Genetic and biochemical characterization by random and site-directed mutagenesis, repressors involved in stress response phenolic acids in Bacillus s…

2009

The aim of this work was to characterize the genetic and biochemical mechanisms involved in the PASR, the phenolic acid stress response induced by phenolic acids Bacillus subtilis. By genetics and molecular biology strategies, like reverse genetic, PCR or transposon random mutagenesis, gene deletion, complementation of strains, and gene expression, the main components of the PASR were identified and quantified. Site-directed and random mutagenesis of respectively the promoter of padC encoding the phenolic acid decaroxylase, and PadR, the repressor of the padC expression, allowed us to identify the site of binding of PadR with this promoter, as well as the amino-acid residues involved in the…

Observing heme doming in myoglobin with femtosecond X-ray absorption spectroscopy.

2015

International audience; We report time-resolved X-ray absorption measurements after photolysis of carbonmonoxy myoglobin performed at the LCLS X-ray free electron laser with nearly 100 fs (FWHM) time resolution. Data at the Fe K-edge reveal that the photoinduced structural changes at the heme occur in two steps, with a faster (∼70 fs) relaxation preceding a slower (∼400 fs) one. We tentatively attribute the first relaxation to a structural rearrangement induced by photolysis involving essentially only the heme chromophore and the second relaxation to a residual Fe motion out of the heme plane that is coupled to the displacement of myoglobin F-helix

A study of the potential influence of frame coolant on HCLL-TBM nuclear response

2007

Abstract Within the European Fusion Technology Programme, the Helium-Cooled Lithium Lead (HCLL) breeding blanket concept is one of the two EU lines to be developed for a long term fusion reactor, in particular with the aim of manufacturing a test blanket module (TBM) to be implemented in ITER. The HCLL-TBM is foreseen to be located in an ITER equatorial port, being housed inside a steel-supporting frame, actively cooled by pressurized water. This supporting frame has been designed to house two different TBMs providing two cavities separated by a dividing plate 20 cm thick. As the nuclear response of HCLL-TBM could vary with the supporting frame configuration and composition, a parametric st…

IMAGINE: A Cold Neutron Imaging Station at the Laboratoire Léon Brillouin

2015

International audience; A second cold neutron imaging station has been open to users at the Laboratoire Léon Brillouin. The station is designed for high resolution neutron imaging and tomography. The typical field of view is 100x100 mm2 with a spatial resolution of 100 μm. Betterspatial resolutions (∼50 μm) can be achieved when reducing the field of view down to 30x30mm2. The L/D ratio can be varied from 200 to1000with pinhole sizes ranging from 20 to7 mm. Future upgrades will provide capabilities for energy resolved measurements using either a velocity selector or a double crystal monochromator. The possibility to perform polarized neutron experiments will also be provided next year.

Rare isotopic species of hydrogen sulfide: the rotational spectrum of H236S

2014

The rotational spectrum of the 36 S-bearing isotopologue of hydrogen sulfide (H2S) has been investigated for the first time in the 167 GHz−1.6 THz frequency range, thus providing an accurate and reliable set of spectroscopic parameters. The experimental investigation was backed up by state-of-the-art quantum-chemical calculations, which also allowed us to demonstrate the incorrectness of the previously reported spectroscopic constants. The present results are of suitable accuracy to attempt the astrophysical detection of the isotopic species under consideration. Finally, reliable predictions for the spectroscopic constants of other rare isotopologues of H2S, namely the mono- and bi-deuterat…

Body composition in children and adolescents residing in southern Europe: Prevalence of overweight and obesity according to different international r…

2019

The objective was to analyze body composition in children and adolescents of Southern Europe to identify prevalence of overweight and obesity. This investigation involved 512 girls and 488 boys between 7-to 19-years. Variables evaluated were Body Mass Index (BMI) and Fat Mass (FM; electrical bioimpedance). The references used to establish prevalence according to BMI were those of the World Health Organization (WHO) and the International Obesity Task Force (IOTF); in the case of FM, the Child Growth Foundation (CGF) reference was used. There were significant differences (p < 0.05) in the prevalence of overweight and obesity between the three classifications (32.3% according to IOTF, 37.3%…

Flux of protons released by wild type and ferritin over-expressor tobacco plants : effect of phosphorus and iron nutrition

2003

Abstract Tobacco ( Nicotiana tabacum ) plants over-expressing the iron storage protein ferritin, either in the cytoplasm or in the plastids, were grown under various P and Fe conditions. The crossed effects of both the genotypes and the environmental conditions on iron and chlorophyll concentrations in leaves, ferric reductase (EC 1.6.99.13) and plasmalemma H + -ATPase (EC 3.6.3.6) activities in roots, and fluxes of H + released by roots were determined. The increase in leaf Fe concentration observed in plants over-expressing ferritin was accompanied by an increase in root ferric reductase and H + -ATPase activities. Iron deficient conditions induced a decrease in Fe and chlorophyll concent…

Moderate-Vigorous Physical Activity across Body Mass Index in Females : Moderating Effect of Endocannabinoids and Temperament

2014

Altres ajuts: This manuscript was supported by grants from Instituto Salud Carlos III (FIS PI11/210 and CIBERobn). Sarah Sauchelli is recipient of a pre-doctoral Grant (2013-17) by IDIBELL. Jose C. Fernández-García is recipient of a 'Rio Hortega' contract from 'Instituto de Salud Carlos III', Madrid, Spain (CM12/00059). The funders had no role in the study design, data collection and analysis, decision to publish, or preparation of the manuscript. Endocannabinoids and temperament traits have been linked to both physical activity and body mass index (BMI) however no study has explored how these factors interact in females. The aims of this cross-sectional study were to 1) examine differences…

Defense Responses of Fusarium oxysporum to 2,4-Diacetylphloroglucinol, a Broad-Spectrum Antibiotic Produced by Pseudomonas fluorescens

2004

A collection of 76 plant-pathogenic and 41 saprophytic Fusarium oxysporum strains was screened for sensitivity to 2,4-diacetylphloroglucinol (2,4-DAPG), a broad-spectrum antibiotic produced by multiple strains of antagonistic Pseudomonas fluorescens. Approximately 17% of the F. oxysporum strains were relatively tolerant to high 2,4-DAPG concentrations. Tolerance to 2,4-DAPG did not correlate with the geographic origin of the strains, formae speciales, intergenic spacer (IGS) group, or fusaric acid production levels. Biochemical analysis showed that 18 of 20 tolerant F. oxysporum strains were capable of metabolizing 2,4-DAPG. For two tolerant strains, analysis by mass spectrometry indicated…
