Effect of Light on the Composition of Mitochondrial Lipids from Hypocotyl Hooks of Bean Seedlings (Phaseolus vulgaris L.).

Summary Mitochondria were isolated and roughly purified from bean seedlings (phaseolus vulgaris) treated with different red light (660 nm) irradiation programmes. Etiolated seedlings were irradiated for short terms or grown in red light. The mitochondria from differently treated seedlings were analysed for their phospholipid and fatty acid patterns. The mitochondrial phospholipid patterns from green and etiolated seedlings were rather similar. This was also valid for the fatty acids from the phospholipid fraction whereas the fatty acid patterns of the glycolipids showed differences. Quantitative separation of Trichoderma lipid classes on a bonded phase column.

Bond Elut aminopropyl columns were used to purify the different lipid classes of Trichoderma harzianum and Trichoderma viride. This methodology permitted good separation of the fungal lipid classes in less time than traditional techniques. The incorporation of [1 (14)C]linoleic acid into neutral lipids, free fatty acids and phospholipids was quantified for both strains. The fatty acid profile of the different lipid classes of these fungal strains was determined as a function of culture time.

Pulmonary surfactant (PS) constitutes the first line of host defense in the deep lung. Because of its high content of phospholipids and surfactant specific proteins, the interaction of inhaled nanoparticles (NPs) with the pulmonary surfactant layer is likely to form a corona that is different to the one formed in plasma. Here we present a detailed lipidomic and proteomic analysis of NP corona formation using native porcine surfactant as a model. Phospholipid-based reverse micelles.

Physicochemical investigations on the aggregation of phospholipids (mainly phosphatidylcholines) in organic solvents are reviewed and compared with the aggregation behaviour of phospholipids in aqueous medium. In particular we review the data showing that phosphatidylcholines (lecithins) form reverse micellar structures in certain apolar solvents. In these systems not only low molecular weight compounds but also catalytically active enzymes and entire cells can be solubilized. In addition, highly viscous phosphatidylcholine gels can be obtained in organic solvents upon solubilizing a critical amount of water. Dietary arachidonic acid and hepatic desaturation of fatty acids in obese zucker rats

The effect of low levels of dietary arachidonic acid (20:4n-6) on Δ6 desaturation of linoleic acid (18:2n-6) and α-linolenic acid (18:3n-3), and on Δ5 desaturation of dihomo-γ-linolenic acid (20:3n-6) were studied in liver microsomes of obese Zucker rats, in comparison with their lean littermates. Fatty acid composition of serum total lipids and of phospholipids from liver microsomes and from total heart and kidney was determined to see whether modifications of desaturation rate, if any, were reflected in the tissue fatty acid profiles. Influence of a hydrophilic spacer on the structure of a phospholipid monolayer

We present the results of thermodynamical and X-ray grazing incidence diffraction experiments performed on Langmuir monolayers of ether lipids with hydrophilic spacers between the glycerol backbone and the phosphatidylcholine head group: DHPC, DH-E01-PC, and DH-E02-PC. As a spacer the oxy-ethylene group is used, which is a typical hydrophilic unit of non-ionic tensides. It turns out that the introduction of the hydrophilic spacers leads to a fluidization of the monolayer and reduces the tilt angle of the chains.

Block copolymers in giant unilamellar vesicles with proteins or with phospholipids

Biocompatible, highly water-soluble, nonionic, amphiphilic block copolymers having different hydrophobic blocks and architectures, but similar molecular size and chemical nature of the hydrophilic blocks, were investigated to check for their ability to form hybrid giant unilamellar vesicles with proteins, and for their interactions with giant unilamellar phospholipid vesicles (GUV). Abstract The use of an Ultrastyragel 500 A column for the study of interactions between phosphatidylcholine and gramicidin A in tetra-hydrofuran is described. Analysis of vacant peak has allowed to establish the influence that eluent lipid composition, concentration of injected gramicidin and temperature have on the interaction. At 20[ddot] C, for the assayed phospholipid concentration range (0.04 to 0.14 %, w/v), the lipid/polypeptide molar binding ratio, BR, varies from 1.3 to 6.9.
