Abstract Prostate cancer is the second leading cause of cancer deaths in men and there is an increasing interest in chemoprevention to fight it. The authors sought a scientific rationale for the traditional use of six Cameroonian medicinal plants for the treatment of prostate inflammation/tumour. Aim: To study a new series of [1,2,3]triazolo[1,5-α]pyridine derivatives as trypanocidal agents because current antichagasic pharmacologic therapy is only partially effective. Materials & methods: The effect of the series upon Trypanosoma cruzi epimastigotes and murine macrophages viability, cell cycle, cell death and on the metabolites of the sterol biosynthesis pathway was measured; also, docking in 14α-demethylase was analyzed. Computational identification of chemical compounds with potential anti-Chagas activity using a classification tree

Chagas disease is endemic to 21 Latin American countries and is a great public health problem in that region. Current chemotherapy remains unsatisfactory; consequently the need to search for new drugs persists. Here we present a new approach to identify novel compounds with potential anti-chagasic action. A large dataset of 584 compounds, obtained from the Drugs for Neglected Diseases initiative, was selected to develop the computational model. Dragon software was used to calculate the molecular descriptors and WEKA software to obtain the classification tree. Cytotoxic Spirostane-Type Saponins from the Roots of Chlorophytum borivilianum

Four new spirostane-type saponins named borivilianosides E-H (1-4) were isolated from an ethanol extract of the roots of Chlorophytum borivilianum together with two known steroid saponins (5 and 6). The structures of 1-4 were elucidated using mainly 2D NMR spectroscopic techniques and mass spectrometry. The cytotoxicity of borivilianosides F (2), G (3), and H (4) and three known compounds was evaluated using two human colon cancer cell lines (HT-29 and HCT 116).

Abstract Five sesterterpenes (1–5) including two new compounds (1 and 2), as well as a new (6) and a known macrolide (7) were isolated from the endophytic fungus Aplosporella javeedii. The structures of the new compounds were elucidated by analysis of their 1D and 2D NMR and HRMS data as well as by comparison with the literature. Compound 4 and its acetyl derivatives 4a, 4b, 4c which were prepared by acetylation of 4 exhibited moderate cytotoxicity against the mouse lymphoma cell line L5178Y with IC50 values ranging from 6.2 to 12.8 μM, respectively. Racemic and chiral nonracemic alpha-substituted and alpha-unsubstituted beta-fluoroalkyl beta-amino acid derivatives 6 and 9 have been synthesized in two steps starting from fluorinated imidoyl chlorides 1 and ester enolates. This approach is based on the chemical reduction of previously obtained gamma-fluorinated beta-enamino esters 4 by using ZnI(2)/NaBH(4) in a nonchelated aprotic medium (dry CH(2)Cl(2)) as the reducing agent. A metal-chelated six-membered model has been suggested to explain the stereochemical outcome of the reduction reaction. Optically detected magnetic resonance of triplet states populated by photoexcitation in water-soluble chlorophyll proteins (WSCPs) from Lepidium virginicum has been performed using both absorption and fluorescence detection. Well resolved triplet-singlet (T-S) spectra have been obtained and interpreted in terms of electronic interactions among the four chlorophylls (Chls), forming two dimers in the WSCP tetramer. Localization of the triplet state on a single Chl leads to a redistribution of the oscillator strength in the remaining three Chls of the complex. Abstract Vibrio cholerae cytolysin (VCC) forms oligomeric pores in lipid bilayers containing cholesterol. Membrane permeabilization is inefficient if the sterol is embedded within bilayers prepared from phosphatidylcholine only but is greatly enhanced if the target membrane also contains ceramide. Although the enhancement of VCC action is stereospecific with respect to cholesterol, we show here that no such specificity applies to the two stereocenters in ceramide; all four stereoisomers of ceramide enhanced VCC activity in cholesterol-containing bilayers. Abstract Four hitherto unreported cycloartane-type triterpenes (1-4) with C-28/C-30 oxidized methyl groups together with two known compounds (5-6) have been obtained from the leaves and twigs of Caloncoba dusenii Gilg. The compounds were isolated through extraction and silica gel column chromatography. Their structures and relative stereochemistry were determined by extensive use of spectroscopic techniques including IR, 1D and 2D-NMR as well as mass spectrometry. This is the first report of C-28/C-30 oxidized positions in cycloartanes from nature.
