Determination of heavy metal content and lipid profiles in mussel extracts from two sites on the moroccan atlantic coast and evaluation of their biol…
2012
Mussels may concentrate pollutants, with possibly significant side effects on human health. Therefore, mussels (Mytilus galloprovincialis) from two sites of the Moroccan Atlantic coast (Jorf Lasfar [JL], an industrial site, and Oualidia [OL], a vegetable-growing area), were subjected to biochemical analyses to quantify the presence of heavy metals (Cd, Cr, and Pb) and to establish the lipid profile: fatty acid, cholesterol, oxysterol, phytosterol and phospholipid content. In addition, mussel lipid extracts known to accumulate numerous toxic components were tested on murine pancreatic β-cells (MIN6), and their biological activities were measured with various flow cytometric and biochemical m…

A new sulfated steroidal derivative (fruticogenin A: 1-sulfo-australigenin-3-sodium sulphate, 1) and three new steroidal saponins named fruticoside K (3-sulfo-spirostan-25(27)-ene-1β,3β-diol-1-O-[α-L-rhamnopyranosyl-(1 → 4)-β-D-fucopyranoside], 2), fruticoside L (3-sulfo-spirostan-25(27)-ene-1β,3β,6α-triol-1-O-[α-L-rhamnopyranosyl-(1 → 4)-β-D-fucopyranoside], 3) and fruticoside M (spirostan-25(27)-ene-1β,3α-diol-1-O-[α-L-rhamnopyranosyl-(1 → 2)-α-L-rhamnopyranoside], 4) were isolated from the aerial parts of Cordyline fruticosa L. var. strawberries. Their structures were established on the basis of 1D and 2D NMR data, mass spectrometry and chemical methods. Compounds 2 and 4 exhibited weak …
Highlighting the divertsity of cactus populations (Opuntia spp.) in Morocco and modulation of lipid metabolism by natural extracts and phytosterols f…
2015
The objective of this PhD thesis is to characterize the Moroccan germplasm of the cactus Opuntia collected from different regions in Morocco, by studying the phenologic behavior, the genetic features and physicochemical composition, along with the molecules of high therapeutic potential which may have beneficial effects on the central nervous system cells from cactus extracts (fruit, flower, seed, oil, cladodes and spine) and compared to Argan oil. This work has been conducted in fours different laboratories sharing complementary analyses skills at agronomic, biochemistry, molecular levels. Our results are presented in three parts : (i) cactus agronomic characteristics, (ii) purification an…
Current and new insights on phytosterol oxides in plant sterol-enriched food.
2011
Over the past 15 years, plant sterol-enriched foods have faced a great increase in the market, due to the asserted cholesterol-lowering effect of plant sterols. However, owing to their chemical structures, plant sterols can oxidize and produce a wide variety of oxidation products with controversial biological effects. Although oxyphytosterols can derive from dietary sources and endogenous formation, their single contribution should be better defined. The following review provides an overall and critical picture on the current knowledge and future perspectives of plant sterols-enriched food, particularly focused on occurrence of plant sterol oxidation products and their biological effects. T…
High Performance Liquid Chromatografy-Mass Spectrometry based chemometric characterization of olive oils
2005
In this study the effective discrimination of extra virgin olive oils is described using HPLC-MS, combined with chemometric evaluation. The presented method is simple since the diluted oil sample is directly injected into the system, without any preliminary chemical derivatization or purification step. Separation of diacylglycerols, triacylglycerols and sterols occurs within 20 min and is achieved using an octadecyl-silica column. Detection is performed by positive APCI mass spectrometry which provided sensitivity to detect over 50 compounds in the sample. After extraction of data, stepwise discriminant function analysis is used to select the variables with the highest discriminative power.…
Evaluation of the toxicity of mussels from 2 sites of the Moroccan Atlantic Coast (Jorf Lasfar and Oualidia) used as bioindicators of contamination :…
2012
Toxic substances generated by various human activities are spilled on different area of the Moroccan coast. Shellfishes can concentrate pollutants and have some adverse effects on human health through the food chain. Despite the strengthening of food safety rules, the involvement of chemical pollution of food on metabolic disorders is not known. To predict the impact of pollutants on the aquatic ecosystem and human health, the development of appropriate biomonitoring tools is required. We quantified heavy metals (Cd, Cr and Pb) in mussels (Mytilus galloprovincialis) from two sites of Moroccan Atlantic coast (industrial site Jorf Lasfar (JL) and touristic site Oualidia (OL)) due to the proxim…
Cytotoxic steroidal glycosides from Allium flavum.
2014
Abstract Three new spirostane-type glycosides ( 1 – 3 ) were isolated from the whole plant of Allium flavum . Their structures were elucidated mainly by 2D NMR spectroscopic analysis and mass spectrometry as (20 S ,25 R )-2α-hydroxyspirost-5-en-3β-yl O -β- d -xylopyranosyl-(1 → 3)-[β- d -galactopyranosyl-(1→2)]-β- d -galactopyranosyl-(1→4)-β- d -galactopyranoside ( 1 ), (20 S ,25 R )-2α-hydroxyspirost-5-en-3β-yl O -β- d -xylopyranosyl-(1 → 3)-[β- d -glucopyranosyl-(1→2)]-β- d -galactopyranosyl-(1→4)-β- d -galactopyranoside ( 2 ), and (20 S ,25 R )-spirost-5-en-3β-yl O -α- l -rhamnopyranosyl-(1 → 4)-[β- d -glucopyranosyl-(1→2)]-β- d -glucopyranoside ( 3 ). The three saponins were evaluated f…
Lymphatic absorption of phytosterol oxides in rats
1999
Two of the main classes of oxyphytosterols (7-keto and epoxides) were synthesized from sitosterol and campesterol and given to mesenteric duct-cannulated adult male rats. Lymph was collected during 24 h and was analyzed for oxysterols. The results showed that the lymphatic recovery of the phytosterol oxides was low: 4.7% of the given dose for epoxy derivatives and 1.5% for 7-keto compounds. The campesterol oxides presented a better absorption than the sitosterol oxides. During the process of absorption, the epoxyphytostanols were also partly transformed in campestanetriol and stigmastanetriol.

Testing the Short-Term Efficacy of a Lipid-Lowering Nutraceutical in the Setting of Clinical Practice: A Multicenter Study
2015
Abstract The main guidelines for cardiovascular disease prevention suggest that nutraceuticals could be an efficacious tool to improve lipid pattern. Our aim was to carry out a clinical trial comparing the metabolic effects of a combined nutraceutical containing both red yeast rice and polyunsaturated fatty acids (PUFAs) and a phytosterol-based approach in a setting of clinical practice. This was a multicenter open study with parallel control. We consecutively enrolled 107 pharmacologically untreated subjects affected by primary polygenic hypercholesterolemia and metabolic syndrome, assigned to 8-week treatment with a combined treatment with red yeast rice (Dif1Stat®, including 5 mg monacol…
