Absence of the endo-beta-1,4-glucanases Cel1 and Cel2 reduces susceptibility to Botrytis cinerea in tomato.

2007

Cel1 and Cel2 are members of the tomato (Solanum lycopersicum Mill) endo-beta-1,4-glucanase (EGase) family that may play a role in fruit ripening and organ abscission. This work demonstrates that Cel1 protein is present in other vegetative tissues and accumulates during leaf development. We recently reported the downregulation of both the Cel1 mRNA and protein upon fungal infection, suggesting the involvement of EGases in plant-pathogen interactions. This hypothesis was confirmed by assessing the resistance to Botrytis cinerea infection of transgenic plants expressing both genes in an antisense orientation (Anti-Cel1, Anti-Cel2 and Anti-Cel1-Cel2). The Anti-Cel1-Cel2 plants showed enhanced …

Development of  a qPCR assay for specific quantification of Botrytis cinerea on grapes

2010

The aim of this study was to develop a system for rapid and accurate real-time quantitative PCR (qPCR) identification and quantification of Botrytis cinerea , one of the major pathogens present on grapes. The intergenic spacer (IGS) region of the nuclear ribosomal DNA was used to specifically detect and quantify B. cinerea . A standard curve was established to quantify this fungus. The qPCR reaction was based on the simultaneous detection of a specific IGS sequence and also contained an internal amplification control to compensate for variations in DNA extraction and the various compounds from grapes that inhibit PCR. In these conditions, the assay had high efficiency (97%), and the limit o…

Analysis of thin film cracking and buckling on compliant substrate by fragmentation test

2007

Application of coating fragmentation test for determination of coating properties is considered. A probabilistic model of coating fragmentation under uniaxial tensile loading is applied for coating and interface property identification of thin brittle coating/polymer substrate system. A finite element model is developed to simulate the process of buckle formation in coating strips during fragmentation test. The measured buckle geometry parameters and buckle density evolution as a function of the applied strain is used to estimate the interface toughness.

Validation and application of a PCR primer set to quantify fungal communities in the soil environment by real-time quantitative PCR

2011

Fungi constitute an important group in soil biological diversity and functioning. However, characterization and knowledge of fungal communities is hampered because few primer sets are available to quantify fungal abundance by real-time quantitative PCR (real-time Q-PCR). The aim in this study was to quantify fungal abundance in soils by incorporating, into a real-time Q-PCR using the SYBRGreen (R) method, a primer set already used to study the genetic structure of soil fungal communities. To satisfy the real-time Q-PCR requirements to enhance the accuracy and reproducibility of the detection technique, this study focused on the 18S rRNA gene conserved regions. These regions are little affec…

Multinuclear and Solid State NMR of Gels

2020

Over the past six decades, nuclear magnetic resonance spectroscopy has been an integral part of synthetic organic and organometallic chemistry, as well as biochemistry. Beyond solution state experiments, increasing developments have opened new avenues to study materials in their solid state. Between two extremes (i.e., solution and solid), there exist several other forms of materials, especially soft materials such as gels and liquid crystals. Traditionally gels have been studied using solution state NMR spectroscopic methods. However, the viscosity of complex viscoelastic fluids such as gels affects the molecular tumbling, which in turn affects the chemical shift anisotropy and dipolar and…

Multivalent N-donor ligands for the construction of coordination polymers and coordination polymer gels

2018

This work describes the synthesis and characterisation of several multivalent N-donor ligands and their coordination compounds and the use of these ligands in the construction of coordination polymers and coordination polymer gels. The project can be divided into two parts. The first part of the research is focused on the coordination chemistry of ring-substituted biimidazoles in acidic media. The dependence of the formation of ion pairs and zwitterionic, especially zinc and copper containing coordination compounds on the pH of the reaction medium and ring-substituents of the ligand were examined. The second part of the study deals with the preparation of Ag, Zn and Cu coordination polymers…

Oligonucleotides-decorated-poly(N-vinyl pyrrolidone) nanogels for gene delivery

2013

Pulsed electron-beam irradiation of a semi-dilute poly(N-vinyl pyrrolidone) (PVP) aqueous solution in the presence of acrylic acid has led to a carboxyl functionalized nanogel system. Nanoparticles hydrodynamic size and surface charge density, in water and as a function of pH, were investigated by dynamic light scattering and laser doppler velocimetry, respectively. Nanogels (NGs) were proved not to be cytotoxic at the cellular level. Indeed, they rapidly bypass the cellular membrane to accumulate in specific cell portions of the cytoplasm, in the perinuclear area. The availability of pendant carboxyl groups on the crosslinked PVP NGs core prompted us to attempt their decoration with a sing…

The adsorption of water vapor on super absorbent product at low temperatures and low mass.

2006

The adsorption isotherms of water vapor on super absorbent product have the same form of type III isotherm at ambient temperature with the hysteresis phenomena. For temperatures lower than ambient, the isotherms become deformed because of the chains effect. The polymer is characterized by a multilayer adsorption, which occurs before a full-course is complete. During adsorption reactions, the polymer undergoes rearrangement polymeric network, which results from a cooperative diffusion of the water molecules and from a chain spacing followed by an expansion of the polymeric network. Three types of water molecules adsorbed on polymer were identified: strongly dependent water, adsorbed water, a…
