Mitophagy in myeloid cells : role in infection with gram-negative bacteria

Sepsis and related organ dysfunctions remain a leading cause of mortality in intensive care units. Increasing evidences have shed light on an unexpected link between mitochondria and immune cell functions. Alterations in mitochondrial functions have been reported in peripheral blood cells in sepsis. We hypothesize here that mitophagy might impact on phagocyte functions in the context of bacterial infection. Mitophagy is a mitochondria-dedicated autophagy that governs the elimination of dysfunctional mitochondria. Chronic myelomonocytic leukemia (CMML) is a hematologic stem cell disease whose characteristics correspond to myelodysplastic/myeloproliferative syndroms (MDS/MPS). Hematopoietic conditional deletion of Tif1γ in mice leads to the development of a MDS/MPS, mimiking human CMML, when age is comprised between 6 to 10 months, defining Tif1γ as a tumour suppressor gene. Among its multiple roles in the body, it is a component of cell membranes that is crucial for the maintenance of their structure and fluidity and is thus implicated in the modulation of many signalling pathways. Functional characterisation of the first HSP110 inhibitors.

Heat shock proteins are molecular chaperones highly expressed in haematological malignancies. My laboratory has shown that the heat shock protein HSP110 is a new and important therapeutic target In colorectal cancer and in non-Hodgkin's lymphoma. As there were no existing inhibitors of HSP110, a screening strategy of a chemical library was carried out and allowed the identification of two molecules capable of specifically inhibiting the chaperone activity of HSP110. Study of the dynamics of biomolecules by high speed atomic force microscopy and surface enhanced Raman spectroscopy

This thesis focuses on the coupling of High–Speed Atomic Force Microscopy (HS-AFM) and Surface Enhanced Raman Spectroscopy (SERS) for biomolecule analysis. We have designed a fabrication protocol to manufacture “SERS-active” substrates. The efficacy of gold, silver and gold-silver bimetallic crystalline nanoparticle substrates were evaluated. Bleomycin induced pulmonary toxicity and its subpleural onset

Idiopathic Pulmonary Fibrosis (PF) is a rare and devastating disease without efficient treatment at this time. Idiopathic FP is characterized by accumulation of myofibroblasts and has a typical sub-pleural onset suggesting a role of the pleura in the disease. Transforming Growth Factor (TGF)-ß1 induces transformation of pleural mesothelial cells (PMC) into active cells exhibiting myofibroblast phenotype. Selective reflection spectroscopy of alkali vapors confined in nanocells and emerging sensing applications

This thesis is aimed at studying the resonant interaction of a laser radiation with an atomic alkali vapor layer of wavelength-scale thickness confined in an optical nanocell; and emerging sensing applications.We focus our attention on the selective reflection arising at the interface between a dielectric window and a resonant alkali vapor, and show that the derivative of selective reflection spectra exhibit narrow resonances whose maxima are located exactly at atomic resonance frequencies.
