Human Topoisomerase I interaction with DNA on flexible nylon substrate for malaria diagnosis
The study of interaction between human Topoisomerase I (hTopo I) and the appropriate sequences of DNA is shown, with the aim to realize a biosensor for malaria diagnosis. Topoisomerases alter the topological state of DNA by catalysing the breaking and rejoining of DNA strands1. The ligase activity is fundamental for our biosensing strategy, revealing the pathology. hTopo I was chosen as a model system for the Plasmodium analogous. The biosensor is a new flexible and easy to degrade biochip printed on nylon by dip-pen lithography (DPL). It is realized exploiting the high specificity of complementary ssDNA2 and hTopo I ligase activity. DPL operation needs ultra-low amounts of DNA (0.5 µL) to …

Extracellular Vesicle microRNAs Contribute to the Osteogenic Inhibition of Mesenchymal Stem Cells in Multiple Myeloma
2020
Osteolytic bone disease is the major complication associated with the progression of multiple myeloma (MM). Recently, extracellular vesicles (EVs) have emerged as mediators of MM-associated bone disease by inhibiting the osteogenic differentiation of human mesenchymal stem cells (hMSCs). Here, we investigated a correlation between the EV-mediated osteogenic inhibition and MM vesicle content, focusing on miRNAs. By the use of a MicroRNA Card, we identified a pool of miRNAs, highly expressed in EVs, from MM cell line (MM1.S EVs), expression of which was confirmed in EVs from bone marrow (BM) plasma of patients affected by smoldering myeloma (SMM) and MM. Notably,we found that miR-129-5p, whic…
The prolyl-isomerase PIN1 is essential for nuclear Lamin-B structure and function and protects heterochromatin under mechanical stress.
2021
Summary: Chromatin organization plays a crucial role in tissue homeostasis. Heterochromatin relaxation and consequent unscheduled mobilization of transposable elements (TEs) are emerging as key contributors of aging and aging-related pathologies, including Alzheimer’s disease (AD) and cancer. However, the mechanisms governing heterochromatin maintenance or its relaxation in pathological conditions remain poorly understood. Here we show that PIN1, the only phosphorylation-specific cis/trans prolyl isomerase, whose loss is associated with premature aging and AD, is essential to preserve heterochromatin. We demonstrate that this PIN1 function is conserved from Drosophila to humans and prevents…
Extracellular Vesicles as Shuttles of Tumor Biomarkers and Anti-Tumor Drugs
2014
Extracellular vesicles (EV) include vesicles released by either normal or tumor cells. EV may exceed the nanometric scale (microvesicles), or to be within the nanoscale, also called exosomes. Thus, it appears that only exosomes and larger vesicles may have the size for potential applications in nanomedicine, in either disease diagnosis or therapy. This is of particular interest for research in cancer, also because the vast majority of existing data on EV are coming from pre-clinical and clinical oncology. We know that the microenvironmental features of cancer may favor cell-to-cell paracrine communication through EV, but EV have been purified, characterized, and quantified from plasma of tu…

Somekohun anatomia : eli miten somekohut ja somekohuotsikot rakentuvat
2017
Tutkimuksen tavoitteena on selvittää, millainen uuden median luoma ilmiö somekohu on ja miten somekohuista uutisoidaan. Tutkimuskysymyksiä ovat: Millaisia somekohu-uutisten otsikot ovat? Millaisista aineksista somekohu rakentuu? Aineisto koostuu 77 somekohuaiheisesta verkko-otsikosta. Tutkimus pohjautuu käsitykselle, että kielellä on valtaa ja kielenkäyttäjän valinnat rakentavat erilaisia representaatioita todellisuudesta. Tutkimuksen teoreettis-metodologisena viitekehyksenä on kriittinen diskurssintutkimus: kiinnostavaa on, miten somekohuista uutisoidaan ja miten uutisotsikossa rakennetaan merkityksiä, diskursseja ja valtarakenteita somekohun ympärille kuvatuista tapahtumista sekä toimijoi…
The activation of aggresomal pathway in Coxsackievirus B3 infection
2016
Transfection of lipoma cells with papilloma bovine virus subgenomic fragment.
1991
Abstract Lipoma cells with consistent chromosomal aberration have been transfected with plasmids carrying papilloma bovine virus subgenomic fragment (PBV 69). The succesful transformation of the cells was ascerted on the changed growth pattern of the cells in liquid medium, colony formation in soft agar and modified cell appearrance in electron microscopy; transfection with PBV 69 has not been, however, sufficient to immortalize lipoma cells.
