Functional recovery after ischemic stroke largely involves brain plasticity and more accurately its ability to reorganize the neuronal networks spared by the infarct. BDNF and nitric oxide signaling both contribute to plasticity at glutamatergic synapses. However, the role of combined signaling of both pathways at the same synapse is largely unknown. Using NO imaging with diaminofluoresceine in cultured hippocampal neurons we analyzed the time course of neurotrophin-induced NO signals. Application of exogenous BDNF, NT-4, and NT-3 (but not NGF) induced NO signals in the soma and in proximal dendrites of hippocampal neurons that were sensitive to NO synthase activity, TrkB signaling, and intracellular calcium elevation. BDNF as a Mediator of Antidepressant Response: Recent Advances and Lifestyle Interactions

Conventional antidepressants are widely employed in several psychiatric and neurologic disorders, yet the mechanisms underlying their delayed and partial therapeutic effects are only gradually being understood. This narrative review provides an up-to-date overview of the interplay between antidepressant treatment and Brain-Derived Neurotrophic Factor (BDNF) signaling. In addition, the impact of nutritional, environmental and physiological factors on BDNF and the antidepressant response is outlined. This review underlines the necessity to include information on lifestyle choices in testing and developing antidepressant treatments in the future.

Jauna dihidropiridīna atvasinājuma AV-6-93 efekts uz biomarķiera BDNF ekspresiju stresa modelī žurkām

Stresa izraisītie atmiņas bojājumi ir iemesls jaunu zāļu vielu meklējumiem, kas spētu aizkavēt smadzeņu bojājumus. Mūsu pētījuma mērķis bija noteikt jauna Latvijas Organiskās sintēzes institūtā sintezētās zāļu vielas AV-6-93, kas sastāv no dihidropiridīna un adamantāna atvasinājumiem, ietekmi uz neirotrofā faktora BDNF ekspresiju imobilizācijas stresētām žurkām hipokampā ar Western blota metodi. Petījuma rezultāti rāda, ka imobilizācijas stress ir palielinājis BDNF ekspresiju žurku hipokampā salīdzinājumā ar kontroles hipokampa žurkām, un AV-6-93 piemīt spēja normalizēt BDNF ekspresiju stresēto žurku hipokampā. Postmenopausal women may be more vulnerable to cognitive loss and Alzheimer's disease (AD) than premenopausal women because of their deficiency in estrogens, in addition to their usually older age. AbstractNeurotrophins promote the survival of specific types of neurons during development and ensure proper maintenance and function of mature responsive neurons. Significant effects of BDNF (Brain-Derived Neurotrophic Factor) on pain physiology have been reported but the contribution of this neurotrophin to the development of nociceptors has not been investigated. We present evidence that BDNF is required for the survival of a significant fraction of peptidergic and non-peptidergic nociceptors in dorsal root ganglia (DRG) postnatally.
