Selection of single-chain antibodies against the VP8* subunit of rotavirus VP4 outer capsid protein and their expression in Lactobacillus casei.

2004

ABSTRACTSingle-chain antibodies (scFv) recognizing the VP8* fraction of rotavirus outer capsid and blocking rotavirus infection in vitro were isolated by phage display. Vectors for the extracellular expression inLactobacillus caseiof one of the scFv were constructed.L. caseiwas able to secrete active scFv to the growth medium, showing the potential of probiotic bacteria to be engineered to express molecules suitable for in vivo antirotavirus therapies.

Single-chain variable fragment (scFv) antibodies against rotavirus NSP4 enterotoxin generated by phage display.

2004

The rotavirus non-structural NSP4 protein causes membrane destabilization as well as an increase in intracellular calcium levels in eukaryotic cells and induces diarrhea in young mice, acting as a viral enterotoxin. In this study the phage display technique was used to generate a panel of single-chain variable fragment (scFv) antibodies specific for the NSP4 protein of the human rotavirus strain Wa from a human semi-synthetic scFv library. After several rounds of panning and selection on NSP4 adsorbed to polystyrene tubes, individual scFv were isolated and characterised by fingerprinting and by sequencing the VH and VL genes. The isolated scFv antibodies specifically recognize NSP4 in enzym…

Investigating Potential Alignments between Modelica Standard Library and SAREF Ontologies

2021

International audience; Simulation tools based on the Modelica language provide comprehensive modelling and simulation approaches for building energy systems. However, the simulation and optimisation of such systems are data-driven processes, lacking a common understanding of information structure within the process. This paper investigates the possible semantic alignments of the Smart Appliances REFerence (SAREF) ontology and its extension for building domain, SAREF4BLDG, with the Modelica Standard Library (MSL). Using the MSL, a residential heating system has been modelled in OpenModelica, an open-source modelling and simulation environment. Then, SAREF and its extension SAREF4BLDG semant…

What to Expect and What to Focus on in SQL Query Teaching

2019

In the process of learning a new computer language, writing erroneous statements is part of the learning experience. However, some errors persist throughout the query writing process and are never corrected. Structured Query Language (SQL) consists of a number of different concepts such as expressions, joins, grouping and ordering, all of which by nature invite different possible errors in the query writing process. Furthermore, some of these errors are relatively easy for a student to fix when compared to others. Using a data set from three student cohorts with the total of 744 students, we set out to explore which types of errors are persistent, i.e., more likely to be left uncorrected by…

Molecular cloning and characterization of a Candida albicans gene coding for cytochrome c haem lyase and a cell wall-related protein.

1998

Immunoscreening of a Candida albicans cDNA library with a monoclonal antibody (mAb 4C12) recognizing an epitope present in high-molecular-weight mannoprotein (HMWM) components specific for the mycelial cell walls (a 180 kDa component and a polydispersed 260 kDa species) resulted in the isolation of the gene CaCYC3 encoding for cytochrome c haem lyase (CCHL). The CaCYC3 gene was transcribed preferentially in mycelial cells in which two mRNA transcripts of 0.8 and 1 kb were found. The nucleotide and the deduced amino acid sequences of this gene displayed 45% homology and 46% identity, respectively, to the Saccharomyces cerevisiae CYC3 gene and shared common features with other reported genes …

Oral toxicity produced by chemotherapy: A systematic review

2014

Introduction: Antineoplastic chemotherapy remains one of the most widely used management strategies in cancer, either alone or in combination with other types of treatment. The main inconvenience of chemotherapy is its lack of selectivity, since it acts upon both tumor cells and rapidly multiplying normal cells such as bone marrow cells, hair follicle cells and oral and gastrointestinal mucosal cells. Material and method: An exhaustive search was made of the main oral toxic effects of chemotherapy in the PubMed-Medline, Cochrane Library and Scopus databases. A total of 1293 articles were identified, of which 333 met the study inclusion criteria. Results: The toxic effects of chemotherapy at…

De Novo Assembly and Functional Annotation of the Olive (Olea europaea) Transcriptome

2013

Olive breeding programmes are focused on selecting for traits as short juvenile period, plant architecture suited for mechanical harvest, or oil characteristics, including fatty acid composition, phenolic, and volatile compounds to suit new markets. Understanding the molecular basis of these characteristics and improving the efficiency of such breeding programmes require the development of genomic information and tools. However, despite its economic relevance, genomic information on olive or closely related species is still scarce. We have applied Sanger and 454 pyrosequencing technologies to generate close to 2 million reads from 12 cDNA libraries obtained from the Picual, Arbequina, and L…

A Dynamic Attribute-Based Authentication Scheme

2015

Attribute-based authentication (ABA) is an approach to authenticate users by their attributes, so that users can get authenticated anonymously and their privacy can be protected. In ABA schemes, required attributes are represented by attribute trees, which can be combined with signature schemes to construct ABA schemes. Most attribute trees are built from top to down and can not change with attribute requirement changes. In this paper, we propose an ABA scheme based on down-to-top built attribute trees or dynamic attribute trees, which can change when attribute requirements change. Therefore, the proposed dynamic ABA scheme is more efficient in a dynamic environment by avoiding regenerating…

A Performance-Based Test for Assessing Students’ Online Inquiry Competences in Schools

2018

In this paper, we introduce a performance-based test for measuring adolescents’ competences in online inquiry. The test covers four competence dimensions: (1) searching and selecting relevant sources, (2) identifying the main ideas presented in the sources, (3) evaluating the credibility of the sources, and (4) synthesizing information across the sources. We implement a technological solution called NEURONE to carry out this routine. The scoring of the test data is demonstrated by presenting preliminary results of a case study. Finally, we discuss the strengths and limitations of the test.
