Efficient Production of Isoflavonoids by Astragalus membranaceus Hairy Root Cultures and Evaluation of Antioxidant Activities of Extracts

In this study, Astragalus membranaceus hairy root cultures (AMHRCs) were established as an attractive alternative source for the efficient production of isoflavonoids (IF). A. membranaceus hairy root line II was screened as the most efficient line and was confirmed by PCR amplification of rolB, rolC and aux1 genes. Culture parameters of AMHRCs were systematically optimized, and five main IF constituents were quali-quantitatively determined by LC-MS/MS. Under optimal conditions, the total IF accumulation of 34 day old AMHRCs was 234.77 μg/g dry weight (DW). A Multiplicity result for a class of strongly indefinite asymptotically linear second order systems

We prove a multiplicity result for a class of strongly indefinite nonlinear second order asymptotically linear systems with Dirichlet boundary conditions. The key idea for the proof is to bring together the classical shooting method and the Maslov index of the linear Hamiltonian systems associated to the asymptotic limits of the given nonlinearity.

The identity type weak factorisation system

We show that the classifying category C(T) of a dependent type theory T with axioms for identity types admits a non-trivial weak factorisation system. We provide an explicit characterisation of the elements of both the left class and the right class of the weak factorisation system. This characterisation is applied to relate identity types and the homotopy theory of groupoids.

ABSTRACT Between 22 and 45% of HIV-positive subjects are likely to report symptoms of depression. Considering this background, a potential pharmacokinetic interaction between the nonnucleoside reverse transcriptase inhibitor efavirenz (EFV) and two antidepressants, sertraline (SRT) and nortriptyline (NT), was studied. Rats were administered EFV alone or together with the antidepressants, and changes in the plasma levels and pharmacokinetic parameters of EFV were analyzed. Analysis of Chlorpyrifos in Water, Fruit Juice, and Honeybee Extract by Chemiluminescent Elisa

Abstract The suitability of competitive enzyme-linked immunosorbent assays (ELISAs) with chemiluminescent detection-based immobilized antigen (indirect assay) for rapid and accurate determination of chlorpyrifos in various food matrices was tested. The limit of detection (LOD) values were 1–1.75 ng mL−1, the standard curve midpoint (IC50) was 3.5 ng mL−1, and the assay duration was 1.5 h. Assay application to the analysis of honeybee extract resulted in chlorpyrifos recoveries varying between 62 and 83% in 5–15 ng mL−1 herbicide concentration range.

Effect of Sublethal Concentrations of Pesticides on the Feeding Behavior of Daphnia magna

Daphnia magna was exposed to sublethal levels (1/4, 1/2, and 2/3 LC50 and LC50) of endosulfan and diazinon to determine the effect of these pesticides on filtration and ingestion rates. The experiments were performed with the unicellular algae Nannochloris oculata in a density of 5 x 10(5) cells/ml. Prior to these experiments, the acute toxicity of both pesticides was evaluated to calculate the LC50s. The 24-hr LC50 values were 0.62 mg/liter and 0.9 microgram/liter for endosulfan and diazinon, respectively. Coscinolactams A and B: new nitrogen-containing sesterterpenoids from the marine sponge Coscinoderma mathewsi exerting anti-inflammatory properties

Abstract Investigation of the marine sponge Coscinoderma mathewsi led to the isolation of two novel nitrogen-containing cheilanthane sesterterpenoids, coscinolactams A and B, together with known suvanine. The structures were elucidated by extensive spectroscopic measurements including NOE experiments to deduce the stereochemistry. The natural compounds, as well as a semisynthetic derivative, showed moderate anti-inflammatory activity measured as their capability to inhibit PGE2 and NO production. The suvanine aldehyde derivative 4 inhibited inducible nitric oxide protein expression with an IC50 value of 7.3 μM.

We have previously investigated the effects of a series of dimethoxy- and trimethoxychalcone derivatives, with various patterns of fluorination, on nitric oxide production in LPS-stimulated murine RAW 264.7. The present study was designed to determine if 3,4,5-trimethoxy-4'-fluorochalcone (CH 17) could modulate the production of NO and/or prostaglandins in vivo. On the mouse macrophage cell line RAW 264.7 CH 17 inhibited dose-dependently NO production, with an IC(50) value in the nanomolar range, and reduced PGE(2) levels by a 58% at 10 microM. Effect of the lipid regulator Gemfibrozil in the Cladocera Daphnia magna at different temperatures.

ABSTRACTIn the present study, an ecotoxicological approach to the evaluation of Gemfibrozil (GEM) as an emerging organic pollutant was done. In order to assess its toxicity, tests were conducted using the cladocera Daphnia magna. Experiments were carried out at 22°C and 28°C. EC50, feeding behavior, and chronic toxicity tests (21 days) were evaluated in D. magna exposed to GEM as well as cholesterol levels at 21-day chronic exposure. D. magna GEM EC50 values (24 h) in our experimental conditions were 148.75 and 116.24 mg L−1 at 22°C and 28°C, respectively.
