Molecular mechanisms involved in the regulation of stress response in Oenococcus oeni and experimental evolution

Oenococcus oeni is the main bacterium responsible of malolactic fermentation in wine. This lactic acid bacteria grow in the stressful wine environment (high ethanol content, sulfites, low pH and low temperatures….). To maintain its cellular homeostasis, O. oeni has established mechanisms of resistance to its ecological niche. This research work focuses on the adaptative response of O. oeni to its environment and especially to acidity. Mass spectral identification of the blocked N-terminal tryptic peptide of the ATPase inhibitor from beef heart mitochondria

AbstractThe presence of a formyl blocking group at the N-terminus of the ATPase inhibitor has been identified and the partial sequence of the N-terminal peptide has been determined by fast atom bombardment and field desorption coupled to mass spectrometry. Minor discrepancies in amino acid sequence of the inhibitor between the present and published data [(1981) Proc. Natl. Acad. Sci. USA 78, 7403-7407] are reported and its relationships with other inhbitors are briefly discussed.

The plasma membrane ATPase of Kloeckera apiculata: purification, characterization and effect of ethanol on activity

Partially (6-fold) purified plasma membrane ATPase from an ethanol-sensitive yeast, Kloeckera apiculata, had an optimum pH of 6.0, an optimum temperature of 35°C, a K m of 3.6 mM ATP and a V max of 11 μmol Pi/min.mg protein. SDS-PAGE of the semi-purified plasma membrane showed a major band of 106 kDa. No in vivo activation of the ATPase by glucose was observed. Although 4% (v/v) ethanol decreased the growth rate by 50% it did not affect the ATPase. Concentrations of ethanol ≥2% (v/v) did, however, inhibit the enzyme in vitro. The characteristics of the enzyme did not change during growth in the presence of ethanol.

Nucleotide sequence of plasmid p4028, a cryptic plasmid from Leuconostoc oenos.

Abstract TheLeuconostoc oenosplasmid p4028 was cloned in pBlueScript (SK+), and its complete nucleotide sequence was determined. The analysis of the nucleotide sequence revealed five open reading frames, all of them located on the same strand and grouped in two clusters separated by a short noncoding stretch. A similarity search against the other sequences deposited in the EMBL and GenBank databases showed that p4028 has no significant similarity with any of the sequences checked. Nevertheless, a putative ATP-binding motif was found in ORF2. A more detailed analysis of this ORF suggests that it could encode for a DNA-dependent ATPase.

Attenuated Carbohydrate and Gill Na+ , K+-ATPase Stress Responses in Whitefish Caged near Bleached Kraft Mill Discharges

Exposure to biologically treated bleached kraft mill effluent (BKME) is demonstrated to greatly modify the acute physiological stress response in fish and, accordingly, to lead to inconsistencies in data interpretation due to dissimilar effects of handling procedures on reference and exposed fish. Cadmium, mercury, and lead effects on gill tissue of freshwater crayfishProcambarus clarkii (girard)

Intermolt adult crayfish P. clarkii were used for this work. After acclimatization to laboratory conditions crayfish were exposed to sublethal concentrations of cadmium, mercury, and lead for 96 h. Gills of control and exposed crayfish were removed and ATPase activity and oxygen uptake rate were determined. Structural damage of gill filaments was also observed. Gill tissue respiration rates were measured for individual crayfish using a Gilson differential respirometer. Lead causes a decrease of gill oxygen uptake, but neither cadmium nor mercury seems to affect it at the concentrations employed. Inhibition of Gill Na+,K+-ATPase Activity in the Eel,Anguilla anguilla,by Fenitrothion

European eels (Anguilla anguilla) were exposed to sublethal fenitrothion concentrations (0.02 and 0.04 mg/liter) in a continuous flow-through system for 4 days. Gill Mg2+- and Na+,K+-ATPase activities were evaluated after 2, 8, 12, 24, 32, 48, 56, 72, and 96 h of pesticide exposure. Results indicated that ATPase activity in gill tissue decreased as concentration of fenitrothion increased. Pesticide induced significant inhibitory effects on the Na+, K+-ATPase activity of A. anguilla, ranging from >56% inhibition at a sublethal concentration of 0.02 ppm to >73% inhibition at a sublethal concentration of 0.04 ppm. Eel ATPase activity as biomarker of thiobencarb exposure

Abstract European eels ( Anguilla anguilla ) were exposed to a sublethal thiobencarb concentration of 0.22 mg/L in a flow-through system for 96 h. Mg 2+ and Na + –K + adenosine triphosphatase (ATPase) activities were evaluated in gill and muscle tissues at 2, 12, 24, 48, 72, and 96 h of thiobencarb exposure. Gill ATPase activities were rapidly inhibited from 2 h of contact onward. Highest inhibition was registered for Na + , K + -ATPase (85%) from 2 to 12 h. Both Mg 2+ and total ATPase were inhibited (>73%) during the first hours of toxicant exposure. ABSTRACT Hepatitis B virus (HBV) is an enveloped DNA virus that presumably buds at intracellular membranes of infected cells. HBV budding involves two endocytic host proteins, the ubiquitin-interacting adaptor γ2-adaptin and the Nedd4 ubiquitin ligase.
