Sterol 27-hydroxylase polymorphism significantly associates with shorter telomere, higher cardiovascular and type-2 diabetes risk in obese subjects

2018

Background/objectivesThe pathologic relationship linking obesity and lipid dismetabolism with earlier onset of aging-related disorders, including cardiovascular disease (CVD) and type-2 diabetes (T2D), is not fully elucidate. Chronic inflammatory state, in obese individuals, may accelerate cellular aging. However, leukocyte telomere length (LTL), the cellular biological aging indicator, is elusively linked with obesity. Recent studies indicate that sterol 27-hydroxylase (CYP27A1) is an emerging antiatherogenic enzyme, that, by converting extrahepatic cholesterol to 27-hydroxycholesterol, facilitates cholesterol removal via high-density lipoprotein-cholesterol (HDL-C). We tested the hypothes…

Essential Oils, Pituranthos chloranthus and Teucrium ramosissimum, Chemosensitize Resistant Human Uterine Sarcoma MES-SA/Dx5 Cells to Doxorubicin by …

2021

The multidrug resistance phenotype is a global phenomenon and causes chemotherapy failure in various cancers, such as in uterine sarcomas that have a high mortality rate. To overcome this phenotype, there is growing research interest in developing new treatment strategies. In this study, we highlight the potential of two essential oils from the Apiaceae family, Pituranthos chloranthus (PC) and Teucrium ramosissimum Desf. (TR), to act as chemopreventive and chemosensitizing agents against two uterine sarcoma cell lines, MES-SA and P-gp-overexpressing MES-SA/Dx5 cells. We found that PC and TR were able to inhibit the cell viability of sensitive MES-SA and resistant MES-SA/Dx5 cells by a sligh…

Evaluating the stability of pharmacophore features using molecular dynamics simulations.

2016

Abstract Molecular dynamics simulations of twelve protein—ligand systems were used to derive a single, structure based pharmacophore model for each system. These merged models combine the information from the initial experimental structure and from all snapshots saved during the simulation. We compared the merged pharmacophore models with the corresponding PDB pharmacophore models, i.e., the static models generated from an experimental structure in the usual manner. The frequency of individual features, of feature types and the occurrence of features not present in the static model derived from the experimental structure were analyzed. We observed both pharmacophore features not visible in …

FastaHerder2: Four Ways to Research Protein Function and Evolution with Clustering and Clustered Databases.

2016

The accelerated growth of protein databases offers great possibilities for the study of protein function using sequence similarity and conservation. However, the huge number of sequences deposited in these databases requires new ways of analyzing and organizing the data. It is necessary to group the many very similar sequences, creating clusters with automated derived annotations useful to understand their function, evolution, and level of experimental evidence. We developed an algorithm called FastaHerder2, which can cluster any protein database, putting together very similar protein sequences based on near-full-length similarity and/or high threshold of sequence identity. We compressed 50…

Mandarin essential oils inhibit quorum sensing and virulence factors of Pseudomonas aeruginosa

2016

Pseudomonas aeruginosa produces biofilm and several virulence factors coordinated by quorum sensing (QS) in food. The interruption of QS is a target to control the bacterial virulence. Chemical preservatives used to control biofilm give rise to several food safety problems. For this reason, essential oils (EOs), generally recognized as safe products, are a hopeful alternative. The aim of this work was to determine the chemical composition of mandarin EOs obtained by cold-pressing (EOP) and cold-pressing followed by steam distillation (EOPD) and their antipathogenic properties against P. aeruginosa. Both EOs contained the highest quantities of monoterpene hydrocarbons, mainly limonene, follo…

Evidence in immunotherapy for paediatric respiratory allergy: Advances and recommendations. Document of the Immunotherapy Working Group of the Spanis…

2016

Allergic respiratory diseases are major health problems in paediatric population due their high level of prevalence and chronicity, and to their relevance in the costs and quality of life. One of the most important risk factors for the development of airway diseases in children and adolescents is atopy. The mainstays for the treatment of these diseases are avoiding allergens, controlling symptoms, and preventing them through sustained desensitization by allergen immunotherapy (AIT). AIT is a treatment option that consists in the administration of increasing amounts of allergens to modify the biological response to them, inducing long-term tolerance even after treatment has ended. This treat…

Whole-genome sequencing for TB source investigations : principles of ethical precision public health

2021

[Background]: Whole-genome sequencing (WGS) of Mycobacterium tuberculosis allows rapid, accurate inferences about the sources, location and timing of transmission. However, in an era of heightened concern for personal privacy and science distrust, such inferences could result in unintended harm and undermine the public's trust.

Field tests of astrometric subsystem

2016

The paper presents results of astrometric subsystem’s functionality tests for space object laser ranging and astrometric position determination device, which is under construction in the Institute of Geodesy and Geoinformatics (GGI) of the University of Latvia. Properties of hardware performance for astrometric image acquisition were evaluated and optimal parameters have been selected. Software for image processing and calculation of instrument orientation was tested and adjustments have been made for real-time operation support. Object tracking properties were evaluated, and mount error model parameters were calculated, using test measurements.
