This chapter summarizes the influence of polyelectrolyte topology on biological functions and biomedical applications such as cell uptake, drug delivery, and gene transfection. Polyelectrolytes utilized are spherical structures derived from dendrimers and albumin or cylindrical brushes, all of which are decorated with various polypeptide chains.

Two CuII complexes with a new quinoline sulfonamide derivative and phenanthroline (phen), [Cu(QSQ)(phen)]ClO4·0.5H2O (1) and [Cu(QSQ)(phen)(H2O)]ClO4 (2) [HQSQ = N-(quinolin-8-yl)quinolin-8-sulfonamide], have been synthesized and physicochemically characterized. Single-crystal X-ray diffraction studies have revealed a highly distorted trigonal-bipyramidal structure for 1 (τ = 0.68) and an almost perfect trigonal-bipyramidal geometry for 2 (τ = 0.92). The photosensitizing potential of drugs must be related to their photoreactivity towards the target biomolecules. In this context, a representative photosensitizing drug (tiaprofenic acid) was co-irradiated with a model protein, bovine serum albumin (BSA). This led to a significant degree of protein crosslinking and to the formation of trace amounts of drug-BSA photoadducts. Amino acid analysis of the hydrolysed (HC1) protein showed that His and Tyr undergo a dramatic decrease (approx. 90%) as a consequence of drug-mediated photodynamic processes. Abstract The thermal ring-opening reaction of a negative photochronic spirobenzopyran was investigated in the presence and absence of albumins. In the presence of the proteins, the formation of the merocyanine form from the spiro form is enhanced markedly by two orders of magnitude. The results for five different albumins indicate the enzyme-like activity of the proteins. The influence of pH, temperature and various ligands was examined in detail for bovine serum albumin (BSA) and human serum albumin (HSA). Drug carriers were prepared by combining oligopeptidic sequences with synthetic polymeric chains. Soluble copolymers of N-(2-hydroxypropyl)methacrylamide (HPMA) were synthesized containing oligopeptidic side chains terminating in a drug model (p-nitroaniline). The susceptibility of these oligopeptidic sequences to degradation on incubation with lysosomal enzymes and human serum was evaluated by monitoring the p-nitroaniline liberation. It was shown that the drug model can be attached to the polymer carrier by means of an oligopeptidic spacer stable in blood serum and degradable after the polymer has been in contact with intracellular lysosomal enzymes. 1,4 dihidropiridīnu atvasinājumi ir ļoti perspektīvi savienojumi. Pēc jaunāko pētījumu datiem, 1,4 dihidropiridīnu atvasinājumiem piemīt antimutagēnas īpašības, kā arī DNS reparāciju veicinošas spējas. Pašlaik šie savienojumi tiek ļoti plaši pētīti. All-trans to 11-cis retinol isomerization in nuclear membrane fraction from bovine retinal pigment epithelium

Abstract Isomerization of all-trans to 11-cis retinol has been studied in a membrane preparation from the nuclear fraction of bovine retinal pigment epithelium. When the nuclear membrane preparation deprived of endogenous retinoids is incubated with 4·5 μ m all-trans-retinol, the mean value calculated for the isomerase activity is 1·32 nmol 11-cis retinol formed hr−1 mg protein−1. Simultaneous formation of all-trans and 11-cis retinyl esters is also observed in the nuclear preparation. When assayed under the same experimental condition, RPE 150 000 g post-nuclear sediment shows about 70% of the isomerase activity found in the nuclear membrane fraction.
