The loss of the inducible Aspergillus carbonarius MFS transporter MfsA leads to ochratoxin A overproduction

Ochratoxin A (OTA), a nephrotoxic compound produced by certain Aspergillus and Penicillium species, is one of the most abundant mycotoxins in food commodities. Aspergillus carbonarius is the main source of OTA in wine, grape juice and dried vine fruits. Although many studies have focused on OTA production by A. carbonarius, little is known about the genes related to OTA production and transport. We have found a transporter that belongs to the major facilitator superfamily (MfsA) which is highly expressed with a 102-fold induction in an ochratoxigenic A. carbonarius strain compared to a low OTA producer strain. Genes differentially expressed by Aspergillus carbonarius strains under ochratoxin A producing conditions

Aspergillus carbonarius is an important ochratoxin A (OTA)-producing fungus that is responsible for toxin contamination of grapes and wine, coffee and cocoa. A suppression subtractive hybridization (SSH) approach was performed with two strains of A. carbonarius, antagonistic in their OTA-production ability, to identify genes whose expression is linked with the ability to produce OTA. BlastX analysis identified 109 differentially-expressed sequences putatively involved in the production of OTA, with significant similarities (Evalue < 10− 5) to sequences deposited in the NCBI non-redundant protein database. A polyphasic approach to the identification of ochratoxin A-producing black Aspergillus isolates from vineyards in Sicily.

Aspergillus strains belonging to section Nigri isolated during a two year survey in eight Sicilian vineyards located on the slopes of Mount Etna (Sicily, Italy) were analysed analyzed in order to characterize species responsible for ochratoxin A (OTA) contamination of grapes. The polyphasic approach permitted analysis of biodiversity of Aspergillus isolates in relation to their morphology, ochratoxigenicity and genetic variability. Predictive assessment of ochratoxin A accumulation in grape juice based-medium by Aspergillus carbonarius using neural networks

Aims: To study the ability of multi-layer perceptron artificial neural networks (MLP-ANN) and radial-basis function networks (RBFNs) to predict ochratoxin A (OTA) concentration over time in grape-based cultures of Aspergillus carbonarius under different conditions of temperature, water activity (a(w)) and sub-inhibitory doses of the fungicide carbendazim. Methods and Results: A strain of A. carbonarius was cultured in a red grape juice-based medium. Characterization and disruption of the cipC gene in the ochratoxigenic fungus Aspergillus carbonarius

Aspergillus carbonarius is considered the most important ochratoxin A (OTA) producing fungi among those causing OTA contamination in grapes and grape-derived products. CipC is a small protein with unknown function that was previously found to be highly up-regulated in an OTA producer strain of A. carbonarius in comparison to a non OTA producer strain. In this study, cipC was characterized and disrupted via Agrobacterium tumefaciens-mediated transformation in an ochratoxigenic A. carbonarius strain in order to study whether this gene has a role in OTA production. Static output-feedback control under information structure constraints

An important challenge in the static output-feedback control context is to provide an isolated gain matrix possessing a zero-nonzero structure, mainly in problems presenting information structure constraints. Although some previous works have contributed some relevant results to this issue, a fully satisfactory solution has not yet been achieved up to now. In this note, by using a Linear Matrix Inequality approach and based on previous results given in the literature, we present an e cient methodology which permits to obtain an isolated static output-feedback gain matrix having, simultaneously, a zero-nonzero structure imposed a priori. Recent Advances in Static Output-Feedback Controller Design with Applications to Vibration Control of Large Structures

Static output-feedback controller design for vehicle suspensions: an effective two-step computational approach

In this study, a novel two-step methodology is applied in designing static output-feedback controllers for a class of vehicle suspension systems. Following this approach, an effective synthesis of static output-feedback controllers can be carried out by solving two consecutive linear matrix inequality optimisation problems.
