A HPLC-fluorescence method for characterization of proteinaceous glues from binding media used in pictorial works of art prior to conservation or restoration treatment is proposed. Fluorescence derivatization of amino acids released by acid hydrolysis of standard proteins is studied. The derivatization reagent was o-phtalaldehyde with 2-mercaptoethanol as catalyst. Mobile phase was a programmed gradient among two eluents (water buffered at pH 5.8 wit 5% THF, and methanol) and is able to satisfactorily resolve the amino acid derivatives in 45min. The aim of this study was to assess the anaerobic biodegradability limits of urban wastewater with AnMBR technology by studying the possible degradation of the organic matter considered as non-biodegradable as observed in aerobic membrane bioreactors operated at very high sludge retention times. Abstract This study examined acid-catalyzed mechanocatalytic pretreatment of birch sawdust without a separate impregnation step. Catalyst amount and pretreatment time were the key variables. Pretreated material was mixed with water for hydrolysis (100 °C, 60 min). The efficient release of total reducing sugars from birch sawdust is significant to the path towards biofuels and biochemicals. Based on the results, the structure and surface of birch sawdust changed as a function of mechanocatalytic pretreatment. The nucleotide sequence of a new tRNA(Leu)(anticodon U*AA) from Saccharomyces cerevisiae which could recognize exclusively the UUA codon has been determined. Its primary structure is: pGGAGGGUUGm2GCac4CGAGDGmGDCDAAGGCm2(2)GGCAGACmUU*AAm1GA++ + psi CUGUUGGACGGUUGUCCGm5CGCGAGT psi CGm1A(orA)ACCUCGCAUCCUUCACCA. This tRNA has a large extraloop and contains 15 modified nucleotides. So far it is the third isoacceptor tRNA for leucine in yeast. It has 61% homology with tRNA(Leu)(anticodon m5CAA) and 63% homology with tRNA(Leu)(anticodon UAG), the two other known yeast tRNAs(Leu).

Enzymatic methods are sensitive and can be quite specific but they are subject to interference by inhibitors and not all are stoichiometric. We have investigated the potential of structural elucidation of O-linked glycopeptides by post-source decay matrix-assisted laser desorption ionization mass spectrometry (PSD-MALDI-MS). In order to establish detailed fragmentation patterns and to dissect fragment ions with and without carbohydrate content, the same O-linked MUC1-derived glycopeptides with acetylated and non-acetylated sugars were analysed and compared. Furthermore, we were interested to examine possible differences in the fragmentation between glycopeptides with acetylated and non-acetylated sugars. The attachment of carbohydrates to the peptide backbone influences the conformation as well as the solubility of the conjugates and can even be essential for binding to specific ligands in cell-cell interactions or for active transport over membranes. The interest in tumor-associated glycoconjugate antigens was particularly initiated by Springer, who published in 1984 that glycoproteins on the outer cell-membrane of epithelial tumor cells have an altered glycosylation consisting of the Thomsen-Friedenreich (T-) antigen and its precursor the TN-antigen structure (Springer 1984).
