A comparison of the behaviour on the nitrosation of the isomers 5-(2-methoxyphenyl)- (1a) 5-(3-methoxyphenyl)- (1b) and 5-(4-methoxyphenyl)2,3-diphenylpyrroles (1c) carried out in the usual way with iso-amyl nitrite in a solution of sodium ethoxide in ethanol evidences that 1a is dramatically less reactive with respect to 1b and 1c. The different reactivity was ascribed to the occurrence of a strong hydrogen bond involving pyrrole NH and the ortho-methoxy group.

The electrophilic/nucleophilic character of a series of captodative (CD) ethylenes involved in polar cycloaddition reactions has been studied using DFT methods at the B3LYP/6-31G(d) level of theory. The transition state structures for the electrophilic/nucleophilic interactions of two CD ethylenes toward a nucleophilically activated ethylene, 2-methylene-1,3-dioxolane, and an electrophilically activated ethylene, 1,1-dicyanoethyelene, have been studied, and their electronic structures have been characterized using both NBO and ELF methods. I read with great interest the recent article from Fomon1 in which he refers to the well-known hypothesis that views the variable frequencies of lactase persistence in different human populations and, consequently, the possibility for some adults to feed on milk (lactose tolerance) as an advantageous evolutionary trait that has been genetically determined and brought about through centuries of natural selection. Before embarking on a detailed discussion of the titanation of carbanions, synthetic and physical organic aspects of several typical organotitanium (IV) compounds shall be surveyed, including thermal stability, aggregation state, X-ray structural data and bond energies. Some of this information is useful in understanding reactivity and selectivity in reactions with organic substrates. Benzo[a]quinolizine is an important heterocyclic framework that can be found in numerous bioactive compounds. The general scheme for the synthesis of these compounds was based on the preparation of the appropriate dihydroisoquinolines by Bischler-Napieralski cyclization with good yields, followed by the Pemberton method to form the oxazinones or pyridones derivatives via acyl-ketene imine cyclocondensation. In the title compound, C17H15FN2O, the exocyclic bond angles at the C atoms of the isoxazole ring bearing the pyridyl and 4-fluorophenyl substituents are 129.66 (17) and 134.58 (16)°, respectively. The structure was determined in a study of the molecular geometry of isoxazole derivatives with biological activity as MAPK inhibitors.

Silyl isoxazolines have been synthesized by [2+3] cycloaddition reaction of nitrile oxides to vinyl- and allylsilanes. The addition of 3-pyridylnitrile oxide to 1,3-divinyl-1,1,3,3-tetraphenyldisiloxane affords 1,3-bis{5-[3-(3-pyridyl)isoxazolin-2-yl]}-1,1,3,3-tetraphenyldisiloxane; the latter exists as a mixture of trans- and cis-isomers.The bond angle of the Si–O–Si fragment in thetrans-isomer equals 180(3)° and in the cis-isomer it is 162(3)°.The pharmacological properties of 4-[3-(5-trimethylsilylisoxazolin-2-yl)]pyridinium-chloride have been studied.
New Synthesis oftrans-Disubstituted Cyclam Macrocycles – Elucidation of the Disubstitution Mechanism on the Basis of X-ray Data and Molecular Modeling

A new way to synthesize trans-disubstituted cyclam tetraazamacrocycles 1 is reported. The synthesis proceeds in three steps via the tricyclic 1,4,8,11-tetraazatricyclo[9.3.1.14,8]hexadecane system 2, which can be selectively dialkylated and hydrolyzed under basic conditions to give the final product 1. An understanding of the reactivity, based on the X-ray experimental electrostatic potential and molecular modeling of the 1,4,8,11-tetraazatricyclo[9.3.1.14,8]hexadecane macrotricycle, has permitted the elucidation of a new reaction pathway leading to the trans-disubstituted cyclam.
