Effects on Arterial Blood Pressure of the Methanol and Dichloromethanol Extracts from Schinus molle L. in Rats

The effects on arterial blood pressure of the methanol and dichloromethanol extracts from Schinus molle L. were analysed in urethane anaesthetized rats. In normotensive rats, the mean arterial blood pressure was significantly reduced by the i.v. administration of both extracts. The dichloromethanol extract inhibited the effects of noradrenaline on arterial blood pressure in the anaesthetized rat and it reduced the maximal contractile effect (Emax) induced by noradrenaline on rat vas deferens in the organ bath. However, the methanol extract did not modify the effects of noradrenaline in the evaluated tests.

Generation (“follow-up compounds”) bzw. Wirkstoffe mit neuen Strukturen und Wirkmechanismen konnten zukunftig diese Lucke fullen.

The Tolerability of Nilvadipine Compared to Nifedipine in Patients with Essential Hypertension

In one multicenter, double-blind study, 659 hypertensive patients were treated for 16 weeks with either nilvadipine (n = 326) or nifedipine (n = 333). The major objective of the study was to compare the compatibility of the two calcium antagonists with regard to hepatic compatibility and side-effect profiles. The dosages were chosen so that the effective blood pressure reduction in both groups was equally good (mean decreases in systolic pressure of 27 +/- 12 mm Hg with nilvadipine and 26 +/- 15 mm Hg with nifedipine, and in diastolic pressure of 18 +/- 6 mm Hg with nilvadipine and 19 +/- 7 mm Hg with nifedipine). A comparison of the antihypertensive action of propranolol and the optical isomers of N-isopropyl-p-nitrophenylethanolamine (INPEA)

In hypertensive patients, the effect of propranolol and of the optical isomers of N-isopropyl-p-nitrophenylethanolamine (INPEA) on blood pressure was studied. Single oral doses of 40 mg propranolol, 700 mg (−)-INPEA and 700 mg (+)-INPEA decreased supine systolic as well as standing systolic and diastolic blood pressure. Single intravenous doses of 75 mg (−)-INPEA and 75 mg (+)-INPEA lowered supine systolic and diastolic blood pressure. The antihypertensive action of the beta adrenergic blocking levorotatory isomer of INPEA and of the dextrorotatory isomer, which is devoid of beta adrenergic blocking activity, did not differ. Differential effects of anandamide on acetylcholine release in the guinea-pig ileum mediated via vanilloid and non-CB1 cannabinoid receptors

The effects of anandamide on [3H]-acetylcholine release and muscle contraction were studied on the myenteric plexus-longitudinal muscle preparation of the guinea-pig ileum preincubated with [3H]-choline. Anandamide increased both basal [3H]-acetylcholine release (pEC50 6.3) and muscle tone (pEC50 6.3). The concentration-response curves for anandamide were shifted to the right by 1 μM capsazepine (pKB 7.5 and 7.6), and by the combined blockade of NK1 and NK3 tachykinin receptors with the antagonists CP99994 plus SR142801 (each 0.1 μM). Effect of nimodipine on rheologic parameters in patients with chronic cerebrovascular disease

Abstract Sixteen patients with chronic cerebrovascular disease were treated with a monotherapeutic regimen of nimodipine 30 mg orally three times a day. At baseline and after 45 and 90 days of therapy, the patients' whole blood filterability, erythrocyte membrane fluidity, red blood cell membrane protein lateral mobility, and red blood cell Ca 2+ content (total and cytosolic) were evaluated. 1. Cats were given single injections of 1.8 mg/kg reserpine intraperitoneally. After a time interval of 18 hours the animals were killed. The concentration of adrenaline and noradrenaline in the right and left atria and ventricles was estimated by a method reported previously. 2. Reserpine caused a similar loss of catechol amines from all parts of the heart. The concentration of noradrenaline was reduced by 93–97%, the concentration of adrenaline by 59–79%. 3.
