Impact of antigen presentation on TCR modulation and cytokine release: implications for detection and sorting of antigen-specific CD8+ T cells using …
2002
Abstract Soluble MHC class I molecules loaded with antigenic peptides are available either to detect and to enumerate or, alternatively, to sort and expand MHC class I-restricted and peptide-reactive T cells. A defined number of MHC class I/peptide complexes can now be implemented to measure T cell responses induced upon Ag-specific stimulation, including CD3/CD8/ζ-chain down-regulation, pattern, and quantity of cytokine secretion. As a paradigm, we analyzed the reactivity of a Melan-A/MART-1-specific and HLA-A2-restricted CD8+ T cell clone to either soluble or solid-phase presented peptides, including the naturally processed and presented Melan-A/MART-1 peptide AAGIGILTV or the peptide ana…
Disentangling the effects of optimism and attributions on feelings of success
2014
Two experiments examined the effects of dispositional optimism and attributions on feelings of success in a performance setting. In Experiment 1, participants successfully solved three cognitive tasks and attributed the success either internally (i.e., to themselves) or externally (i.e., to a teammate). We found no effect of optimism, but a significant effect of the attribution: Internal attribution predicted an increase in feelings of success. In Experiment 2, we replicated the design and adopted an extreme groups approach in order to include the extremes of the optimism dimension. Only optimism affected feelings of success in this sample: Pessimistic participants showed higher increases i…
Identification of NY-ESO-1 epitopes presented by human histocompatibility antigen (HLA)-DRB4*0101-0103 and recognized by CD4(+) T lymphocytes of pati…
2000
NY-ESO-1 is a member of the cancer-testis family of tumor antigens that elicits strong humoral and cellular immune responses in patients with NY-ESO-1–expressing cancers. Since CD4+ T lymphocytes play a critical role in generating antigen-specific cytotoxic T lymphocyte and antibody responses, we searched for NY-ESO-1 epitopes presented by histocompatibility leukocyte antigen (HLA) class II molecules. Autologous monocyte-derived dendritic cells of cancer patients were incubated with recombinant NY-ESO-1 protein and used in enzyme-linked immunospot (ELISPOT) assays to detect NY-ESO-1–specific CD4+ T lymphocyte responses. To identify possible epitopes presented by distinct HLA class II allele…
A 3-week post-weaning restricted feeding as alternative to an ad libitum antibiotic-medicated feed: Effects on growth, carcass and meat of rabbits di…
2022
In this investigation, the impact of a 3-week post-weaning restricted feeding (RF) using an antibiotic-free feed, applied to rabbits from 36 to 56 days of age to preserve their health status as alternative to an ad libitum medicated feed with antibiotics (AMF), was verify in relation to rabbit genotype and slaughter age (SA). The RF and AMF treatments were compared evaluating their effects on feed intake, growth, mortality, carcass and meat quality of rabbits of two genotypes, Italian White purebred (IWP) and hybrid Hycole×IWP crossbred (HIWC), slaughtered at 78 or 92 days of age to obtain light or heavy carcasses. At 36 days of age, 256 weaned rabbits of both sexes were divided into 4 homo…
Molecular characterisation of the species of the genus Zygosaccharomyces.
2003
The restriction fragments polymorphisms of the mitochondrial DNA and the PCR fragment that comprised the internal transcribes spacers and the 5.8S rRNA gene, together with the electrophoretic karyotypes of 40 strains from the 10 species of the genus Zygosaccharomyces, including the new species Z. lentus were examined. The RFLP's of the ITS-5.8S region showed a specific restriction pattern for each species, including the new species Z. lentus. The only exception were the species Z. cidri and Z. fermentati that produced identical restriction profiles. The electrophoretic chromosome patterns confirmed the differences between the species of this genus, including the phylogenetic closest species…
Characterisation of four species of the genus Kluyveromyces by mitochondrial DNA restriction analysis
1997
Summary In the present work, we determine the relationships at the within-species level among strains of Kluyveromyces dobzhanskii, K. lactis, K. marxianus, and K. thermotolerans, through the restriction analysis of their mtDNAs. The three first species showed a high level of intraspecific mtDNA divergence, this polymorphism is correlated to the varieties or species defined according to the original taxonomy of the genus, which is in concordance with that shown by other phenotypic or genotypic markers codified for by the nuclear genome. In these species, the analysis of the relationships among strains based on mtDNA restriction data agrees with previous classifications based on morphologica…
Gray code for permutations with a fixed number of cycles
2007
AbstractWe give the first Gray code for the set of n-length permutations with a given number of cycles. In this code, each permutation is transformed into its successor by a product with a cycle of length three, which is optimal. If we represent each permutation by its transposition array then the obtained list still remains a Gray code and this allows us to construct a constant amortized time (CAT) algorithm for generating these codes. Also, Gray code and generating algorithm for n-length permutations with fixed number of left-to-right minima are discussed.
Metilazione del DNA in artrite reumatoide
2005
GENETIC POLYMORPHISM OF THE FOURTH COMPONENT OF HUMAN COMPLEMENT: POPULATION STUDY AND PROPOSAL FOR A REVISED NOMENCLATURE BASED ON GENOMIC PCR TYPIN…
1996
SUMMARY The fourth component of human complement (C4) is coded for by two homologous genes, C4A and C4B, located in the class III region of the major histocompatibility complex (MHC). Genetic typing of C4A and B alleles is routinely carried out by high-voltage agarose gel electrophoresis. The electrophoretic C4 polymorphism can be further subdivided by the Rodgers (Rg) and Chido (Ch) blood groups, which are antigenic determinants of the C4A and B alpha-chains, respectively. We have used a recently described direct PCR typing method using sequence-specific primers (PCR-SSP) in combination with electrophoretic C4 typing as well as genomic RFLP analysis to determine the frequency of C4 allotyp…
Campylobacter spp. contamination of chicken carcasses during processing in relation to flock colonisation.
2005
The presence and numbers of campylobacters on chicken carcasses from 26 slaughter groups, originating from 22 single-house flocks and processed in four UK plants, were studied in relation to the level of flock colonisation determined by examining the caecal contents of at least ten birds per group. The prevalence of campylobacters on carcasses from five campylobacter-negative flocks processed just after other negative flocks was low (/=30%). Campylobacters were isolated from 90 to 100% of carcasses from three flocks which were partly colonised, with 5, 5 and 30% of caecal contents positive, and which were processed after fully colonised flocks. All carcasses from the remaining fully colonis…
