The contribution of tear osmolarity measurement to ocular surface assessment in contact lens wearers
2012
Purpose: To determine whether tear osmolarity using the TearLabTM Osmolarity System contributes to the assessment of the ocular surface in contact lens (CL) wearers. Methods: Data were collected from 44 CL wearers (28 tolerant and 16 intolerant) and 34 healthy subjects. Every patient underwent a thorough ophthalmic examination and tear osmolarity test, conjunctival impression cytology and meibomian lipid sampling. Symptoms, break-up time (BUT), tear osmolarity, conjunctival expression of HLA-DR and meibomian fatty acid composition were evaluated. Results: Tear osmolarity was significantly higher in the controls compared to tolerant and intolerant CL wearers (p=0.0007). Flow cytometry results …
An efficient esterification of pullulan using carboxylic acid anhydrides activated with iodine
2010
Received August 9, 2009Accepted January 12, 2010Published online February 11, 2010Acylation of alcohols (esterification) with anhydride is an important and routinely utilizedtransformation in organic synthesis. Iodine is a cheap and one of the most powerful anhy-dride activating agents for the esterification of alcohols. Therefore, an efficient and cost ef-fective method was developed for the acylation of pullulan polysaccharide by using differentacid anhydrides activated with iodine. A series of organo-soluble pullulan esters were suc-cessfully synthesized, purified and characterized. Under solvent free conditions, it was feasi-ble to fabricate pullulan nonaacetate and pullulan stearate. A…
Analysis of tear proteins by one- and two-dimensional thin-layer iosoelectric focusing, sodium dodecyl sulfate electrophoresis and lectin blotting. D…
1998
· Background: Isoelectric focusing (IEF) of tear proteins has not yet been carried out in a satisfactory way. Two-dimensional (2D) electrophoresis, especially in the combination of IEF with SDS, is able to differentiate between proteins in detail. The purpose of this study was therefore to analyze tear proteins by 1D IEF alone and in combination with a 2D pattern, and by IEF followed by lectin staining. · Methods: Ampholines, covering a broad range from pH 3 to pH 10, were applied. After IEF, semi-dry blotting and incubation with a group II lectin and two group V lectins was performed. · Results: Tear proteins could be separated into 31 single bands. Tear-specific pre-albumin (TSPA), lactof…
A metabolomic approach to dry eye disorders. The role of oral supplements with antioxidants and omega 3 fatty acids.
2014
Purpose We used nuclear magnetic resonance spectroscopy of hydrogen-1 nuclei (1H NMR S) to analyze the metabolic profile of reflex tears from patients with dry eye disorders. Methods We performed a prospective case-control study involving 90 participants: 55 patients diagnosed with dry eye syndrome (DESG) and 35 healthy subjects (control group, CG). From the DESG, two subgroups were formed: mild DES (n=22) and moderate DES (n=33). Participants were prescribed an oral nutraceutic supplementation containing antioxidants and essential polyunsaturated fatty acids to be taken as three capsules per day for 3 months. Reflex tears (20–30 µl) were collected from the tear meniscus of both eyes of eac…
Characterization of human reflex tear proteome reveals high expression of lacrimal proline-rich protein 4 (PRR4).
2014
In-depth studies on the proteome of reflex tears are still inadequate. Hence, further studies on this subject will unravel the key proteins which are conjectured to possess vital functions in the protection of the ocular surface. Therefore, this study investigated the differences in the expression levels in proteome of reflex compared to basal tears. Basal (n = 10) and reflex (n = 10) tear samples from healthy subjects were collected employing the capillary method, subsequently pooled and the proteomes were characterized employing 1DE combined with LC-ESI-MS/MS strategy for label-free quantitative (LFQ) analysis. The differentially expressed proteins were validated by 2DE combined with LC-E…
Changes in Tear Protein Pattern after Photorefractive Keratectomy
2003
PURPOSE. Changes in tear protein composition of patients who underwent photorefractive keratectomy (PRK) were analyzed. METHODS. Tear samples were obtained from 23 eyes of 23 patients immediately before PRK and on the fourth postoperative day with glass capillaries. Tear proteins were separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Digital image analysis and evaluation of the densitometric data of the electrophoretic separations were done with BioDoc-Analyze. RESULTS. Analysis of discriminance found a significant difference in the protein patterns (p<0.001). This type of analysis of the electrophoretic densitographs uses all peak information simultaneously. A signifi…
Proinflammatory Cytokine Profiling of Tears from Dry Eye Patients by Means of Antibody Microarrays
2011
In the pathogenesis of keratoconjunctivitis sicca, immune processes are thought to play an important role. However, the exact details of the pathomechanisms are still unknown. In this study, the expression patterns of proinflammatory cytokines in the tears of patients with different subtypes of dry eye were analyzed. One hundred forty-three subjects subdivided into healthy controls (CTRL, n = 38), patients with aqueous-deficient dry eye (DRYaq, n = 35), patients with changes of the lipid layer (DRYlip, n = 36), and patients with a combination of both (DRYaplip, n = 34) were examined. Expression patterns of proteins (e.g., IL-1β, IL-6, ITNF-α, and IFN-γ) were examined using an advanced antibo…
Gentamicin, norfloxacin and lysozyme concentration in human tears: in vivo and in vitro study
1992
Hen's egg lysozyme (HEL) activity was measured in vitro with gentamicin and norfloxacin by a turbidimetric technique. Gentamicin at the concentration of 10(-3) M inhibited HEL activity by 39%, while 10(-3) M norfloxacin did not affect HEL activity. However, an in vivo study in healthy persons did not show any significant statistical difference in tear lysozyme activity when 0.3% gentamicin or 0.3% norfloxacin were topically applied.
Characterization of lacrimal proline-rich protein 4 (PRR4) in human tear proteome
2014
This study was initiated considering the lack of comprehensive characteristics profile of PRR4 in tears of healthy subjects. Therefore, detailed characterizations of PRR4 from basal tears employing in-gel and in-solution digestions for MS systems are presented herein. First, pooled tear samples (n = 10) were utilized to identify PRR4-rich region/spots in 1DE/2DE gels employing LC-MALDI-MS and 1DE-LC-ESI-LTQ-Orbitrap-MS systems. PRR4-rich region and ten spots with vast polymorphisms (Mr : 17-30 kDa, pI: 3.0-6.6) were identified in 1DE and 2DE gels, respectively. In addition, combinations of four types of PTMs, which are methylation, acetylation, oxidation, and pyroglutamate formation, were i…
