Organocatalytic enantioselective Strecker reaction with seven-membered cyclic imines

[EN] A highly enantioselective Strecker reaction with dibenzo[b,f][1,4]oxazepines has been described using a dihydroquinine-derived thiourea as organocatalyst. The reaction affords chiral 10,11-dihydrodibenzo[b,f][1,4] oxazepine 11-carbonitrile derivatives in excellent yields (up to 99%) and excellent enantioselectivities (up to 98%) under mild reaction conditions.

Catalytic Asymmetric Reactions Involving the Seven-Membered Cyclic Imine Moieties Present in Dibenzo[b,f][1,4]oxazepines

The dibenzo[b,f][1,4]oxazepine scaffold is a privileged structure in medicinal chemistry that displays a wide variety of biological and pharmacological activities. However, catalytic asymmetric methodologies for the synthesis of chiral dibenzo[b,f][1,4]oxazepine derivatives are scarce in the literature. This microreview presents an overview of enantioselective reactions in which these cyclic seven-membered imines are used as electrophiles, including their substrate scope, limitations and application to the synthesis of related compounds.

The Oriental burrower bug Macroscytus subaeneus (Dallas, 1851) (Hemiptera: Heteroptera: Cydnidae: Cydninae), presently known only from single localities in Flores (Indonesia), the Philippines and Thailand, is recorded from the United Kingdom (hereafter U.K.). A single female specimen was collected on the semi-rural edge of Bath city by a Starling (Sturnus vulgaris L.) and taken to its nest from which it was retrieved during 2015. The specimen is illustrated and its diagnostic characters are provided. We hypothesize that this specimen could have been imported with a shipping container from Thailand.

The deep-sea holasteroid Calymne relicta was first described from a few fragments discovered by the HMS ‘Challenger’ in the Bermuda abyssal plain more than a century ago. In addition to re-examining the type material, we describe herein new specimens from unpublished material collected between 3720 and 4860 m during three scientific expeditions that took place on both sides of the North Atlantic between 1966 and 1991. The new material includes juvenile and adult specimens in sufficiently good preservational state to allow a full redescription, including all types of appendages, some of which have never been described. Dopamine D2 receptors (D2R) in the medial prefrontal cortex (mPFC) are key players in the etiology and therapeutics of schizophrenia. The overactivation of these receptors contributes to mPFC dysfunction. Chronic treatment with D2R agonists modifies the expression of molecules implicated in neuronal structural plasticity, synaptic function, and inhibitory neurotransmission, which are also altered in schizophrenia. Altered gastrointestinal motility in an animal model of Lesch-Nyhan disease.

Mutations in the HGPRT1 gene, which encodes hypoxanthine-guanine phosphoribosyltransferase (HGprt), housekeeping enzyme responsible for recycling purines, lead to Lesch-Nyhan disease (LND). Clinical expression of LND indicates that HGprt deficiency has adverse effects on gastrointestinal motility. Therefore, we aimed to evaluate intestinal motility in HGprt knockout mice (HGprt(−)). Spontaneous and neurally evoked mechanical activity was recorded in vitro as changes in isometric tension in circular muscle strips of distal colon. The adult brain retains a substantial capacity for synaptic reorganization, which includes a wide range of modifications from molecular to structural plasticity. Previous reports have demonstrated that the structural remodeling of excitatory neurons seems to occur in parallel to changes in GABAergic neurotransmission.
