractile activities and the response to beta-adrenergic stimulation of rat cardiac muscle cells in culture. Cells were grown for 3 days in a conventional serum culture medium and then incubated for 24 h in synthetic media containing either n-6 or n-3 as the sole source of polyunsaturated fatty acids (PUFA). The n-6/n-3 ratio in the phospholipids was 0.9 in the n-3 cells and 13.1 in the n-6 cells compared with 6.3 in controls cells. Such modifications did not alter action potentials and the main parameters related to contraction, although shortening was slightly acce…

Conformational transitions of gramicidin A in phospholipid model membranes. A high-performance liquid chromatography assessment.

1991

We have investigated the conformation of gramicidin A reconstituted in different phospholipid environments, small unilamellar vesicles, extensive bilayers, and micelles by exploiting a recently proposed experimental approach based on high-performance liquid chromatography [Bano et al. (1988) J. Chromatogr. 458, 105; Bano et al. (1989) FEBS Lett. 250, 67]. The method allows the separation of conformational species of the peptide namely, antiparallel double-stranded (APDS) dimers and β 6.3 -helical monomers, and quantitation of their proportions in the lipid environment. Various experimental parameters (e.g., nature of organic solvent, time of incubation in organic solvent, lipid-to-peptide m…
