We report in this paper on isolation and characterization of two unknown nucleosides G* and [A*] located in the T-psi-stem of yeast methionine initiator tRNA, using the combined means of HPLC protocols, real time UV-absorption spectrum, and post-run mass spectrometry by electron impact or fast atom bombardment. The G* nucleoside in position 65 was identified as unmodified guanosine. Abstract In this work, a monolithic polymer based on poly(methacrylic acid-co-ethylene glycol dimethacrylate) (MAA-co-EDMA) was prepared inside 200 μL pipette tips for the extraction of drug of abuse from oral fluid samples. After an appropriate surface tip modification, several polymerization mixtures with different monomer/cross-linker ratios, and percentage of porogen were studied. The most appropriate monolith to easily flow organic solvents and oral fluid samples was prepared with a MAA/EDMA ratio of 8:92 wt/wt and dodecanol containing 10 wt% toluene, as porogenic solvent. Most nanomaterials acquire a protein corona upon contact with biological fluids. The magnitude of this effect is strongly dependent both on surface and structure of the nanoparticle. New background correction approach based on polynomial regressions for on-line liquid chromatography-Fourier transform infrared spectrometry.

Abstract In the present study a new approach for the chemometric background correction in on-line gradient LC–FTIR is introduced. For this purpose, the spectral changes of the elution mixture during gradient elution were analyzed applying 2D correlation spectroscopy. The fundamentals of the new background correction algorithm, based on polynomial fits calculated from a reference spectra matrix (Polyfit-RSM method) are explained. The cocoa varieties Criollo, Forastero and Trinitario, which have different organoleptic properties, quite often are mixed together in cocoa-based products. The objective of this work was to develop a rapid method for the simultaneous determination of polyphenols (n = 35) and alkaloids (n = 2) in cocoa-based products by using the high throughput advantages provided by the Q Exactive Hybrid Quadrupole-Orbitrap Mass Spectrometer (FIRMS). An extensive survey of wild Sicilian oregano was made. A total of 57 samples were collected from various sites, followed by taxonomic characterization from an agronomic perspective. Based on morphological and production characteristics obtained from the 57 samples, cluster analysis was used to divide the samples into homogeneous groups, to identify the best biotypes. All samples were analyzed for their phytochemical content, applying a cascade-extraction protocol and hydrodistillation, to obtain the non volatile components and the essential oils, respectively. Relative contribution of different l-arginine sources to the substrate supply of endothelial nitric oxide synthase

In certain cases of endothelial dysfunction l-arginine becomes rate-limiting for NO synthesis in spite of sufficiently high plasma concentrations of the amino acid. To better understand this phenomenon, we investigated routes of substrate supply to endothelial nitric oxide synthase (eNOS). We have taken advantage of our previously reported high performance liquid chromatographic (HPLC) strategy to investigate the conformational behavior of the optically reversed gramicidin M (gM-), an analog of gramicidin A with all tryptophans replaced by phenylalanines, in different model membranes.
