Preparation of extracts from mature spruce needles for enzymatic analyses

It was possible to extract simultaneously several active enzymes involved in the carbohydrate or the amino acid metabolism from spruce needles [Picea abies (L.) Karst.] when a) a 100 mM Na-Pi buffer of pH 7.5 containing 5% PVPP and 0.5% Triton X-100 was used and when b) the resulting crude extracts were freed from lowmolecular-weight compounds by gel-chromatography using the separation medium Fractogel TSK HW-40. Besides Triton X-100, Triton X-305, Myrij-52 and Brij-35 were tested, but 0.5% Triton X-100 brought about the most active enzyme extracts. n-3 Polyunsaturated fatty acids (PUFA) are considered to be authentic immunosuppressors and appear to exert beneficial effects with respect to certain immune-mediated diseases. In addition to promoting T-helper 1 (Th1) cell to T-helper 2 (Th2) cell effector T-cell differentiation, n-3 PUFA may also exert anti-inflammatory actions by inducing apoptosis in Th1 cells. With respect to mechanisms of action, effects range from the modulation of membrane receptors to gene transcription via perturbation of a number of second messenger cascades. Two ATP molecules are used in the reaction catalyzed by carbamoyl-phosphate synthetase I. One molecule (ATPA) phosphorylates HCO3- and the other (ATPB) phosphorylates carbamate. Carbamoyl-phosphate synthetase I is a 160-kDa polypeptide consisting of a 40-kDa N-terminal moiety and a 120-kDa C-terminal moiety, the latter being composed of two similar halves of molecular mass 60 kDa. The comparative behavior of Lactococcus lactis in free and immobilized culture processes

This study presents the changes in the physiological state of Lactococcus lactis in different actively growing systems. The intracellular pH (pHin), NADH/NAD ratio and lactate dehydrogenase (LDH) activity were measured in free cell batch culture with or without external pH (pHout) control, and in continuous free or immobilized cell reactors. In free cell batch culture it has been shown that the pHin is dependent both on pHout and age of the culture, while in free cell continuous culture the pHin and the pHout are close together. Inorganic polyphosphates (polyP) are linear polymers of orthophosphate (Pi) residues linked by high-energy phosphoanhydride bonds. These polymers are widely distributed in nature, from archaebacteria, eubacteria, fungi, algae, and protozoa to higher plants and animals (for reviews, see Kulaev 1979; Wood and Clark 1988; Kornberg 1994, 1995). PolyP molecules are stable in neutral aqueous solutions, but are hydrolyzed by heat-treatment, and under acidic or alkaline conditions. Abstract Marine sponges as sessile filter feeders are inevitably under a constant influence of changes in their environment. Mediation of extracellular signals and regulation of cellular response to environmental stress is a key function of cellular protein kinases. Expression, proteolytical cleavage and phosphorylation of stress-responsive KRS_SD protein kinase, in control and tributyl-tin (TBT) treated sponges were investigated. Females live longer than males. This could be in part due to the higher levels of estrogens in females, which protect them against aging. Physiological concentrations of estrogens have antioxidant effects as they induce the expression of manganese superoxide dismutase and glutathione peroxidase by stimulating estrogen receptors and the mitogen-activated protein kinase and nuclear factor kappa B pathways. However, estrogens can have undesirable effects such as they are feminizing to males, so other alternatives need to be searched. Electrophilic substitution and cyclization of 2,2′-bis(N-methylindolyl): A simple access to potential protein kinase C inhibitor

A strategy is described for the synthesis of functionalized and cyclized 2,2′-bisindolyl derivatives related to several basic systems of natural products. The starting 2,2′-bis(N-methylindolyl) (8) reacts with a variety of electrophiles and electrophilic dienophiles to furnish the novel, functionalized and cyclized bisindolyl derivatives 9–16. In addition, some reactivity and structural aspects are discussed; an X-ray crystallographic analysis of the 2,2′-bisindolyl 8 provided valuable information for the conformational analyses.
