Identification and quantification of microbial populations in activated sludge and anaerobic digestion processes

Eight different phenotypes were studied in an activated sludge process (AeR) and anaerobic digester AnD) in a full-scale wastewater treatment plant by means of fluorescent in situ hybridization (FISH) and automated FISH quantification software. The phenotypes were ammonia-oxidizing bacteria, nitrite-oxidizing bacteria, denitrifying bacteria, phosphate-accumulating organisms (PAO), glycogen-accumulating organisms (GAO), sulphate-reducing bacteria (SRB), methanotrophic bacteria and methanogenic archaea. In vitro differentiation of E-N-CAM expressing rat neural precursor cells isolated by FACS during prenatal development

Most fetal rat brain cells expressing the embryonal, highly sialylated form of the cell adhesion molecule N-CAM (E-N-CAM) are precursor cells, as judged from the absence of marker molecules specific for mature neural cell types. However, the detection of E-N-CAM+ cells in frozen sections does not provide information on the lineage-specific differentiation of these cells during development. Jaunu Staphylococcus Epidermidis sortāzes A inhibitoru atlase izmantojot FRET skrīninga sistēmu

Pētījuma mērķis ir atrast jaunus zāļu kandidātus, kuri spēj inhibēt Staphylococcus epidermidis sortāzi A, vienlaicīgi mazinot patogēnu multirezistēnces izplatīšanos. Šim mērķim tika izveidoti ģenētiski DNS konstrukti, kuri kodē S.epidermidis sortāzes A katalītisko domēnu, iebūvējot pET22b un pET28a vektoros srtA gēnu. Rekombinantā sortāzes proteīna sintēze tika inducēta ar IPTG, izmantojot BL21(DE3) celmu Escherichia coli ekspresijas sistēmā. There is an urgent need of anti-virulence agents effective in the prevention or eradication of biofilms that are intrinsically resistant to conventional antibiotics. If we consider that the first step of staphylococcal pathogenesis and of biofilm formation is the bacterial adhesion, promoted by the surface exposed proteins at the cell wall, we believe that new anti-virulence agents could be developed by using as a target the Sortase A (SrtA), the enzyme responsible of linking surface exposed proteins to peptidoglycan. Five studies across a range of domains show that consumers who can choose as many alternatives as they wish (“flexible choice”), report more positive affective states and purchase more than those who have to choose a pre-defined quantity of products (“constrained” choice). Darba mērķis ir atrast jaunas novatoriskas vielas, kas inhibē Grampozitīvās baktērijas Streptococcus pneumoniae transpeptidāzi sortāzi A. Šo inhibitoru atlasei tika izmantota rekombinanta S.pneumoniae sortāzes A katalītisko domēnu kodējoša konstrukcija bez N terminālā reģiona. Šāda rekombinanta transpeptidāze tika ekspresēta E.coli ekspresijas sistēmā un sintezētais proteīns izrādījās šķīstošs. Pēc attīrīšanas sortāze tika izmantota inhibitoru atlasei, izmantojot skrīningu, kas balstās uz fluorescences rezonanses enerģijas pārnesi (FRET).
