Serological and virological evidence of non-sexual transmission of human herpesvirus type 8 (HHV8).

To evaluate whether or not human herpesvirus 8 (HHV8) can be transmitted through a non-sexual route a serological survey was carried out in a group of 51 catholic nuns. The seroprevalence rate and the geometrical mean antibody titre to anti-latent HHV8 antigen were similar in nuns and in a group of 60 women, matched by age, in the general population (27 vs. 24%; 1028 vs. 1575, respectively). Moreover, by using nested polymerase chain reaction (PCR), HHV8 DNA sequences were detected in 7 of 16 (43·8%) saliva and peripheral blood mononuclear cells (PBMC) from patients with classical Kaposi's sarcoma (KS) and in 3 out of 7 (42%) AIDS-KS patients. Amino acids requirements of measles virus in HeLa cells.

A low plating efficiency for measles virus was obtained when arginine, threonine and isoleucine were omitted from Eagle's basal medium and a stimulatory effect on the plaque production was produced by increasing the concentration up to 0.5 mm of arginine and glutamine. However, of all amino acids in Eagle's basal medium, only methionine was found to be essential for the synthesis of infectious progeny virus. Uridine enhances the cytotoxic effect of D-glucosamine in rat C6 glioma cells.

This paper studies the influence of uridine on the effects exerted by D-glucosamine in rat C6 glioma cells. 2 mM uridine increased markedly both the cytotoxic effect of the aminosugar and the inhibition of thymidine incorporation into acid-insoluble fraction. Furthermore the complete resumption of the capacity to incorporate either 3H-thymidine or 3H-mannose which was observed after the removal of the aminosugar, was impeded when the cells were treated contemporaneously with D-glucosamine and uridine. An exposure for 4 hr to 20 mM glucosamine alone enhanced about 15-fold the cellular pool of UDP-N-acetylhexosamines; the addition of 2 mM uridine intensified the expansion of this pool, which …

Inhibition of growth of measles virus by mycoplasma in cell-cultures and the restoring effect of arginine

Presence of mycoplasma in MK cell cultures caused an inhibition of measles virus, as revealed by a lower plating efficiency. When arginine is supplemented to the cell-system, mycoplasma contaminated cells yield plaque titers as high as those of uncontaminated cells. The essential role of arginine for plaque formation of measles virus is suggested also by experiments in which arginine added to the agar overlay restored the plating efficiency of measles virus in cultures maintained in a minimal nutrient medium.

Associations of classic Kaposi sarcoma with common variants in genes that modulate host immunity

AbstractClassic Kaposi sarcoma (CKS) is an inflammatory-mediated neoplasm primarily caused by Kaposi sarcoma–associated herpesvirus (KSHV). Kaposi sarcoma lesions are characterized, in part, by the presence of proinflammatory cytokines and growth factors thought to regulate KSHV replication and CKS pathogenesis. Using genomic DNA extracted from 133 CKS cases and 172 KSHV-latent nuclear antigen-positive, population-based controls in Italy without HIV infection, we examined the risk of CKS associated with 28 common genetic variants in 14 immune-modulating genes. HUMAN HERPESVIRUS 8 SEROPREVALENCE AMONG INTERNATIONALLY ADOPTED CHILDREN COMING FROM EASTERN EUROPE

The seroprevalence of Human Herpes Virus 8 (HHV8) and its transmission pattern were assessed testing serum samples of 120 internationally adopted children (aged 1-15 years) coming from Eastern Europe. Determinations of IgG antibodies against both latent and lytic HHV-8 antigens were performed by indirect immunofluorescence assay. Antibodies were detected only for lytic antigen of the virus in 12.5% of children with a seroprevalence significatively higher (19.6%) in young children (age 1-6). No correlation was observed between HHV8 seropositivity and serological markers for hepatitis A, B and C viruses and Human Immunodeficiency virus. Background: The presence of Human Herpesvirus-8 (HHV8) DNA is predictive of Kaposi sarcoma (KS) among patients with HIV-associated or iatrogenic immunosuppression. However, correlates of HHV8-DNA detection in the general population remain undefined. Methods: We assessed correlates of HHV8-DNA detection among Italian adults without KS who had antibodies against HHV8-latent nuclear antigen by immunofluorescence assay.
