Styryl-pyrones from Goniothalamus arvensis

Two novel styrl-pyrones, (+)-garvensintriol and (+)-etharvendiol, together with a known cytotoxic furano-furone, (+)-goniofufurone, have been isolated from the stem bark of Goniothalamus arvensis. A different relative configuration, cis-erythro-erythro for garvensintriol and cis-threo-erythro for etharvendiol, is established, and their absolute stereochemistry is discussed.

3-acetylaltholactone and related styryl-lactones, mitochondrial respiratory chain inhibitors.

A novel furano-pyrone, 3-acetylaltholactone, and two other known styryl-lactones, altholactone and 5-acetoxyisogoniothalamin oxide, have been isolated from Goniothalamus arvensis (Annonaceae) stem bark. We report here the isolation and structural elucidation of these compounds with furane-pyrone and styryl-pyrone skeletons, postulating also for the first time their mechanism of cytotoxicity based on inhibition on mammalian mitochondrial respiratory chain.

The Annonaceae includes cultivated species of economic interest and represents an important source of information for better understanding the evolution of tropical rainforests. In phylogenetic analyses of DNA sequence data that are used to address evolutionary questions, it is imperative to use appropriate statistical models. Annonaceae are cases in point: Two sister clades, the subfamilies Annonoideae and Malmeoideae, contain the majority of Annonaceae species diversity. Phytochemical investigations of ethanol root bark and stem bark extracts of Cleistochlamys kirkii (Benth.) Oliv. (Annonaceae) yielded a new benzopyranyl cadinane-type sesquiterpene (cleistonol, 1) alongside 12 known compounds (2–13). The structures of the isolated compounds were established from NMR spectroscopic and mass spectrometric analyses. Structures of compounds 5 and 10 were further confirmed by single crystal X-ray crystallographic analyses, which also established their absolute stereochemical configuration. Vasodilator Effects of Liriodenine and Norushinsunine, Two Aporphine Alkaloids Isolated from &lt;i&gt;Annona cherimolia,&lt;/i&gt;in Rat Aorta

The effect of two aporphines, liriodenine and norushinsunine, isolated from Annona cherimolia, were studied in the rat aorta in order to examine their mechanism of action. Both alkaloids (10–7–10–4 mol/l) showed relaxant effects on the contractions elicited by 10–6 mol/l noradrenaline (NA) or 80 mmol/l KC1, but, while liriodenine showed a nonspecific relaxant action on both spasmogens, norushinsunine was more potent on KC1-induced contraction. In Ca2+-free medium, both alkaloids (0.1 mmol/l) inhibited the responses elicited by NA, but not those elicited by caffeine. Acetogenins, Aporphinoids, and Azaanthraquinone from Annona cherimolia Seeds.

We have isolated several known steroids, glycosides, isoquinoline, and azaanthraquinone alkaloids, and five acetogenins belonging to a new group of secondary metabolites, the bis-tetrahydrofuran gamma-lactones, from the seeds of ANNONA CHERIMOLIA. The azaanthraquinone alkaloid isolated was identified as cleistopholine. One new acetogenin is reported here: laherradurine; a second new one was isolated, but its structure has not been determined. The other three: cherimoline, dihydrocherimoline, and asimicine have already been reported.
