Dynamik des Ketonkörperstoffwechsels bei gesunden Ratten

Bis zu 150 μMol Acetacetat/kg/min besteht bei gesunden Ratten eine lineare Beziehung zwischen Infusionsrate und Blutspiegelanstieg an Ketonkorpern. Zunahme der Infusionsrate um 10 μMol Acetacetat/kg/min bewirkt einen Blutspiegelanstieg um 0,072 μMol Gesamtketonkorper/ml. Die totale Clearance fur Gesamtketonkorper betragt 139±17 ml · kg−1 · min−1. Uberschreitet die Zufuhr 150 μMol · kg−1 · min−1, so steigen die Ketonkorperspiegel rasch auf uberhohte Werte an. Aus den Daten lassen sich endogene Produktion und Verwertung von Ketonkorpern fur jeden stationaren Blutspiegel innerhalb der Umsatzkapazitat berechnen.

The effect of arabinofuranosyl-cytosine upon the synthesis of herpesvirus hominis Electron microscopic observations in relation to viral DNA-synthesis

The paper describes experiments about the degree of dependency of capsid, envelope and antigen synthesis byherpesvirus hominis upon viral DNA synthesis. The DNA synthesis has been blocked by different doses of Ara-C and the remnant DNA-synthesis has been measured by [3H]-thymidine incorporation after CsCl-density gradient centrifugation. Electron microscopic studies were done in parallel after incubation of infected cells with different doses of Ara-C. Finally, antigens were prepared after infection without and with added Ara-C. The biosynthesis of herpesvirus DNA in rabbit kidney cells is inhibited to 50% by PEA (2-Phenylethanol) at 0.65 mg PEA/ml. The inhibition of cellular DNA synthesis in uninfected cells by PEA is about twice as sensitive as that of viral DNA synthesis.

The effect of chemically modified deoxyribonucleic acid (DNA) on enzymatic DNA-synthesis in vitro has been investigated with analogues of DNA prepared without either purine bases (apurinic acid, APA) or pyrimidine bases (apyrimidinic acid, APyA). DNA-synthesis in vitro was studied with the deoxynucleotide polymerizing enzymes purified from calf thymus (replicative and terminal deoxynucleotidyltransferase respectively).

Influence of molecular weight of DNA on the determination of anti-DNA antibodies in systemic lupus erythematosus (SLE) sera by radioimmunoassay

Using a radioimmunoassay (RIA) based on the Farr technique with radioactively labeled 3-H-DNA for quantitative measurements of anti-DNA antibodies in sera of patients with systemic lupus erythematosus (SLE), the influence of molecular weight of DNA (ranging from 0.1 times 10-6 to 22.0 times 10-6 daltons) on binding and precipitation in this system has been investigated. Comparing our results with mathematical models it follows that one antibody molecule is fixed on the average to a statistical DNA segment of 2 times 10-6 to 4 times 10-6 daltons.
