Temporal dominance of sensations and sensory profiling: A comparative study
2008
Abstract Temporal dominance of sensations (TDS) is a recent descriptive sensory method consisting in assessing repeatedly, until the sensations end, which sensation is dominant and in scoring its intensity. Compared to time–intensity, this method considers the multidimensionality of the perceptual space over time. The objectives of this study were first to compare description of gels containing different levels of odorants (peach and mint), citric acid, cooling agent and xanthan gum obtained with TDS and with a conventional descriptive method and then to explore the impact of mint and peach odorant on long lasting perception. TDS provided reliable information close to standard sensory profi…
Genetic dissection of the miR-17∼92 cluster of microRNAs in Myc-induced B-cell lymphomas
2009
The miR-17∼92 cluster is frequently amplified or overexpressed in human cancers and has emerged as the prototypical oncogenic polycistron microRNA (miRNA). miR-17∼92 is a direct transcriptional target of c-Myc, and experiments in a mouse model of B-cell lymphomas have shown cooperation between these two oncogenes. However, both the molecular mechanism underlying this cooperation and the individual miRNAs that are responsible for it are unknown. By using a conditional knockout allele of miR-17∼92, we show here that sustained expression of endogenous miR-17∼92 is required to suppress apoptosis in Myc-driven B-cell lymphomas. Furthermore, we show that among the six miRNAs that are encoded by m…
Mikrokokoistetut leviämisvastus- ja nelipisteanturimittaukset puolijohderakenteen varauksenkuljettajien syvyysjakaumien määrittämisessä
2013
Entistä pienemmät puolijohderakenteet vaativat analyysityökaluilta erittäin hyvää herkkyyttä, jonka on myös kehityttävä rakenteiden vaatimusten mukaisesti -- muussa tapauksessa kehitys voi hidastua hyvin paljon ilman kunnollista tietoa valmistusmenetelmien tuloksista. Yksi oleellinen osa rakenteiden analysoimisessa on tieto seostusatomien ja varauksenkuljettajien pitoisuuksista rakenteen syvyyssuunnassa, eli syvyysjakauma. Vuosikymmeniä syvyysjakauman mittaukseen on käytetty leviämisvastusmittausta (SRP, engl. Spreading Resistance Profiling), ja nelipisteanturimittausta (4PP, engl. Four Point Probe) on käytetty kuvaamaan koko syvyysjakauman vaikutusta sähköisessä kontaktissa. Molempien tekn…
JNK ‐dependent gene regulatory circuitry governs mesenchymal fate
2015
The epithelial to mesenchymal transition (EMT) is a biological process in which cells lose cell-cell contacts and become motile. EMT is used during development, for example, in triggering neural crest migration, and in cancer metastasis. Despite progress, the dynamics of JNK signaling, its role in genomewide transcriptional reprogramming, and involved downstream effectors during EMT remain largely unknown. Here, we show that JNK is not required for initiation, but progression of phenotypic changes associated with EMT. Such dependency resulted from JNK-driven transcriptional reprogramming of critical EMT genes and involved changes in their chromatin state. Furthermore, we identified eight no…
Anti-inflammatory Lactobacillus rhamnosus CNCM I-3690 strain protects against oxidative stress and increases lifespan in Caenorhabditis elegans.
2012
International audience; Numerous studies have shown that resistance to oxidative stress is crucial to stay healthy and to reduce the adverse effects of aging. Accordingly, nutritional interventions using antioxidant food-grade compounds or food products are currently an interesting option to help improve health and quality of life in the elderly. Live lactic acid bacteria (LAB) administered in food, such as probiotics, may be good antioxidant candidates. Nevertheless, information about LAB-induced oxidative stress protection is scarce. To identify and characterize new potential antioxidant probiotic strains, we have developed a new functional screening method using the nematode Caenorhabdit…
Drug-induced chromatin accessibility changes associate with sensitivity to liver tumor promotion
2019
This work explores quantitative chromatin accessibility, transcriptional and cis-acting gene regulatory variations underlying mouse strain–specific differences in drug-induced liver tumor promotion sensitivity.
Evidence for PTGER4 ,PSCA, and MBOAT7 as risk genes for gastric cancer on the genome and transcriptome level
2018
Genetic associations between variants on chromosome 5p13 and 8q24 and gastric cancer (GC) have been previously reported in the Asian population. We aimed to replicate these findings and to characterize the associations at the genome and transcriptome level. We performed a fine-mapping association study in 1926 GC patients and 2012 controls of European descent using high dense SNP marker sets on both chromosomal regions. Next, we performed expression quantitative trait locus (eQTL) analyses using gastric transcriptome data from 143 individuals focusing on the GC associated variants. On chromosome 5p13 the strongest association was observed at rs6872282 (P = 2.53 x 10(-04)) and on chromosome …
Circulating miRNAs as diagnostic biomarkers for adolescent idiopathic scoliosis
2018
AbstractThe aetiology of adolescent idiopathic scoliosis (AIS) has been linked to many factors, such as asymmetric growth, neuromuscular condition, bone strength and genetic background. Recently, epigenetic factors have been proposed as contributors of AIS physiopathology, but information about the molecular mechanisms and pathways involved is scarce. Regarding epigenetic factors, microRNAs (miRNAs) are molecules that contribute to gene expression modulation by regulating important cellular pathways. We herein used Next-Generation Sequencing to discover a series of circulating miRNAs detected in the blood samples of AIS patients, which yielded a unique miRNA biomarker signature that diagnos…
Sepsis in preterm infants causes alterations in mucosal gene expression and microbiota profiles compared to non-septic twins
2016
Sepsis is a life-threatening condition in preterm infants. Neonatal microbiota plays a pivotal role in the immune system maturation. Changes in gut microbiota have been associated to inflammatory disorders; however, a link with sepsis in the neonatal period has not yet been established. We aimed to analyze gut microbiota and mucosal gene expression using non-invasively obtained samples to provide with an integrative perspective of host-microbe interactions in neonatal sepsis. For this purpose, a prospective observational case-control study was conducted in septic preterm dizygotic twins and their non-septic twin controls. Fecal samples were used for both microbiota analysis and host genome-…
Genomic characterization of the Atlantic cod sex-locus
2016
AbstractA variety of sex determination mechanisms can be observed in evolutionary divergent teleosts. Sex determination is genetic in Atlantic cod (Gadus morhua), however the genomic location or size of its sex-locus is unknown. Here, we characterize the sex-locus of Atlantic cod using whole genome sequence (WGS) data of 227 wild-caught specimens. Analyzing more than 55 million polymorphic loci, we identify 166 loci that are associated with sex. These loci are located in six distinct regions on five different linkage groups (LG) in the genome. The largest of these regions, an approximately 55 Kb region on LG11, contains the majority of genotypes that segregate closely according to a XX-XY s…
