Allosteric Models for Multimeric Proteins:  Oxygen-Linked Effector Binding in Hemocyanin

In many crustaceans, changing concentrations of several low molecular weight compounds modulates hemocyanin oxygen binding, resulting in lower or higher oxygen affinities of the pigment. The nonphysiological effector caffeine and the physiological modulator urate, the latter accumulating in the hemolymph of the lobster Homarus vulgaris during hypoxia, increase hemocyanin oxygen affinity and decrease cooperativity of oxygen binding. Homotropic and heterotropic allosteric interactions are important mechanisms that regulate protein function. These mechanisms depend on the ability of oligomeric protein complexes to adopt different conformations and to transmit conformation-linked signals from one subunit of the complex to the neighboring ones. Bohr-effect and buffering capacity of hemocyanin from the tarantula E. californicum.

A previous report showed that binding of oxygen to the 24-meric hemocyanin from E. californicum does not correlate linearly with the release of protons as known from hemoglobin. However, this unusual complex phenomenological observation could not be explained at that time. Here, I present a full analysis of the thermodynamic coupling between protons and oxygen for the 24-meric tarantula hemocyanin in Ringer-solution based on the Nested-MWC-model. Positive Allosteric Control of Guests Encapsulation by Metal Binding to Covalent Porphyrin Cages

The allosteric control of the receptor properties of two flexible covalent cages is reported. These receptors consist of two zinc(II) porphyrins connected by four linkers of two different sizes, each incorporating two 1,2,3‐triazolyl ligands. Silver(I) ions act as effectors, responsible for an on/off encapsulation mechanism of neutral guest molecules. Binding silver(I) ions to the triazoles opens the cages and triggers the coordination of pyrazine or the encapsulation of N,N′‐dibutyl‐1,4,5,8‐naphthalene diimide. Improved stability of solid state light emitting electrochemical cells consisting of ruthenium and iridium complexes

ABSTRACTTwo charged organometallic complexes containing bulky hydrophobic ligands based on ruthenium (II) and iridium (III) were synthesized and their performance in solid state light emitting electrochemical cells is described. The complexes were chosen as due to their large ligands a diminished susceptibility towards the formation of destructive complexes during device operation is expected. The LEC device performances reveal the longest living devices reported so far under dc bias. Quantum chemical calculations confirm that the major effect of the bulky diphenylphenanthroline ligands is of steric origin and not related with changes in the molecular electronic structure of the complexes.

Regulatory effects of polyamines on membrane-bound acetylcholinesterase

The effects of putrescene, spermidine and spermine on membrane-bound acetylcholinesterase from human erythrocyte ‘ghosts’ and the solubilized enzyme of the electric organ of the electric eel were studied by kinetic methods. Measurements were made by using a photometric method which made it possible to record the enzyme reaction in the steady-state phase. Substrate-concentration-dependent activation and inhibition of acetylcholinesterase by polyamines is similar to that by Na+, K+, Ca2+, Mg2+ and certain quaternary and bisquaternary amines.
